The coliform bacterial population in the Grand Forks, N.Dak. sewage system was examined for multiple-antibiotic-resistant organisms over a 1-year period. Multiple-antibiotic-resistant coliforms were found to be common in the sewage, and their numbers remained fairly constant relative to the total coliform population throughout the year. Resistance to kanamycin, tetracycline, and ampicillin was found to be transferable at variable rates. Transfer rates were found to be temperature sensitive and were optimal at 35 degrees C. Although 75% of the multiple-antibiotic-resistant coliforms were capable of transferring resistance at some level, only 25% were capable of transferring resistance at rates greater than 10(-3) transconjugants per initial donor.  相似文献

We show here that progesterone added to the medium of proliferating T47Dco human breast cancer cells is metabolized with a half life of 2-4h. The final metabolic product, 5 alpha-pregnan-3 beta,6 alpha-diol-20-one, (P-metabolite) is released into the medium. This structure suggested that the intracellular metabolism of progesterone involves the enzymes 5 alpha-reductase, 3 beta-hydroxysteroid dehydrogenase, and 6 alpha-hydroxylase. To investigate this pathway, the cells were incubated with a variety of potential substrates. In addition to progesterone, only precursors with the 5 alpha-configuration served as substrates for the enzymes leading to P-metabolite formation. Some precursors with a 5 beta-configuration were also metabolized by T47Dco cells. This metabolism reflected activity by either 3 beta-hydroxysteroid dehydrogenase and/or 6 alpha-hydroxylase but, in contrast to progesterone metabolism, the rates were different and the products were often mixtures. In T47Dco and MCF-7 human breast tumor cells, the reduction at C-3 followed by 6 alpha-hydroxylation, appear to be the major, and possibly only, route of progesterone metabolism. In contrast, preliminary data suggest that in normal human breast epithelial cells, this is not an exclusive route. Androgens are partially subject to the same metabolic enzymes, but synthetic progestins are not metabolized by T47Dco during an 18 h incubation.  相似文献

Insulin release from statically incubated HIT-T15 cells was maximally stimulated by glucose, L-arginine and L-leucine. L-arginine stimulated insulin release in the absence of glucose. Glucose induced insulin release was potentiated by the addition of L-leucine, L-arginine and the two in combination. Both glibenclamide and chlorpropamide stimulated insulin release from HIT-T15 cells. Glibenclamide was the more potent and equivalent in insulinotrophic action to 7.5 mmol/l glucose. Only chlorpropamide significantly potentiated glucose induced insulin release. Perifused HIT-T15 cells produced a reproducible biphasic insulin response to glucose challenge which was characterised by a pronounced and sustained first phase and a reduced second phase. The stimulation of phase I by glibenclamide alone and the inhibition of phase II of glucose induced insulin release by colchicine suggested the presence of a readily available pool of insulin granules which was not rapidly restored by insulin biosynthesis and granule margination.  相似文献

Kinetic parameters of 3-(3, 4-dichlorophenyl)-1, 1-dimethyl urea (DCMU)-induced inhibition of electron transport in chloroplast thylakoids isolated from Phaseolus vulgaris L. cv. Oregon 1604 were determined from analysis of a convergent, parallel electrical circuit. Through this analogue, the apparent affinity of the purported binding site for DCMU (K1) and the relative amount of DCMU-insensitive electron transport (vmax1/vo) were obtained using a reiterative non-linear least squares curve-fitting procedure. Exposure of thylakoids to heat caused a gradual increase in K1 (or decrease in the affinity of the thylakoid for DCMU) with an apparent activation energy of 134 kJ mol−1. Tryptic susceptibility of a protein region regulating K1 also decreased gradually with exposure to 45°C, suggesting that the heat-induced increase in K1 might be due to a protein conformational change. On the other hand, thylakoid exposure to 45°C resulted in a rapid (<5 min) irreversible increase in vmaxI/vo, which was also the apparent result of a conformational change in a region of the protein which regulates this function. These results are suggestive of the existence of differential thermal sensitivities of proteins within the thylakoids and, perhaps, of different regions within a single membrane protein.  相似文献

From the time of its introduction, in 1973, through November 1983, fine needle aspiration biopsy ( FNAB ) of the lung was used in the diagnostic evaluation of 1,015 patients at the Duke University Medical Center and Durham Veterans Administration Medical Center. A diagnosis of a primary lung cancer was established in 49.4% of the cases and malignant neoplasm metastatic to the lung in 14.1%; a benign diagnosis was made in 31.1% and an inconclusive one in 5.4%. In 123 patients in whom lung tissue was also available, cancer was correctly diagnosed by FNAB in 83.8%, not diagnosed by FNAB in 14.6% and incorrectly diagnosed in 1.6%. In 37 patients without cancer, FNAB detected an infectious organism or a specific morphologic type of inflammation (abscess and granuloma).  相似文献

The ability of an invasive plant to occupy new areas is often attributed to both morphological and physiological plasticities that allow them to remain viable over a wide range of environmental conditions. Studies addressing the ecological requirements of Microstegium vimineum often consider soil moisture or soil moisture along with other factors as important explanatory components for the establishment and persistence of this invasive monocot. However, controlled studies specifically targeting water relations in M. vimineum are needed. Therefore, the purpose of this study was to determine how different water availabilities influence the growth and physiological performance of M. vimineum. This study utilized experimental microcosms to achieve different water availabilities including low soil moisture (<15% water), moderate soil moisture (ca. 20–30%), and flooded conditions. While both flooded and low soil moisture resulted in diminished growth, M. vimineum still survived under these conditions. Physiological processes including C4 metabolism, minimum stress under low water conditions, and the ability to increase tissue rigidity may confer some advantages to M. vimineum during periods of limiting water conditions. Similarly, the proportionally low root biomass, shallow root structure, and its ability to maintain stable water relations during flooding and/or soil waterlogging may facilitate M. vimineum’s ability to invade mesic habitats. It is likely, therefore, that the capacity to tolerate both low soil moistures and flooded conditions has enhanced the ability of M. vimineum to populate disturbed systems in central North Carolina.  相似文献
