SYNOPSIS DNA synthesis of intracellular Trypanosoma cruzi amastigotes, following the infection of bovine embryo skeletal muscle (BESM) cells, was studied by autoradiography. After penetration, there was a prereplicative lag period (∼12 h) followed by a synchronous round of DNA synthesis which was found to be independent of parasite number/BESM cell and the host cell DNA synthesis cycle. Parasite reproduction occurred, for the first time, at ∼ 21 h postinfection. It was concluded that T. cruzi trypomastigotes are in the G1 /G, phase of their cell division cycle and that after penetration parasite reproduction occurs independent of events controlling host cell DNA synthesis and growth. The early synchronous growth of intracellular amastigotes should facilitate further studies on the biochemical events controlling trypomastigote-to-amastigote transformation and amastigote reproduction. A further application is envisaged for studies on the mode of action of drugs with trypanocidal activity. 相似文献

The reproduction of Toxoplasma gondii RH-strain in vertebrate cells was studied in a controlled-environment culture system. The lag period before reproduction and the doubling time of individual parasites were determined using a least-squares linear regression method of analysis which does not artificially constrain the data. In the majority of cases, the time intercept of the linear regression line was either zero, implying the lack of a lag phase before reproduction, or negative, implying the parasite had completed part of its reproductive cycle before entering the host cell. The mean doubling time of T. gondii is 10.9 h in bovine embryo skeletal muscle cells and 8.3 h in HeLa cells. This difference is not significant at the 5% level. Following stratification seeds of Corylus avellana exhibitedtheir characteristic ability to germinate at 20 °C undermoist conditions. Stratification of the intact fruit also stimulatedelongation of the cotyledonary petiole when isolated cotyledonswere transferred to moist conditions at 20 °C. GA3 inducedboth of these effects in non-stratified material. ABA substantiallydecreased seed germination and the response of cotyledonarypetioles to stratification and GA3. CCC2 applied to stratifiedor GA3-pretreated cotyledons did not depress the final percentageof growing petioles. Cotyledons can clearly regulate the development of their petiolesin the absence of the embryonic axis. Secondary tumours were formed on the cotyledonary leaf petiole,the hypocotyl, and first true leaf of castor bean seedlingsafter inoculating the blades of the cotyledonary leaves withAgrobacterium tumefaciens. Depending on the strain of bacteriaemployed, 0 to 80 per cent of the plants developed secondarytumours. The ability of different strains to initiate secondarytumours was not obviously correlated with their relative effectivenessin initiating primary tumours. Though all produced primary tumours,five out of ten auxotrophic strains failed to yield secondarytumours, whereas only one out of 14 prototrophic strains failedto do so. Both the number of plants developing secondary tumoursand the frequency with which these tumours occurred on differentparts of the plant were positively correlated with the concentrationof the primary inoculum. Tumours also developed on the cotyledonaryleaf petiole and on the hypocotyl after vacuum infiltrationof A. tumefaciens into the blade of cotyledonary leaves. Inmost instances (9 out of 11 plants) no tumours were formed onthe blade of the infiltrated leaf. Thus, tumour formation equivalentto secondary tumours can occur in the absence of a primary tumouror an overt primary wound. Excision of inoculated leaves showedthat the stimulus for secondary tumour formation moves fromthe blade to the hypocotyl within 24 h. Attempts to demonstratethe presence of a sub-cellular tumour-initiating agent in homogenatesof inoculated leaves were unsuccessful. A. tumefaciens, however,was found in the petiole of the cotyledonary leaf and in thehypocotyl within 24 h of inoculation. Fluorescence and thermal imaging were used to examine the dynamics of stomatal patches for a single surface of Xanthium strumarium L. leaves following a decrease in ambient humidity. Patches were not observed in all experiments, and in many experiments the patches were short-lived. In some experiments, however, patches persisted for many hours and showed complex temporal and spatial patterns. Rapidly sampled fluorescence images showed that the measurable variations of these patches were sufficiently slow to be captured by fluorescence images taken at 3-min intervals using a saturating flash of light. Stomatal patchiness with saturating flashes of light was not demonstrably different from that without saturating flashes of light, suggesting that the regular flashes of light did not directly cause the phenomenon. Comparison of simultaneous fluorescence and thermal images showed that the fluorescence patterns were largely the result of stomatal conductance patterns, and both thermal and fluorescence images showed patches of stomatal conductance that propagated coherently across the leaf surface. These nondispersing patches often crossed a given region of the leaf repeatedly at regular intervals, resulting in oscillations in stomatal conductance for that region. The existence of these coherently propagating structures has implications for the mechanisms that cause patchy stomatal behaviour as well as for the physiological ramifications of this phenomenon. 相似文献

Idiomorphs mat1-1 and mat1-2 from Mycosphaerella fijiensis , the causal agent of black leaf streak disease of banana, were isolated. Degenerate oligos were used to amplify the HMG box of the mat1-2 idiomorph from M. fijiensis , showing homology with the HMG box of Mycosphaerella graminicola. Using a DNA walking strategy, anchored on the DNA lyase gene towards the HMG box, a 9-kb-long region of mat1-2 was obtained. A 5-kb fragment from the mat1-1 region was obtained by long-range PCR using primers on the flanking regions, which have close to 100% identity between both idiomorphs. High-identity (77–89%), inverted regions within both idiomorphs were found, which suggest unique inversion events, which have not been found before, and that could have been significant in the evolution of this species. The predicted genes showed the conserved introns in both idiomorphs as well as an additional intron within the alpha box. The implications for the evolution of species in the Mycosphaerella complex on banana are discussed. 相似文献

In this article, I explore the lessons that the anthropological debates of the 1980s about writing culture might have for contemporary childhood research within anthropology and the social sciences more generally. I argue that the current rhetoric about "giving voice to children," commonplace both inside and outside the academy, poses a threat to the future of childhood research because it masks a number of important conceptual and epistemological problems. In particular, these relate to questions of representation, issues of authenticity, the diversity of children's experiences, and children's participation in research, all of which need to be addressed by anthropologists in their own research practices with children. Unless anthropologists do so, childhood research risks becoming marginalized once more and will fail to provide an arena within which children are seen as social actors who can provide a unique perspective on the social world about matters that concern them as children. 相似文献

Responses of Respiration to Increases in Carbon Dioxide Concentration and Temperature in Three Soybean Cultivars 总被引:3,自引:1,他引:2
The purpose of this experiment was to determine how respirationof soybeans may respond to potential increases in atmosphericcarbon dioxide concentration and growth temperature. Three cultivarsof soybeans (Glycine max L. Merr.), from maturity groups 00,IV, and VIII, were grown at 370, 555 and 740cm3m-3carbon dioxideconcentrations at 20/15, 25/20, and 31/26°C day/night temperatures.Rates of carbon dioxide efflux in the dark were measured forwhole plants several times during exponential growth. Thesemeasurements were made at the night temperature and the carbondioxide concentration at which the plants were grown. For thelowest and highest temperature treatments, the short term responseof respiration rate to measurement at the three growth carbondioxide concentrations was also determined. Elemental analysisof the tissue was used to estimate the growth conversion efficiency.This was combined with the observed relative growth rates toestimate growth respiration. Maintenance respiration was estimatedas the difference between growth respiration and total respiration.Respiration rates were generally sensitive to short term changesin the measurement carbon dioxide concentration for plants grownat the lowest, but not the highest carbon dioxide concentration.At all temperatures, growth at elevated carbon dioxide concentrationsdecreased total respiration measured at the growth concentration,with no significant differences among cultivars. Total respirationincreased very little with increasing growth temperature, despitean increase in relative growth rate. Growth respiration wasnot affected by carbon dioxide treatment at any temperature,but increased with temperature because of the increase in relativegrowth rate. Values calculated for maintenance respiration decreasedwith increasing carbon dioxide concentration and also decreasedwith increasing temperature. Calculated values of maintenancerespiration were sometimes zero or negative at the warmer temperatures.This suggests that respiration rates measured in the dark maynot have reflected average 24-h rates of energy use. The resultsindicate that increasing atmospheric carbon dioxide concentrationmay reduce respiration in soybeans, and respiration may be insensitiveto climate warming. Glycine max L.
