Xylan is the major hemicellulosic constituent in dicot secondary cell walls. Cell wall composition of cotton fiber changes dynamically throughout development. Not only the amounts but also the molecular sizes of the hemicellulosic polysaccharides show substantial changes during cotton fiber development. However, none of the genes encoding glycosyltransferases (GTs) responsible for synthesizing xylan have been isolated and characterized in cotton fiber. In this study, we applied a bioinformatics approach and identified two putative GTs from cotton, designated GhGT43A1 and GhGT43C1, which belong to the CAZy GT43 family and are closely related to Arabidopsis IRX9 and IRX14, respectively. We show that GhGT43A1 is highly and preferentially expressed in 15 and 20 days post‐anthesis (dpa) cotton fiber, whereas GhGT43C1 is ubiquitously expressed in most organs, with especially high expression in 15 dpa fiber and hypocotyl. Complementation analysis demonstrates that GhG43A1 and GhGT43C1 are orthologs of Arabidopsis IRX9 and IRX14, respectively. Furthermore, we show that overexpression of GhGT43A1 or GhGT43C1 in Arabidopsis results in increased xylan content. We also show that overexpression of GhGT43A1 or GhGT43C1 leads to more cellulose deposition. These findings suggest that GhGT43A1 and GhGT43C1 likely participate in xylan synthesis during fiber development.  相似文献

This review aims to describe changes in breastfeeding and summarise the breastfeeding rates, duration and reasons of discontinuing 'any breastfeeding' or 'exclusive breastfeeding' in P.R. China. Breastfeeding rates in China fell during the 1970s when the use of breast milk substitutes became widespread, and reached the lowest point in the 1980s. As a result many efforts were introduced to promote breastfeeding. The breastfeeding rate in China started to increase in the 1990s, and since the mid-1990s 'any breastfeeding' rates in the majority of cities and provinces, including minority areas, have been above 80% at four months. But most cities and provinces did not reach the national target of 'exclusive breastfeeding' of 80%. The 'exclusive breastfeeding' rates in minority areas were relatively lower than comparable inland provinces. The mean duration of 'any breastfeeding' in the majority of cities or provinces was between seven and nine months. The common reasons for ceasing breastfeeding, or introducing water or other infant food before four months, were perceived breast milk insufficiency, mother going to work, maternal and child illness and breast problems. Incorrect traditional perceptions have a strong adverse influence on 'exclusive breastfeeding' in less developed areas or rural areas. China is a huge country, geographically and in population size, and there is considerable ethnic diversity. Therefore breastfeeding rates in different parts of China can vary considerably.  相似文献

A simple, sensitive and rapid flow injection analysis (FIA) method with resonance light scattering (RLS) was described for the determination of propafenone (PPF). The method was based on the ion‐association reaction of 12‐tungstophosphoric acid (TP) with propafenone. In pH 1.0 acidic medium, TP reacted with PPF to form an ion‐associate complex, which resulted in a significant enhancement of RLS intensity. The maximum scattering peak was located at 340 nm, the RLS intensity was proportional to the concentration of PPF in the range 0.003–9.0 µg/mL, and the detection limit (3σ) of 1.0 ng/mL was obtained at a sampling rate of 60 samples/h. The feasible reaction conditions and FIA parameters for the system were optimized. The method proposed in this paper shows satisfactory reproducibility with a relative standard deviation (RSD) of 2.1% for 10 successive determinations of 2.0 µg/mL PPF. The present method had been successfully applied to the determination of PPF in serum samples and pharmaceutical samples. ZFP245 is a cold- and drought-responsive gene that encodes a zinc finger protein in rice. The ZFP245 protein localizes in the nucleus and exhibits trans-activation activity. Transgenic rice plants overexpressing ZFP245 were generated and found to display high tolerance to cold and drought stresses. The transgenic plants did not exhibit growth retardation, but showed growth sensitivity against exogenous abscisic acid, increased free proline levels and elevated expression of rice pyrroline-5-carboxylatesynthetase and proline transporter genes under stress conditions. Overproduction of ZFP245 enhanced the activities of reactive oxygen species-scavenging enzymes under stress conditions and increased the tolerance of rice seedlings to oxidative stress. Our data suggest that ZFP245 may contribute to the tolerance of rice plants to cold and drought stresses by regulating proline levels and reactive oxygen species-scavenging activities, and therefore may be useful for developing transgenic crops with enhanced tolerance to abiotic stress.  相似文献

The mechanism of blue light-induced retinal ganglion cell (RGC) injury is poorly understood. In this study, we established a patented light-emitting diode-based system to study the effects of long-term blue light exposure under culture conditions on RGC-5 cells. Long-term blue light exposure significantly reduced cell viability in a time-dependent manner and induced apoptosis and necrosis in RGC-5 cells. Long-term blue light exposure marked an increase in the expression of Bax and active Caspase-3 (p17), which was accompanied by Bcl-2 down-regulation, and displayed features of the mitochondria-dependent apoptosis pathway. Blue light exposure also increased the generation of reactive oxygen species (ROS), and was a strong inducer of ROS-sensitive protein nuclear factor erythroid 2-related factor 2 (Nrf2) and heme oxygenase-1 (HO-1) expression. Moreover, blue light exposure constitutively activated p38 mitogen-activated protein kinases and c-Jun NH2-terminal kinase (JNK), as well as induced the phosphorylation of extracellular signal-regulated kinase in the early phase, in blue light-exposed RGC-5 cells. The protein expression of c-jun and c-fos was further enhanced after RGC-5 cells were exposed to blue light. Taken together, these findings indicated that blue light induced RGC-5 cell line death in dependence upon exposure duration. The potential mechanisms for this phenomenon might be via activated mitochondria-dependent apoptosis, increased ROS production and protein expressions of Nrf2 and HO-1, and activated JNK/p38 MAPK signaling pathways.  相似文献

dead end (dnd) was identified in zebrafish as a gene encoding an RNA-binding protein essential for primordial germ cell (PGC) development and gametogenesis in vertebrates. The adult dnd RNA expression has been restricted to the ovary in Xenopus or to the testis in mouse. Its protein product is nuclear in chicken germ cells but both cytosolic and nuclear in mouse cell cultures. Here we report the cloning and expression pattern of Odnd, the medakafish (Oryzias latipes) dnd gene. Sequence comparison, gene structure, linkage analysis and expression demonstrate that Odnd encodes the medaka Dnd orthologue. A systematic comparison of Dnd proteins from five fishes and tetrapod representatives led to the identification of five previously unidentified conserved regions besides the RNA recognition motif. The Odnd RNA is maternally supplied and preferentially segregated with PGCs. Its adult expression occurs in both sexes and is restricted to germ cells. In the testis, Odnd is abundant in spermatogonia and meiotic cells but absent in sperm. In the ovary, Odnd RNA persists throughout oogenesis. Furthermore, we developed a dual color fluorescent in situ hybridization procedure allowing for precise comparisons of expression and distribution patterns between two genes in medaka embryos and adult tissues. Importantly, this procedure co-localized Odnd and Ovasa in testicular germ cells and PGCs. Surprisingly, by cell transfection and embryo RNA injection we show that ODnd is cytoplasmic in cell cultures, cleavage embryos and PGCs. Therefore, medaka dnd encodes a cytoplasmic protein and identifies embryonic and adult germ cells of both sexes.  相似文献

Aims:  To determine Legionella pollution prevalence, describe the amount of Legionellae with respect to temperature in Shanghai cooling tower water (CTWs) in various types of public sites.
Methods and Results:  Six urban districts were selected as the study fields, adopting multiple-phase sampling methods. Routine culture was used to identify Legionellae. Of the samples, 58·9% (189/321) were observed to be positive, 19·9% were isolated over 100 CFU ml−1. Legionella pneumophila serogroup 1 was the most frequently isolated species (155/189, 82·0%), followed by Leg. micdadei that was at the second place (44/189, 23·3%). The mean CFU ml−1 of Legionellae in CTWs reached its peak from July to September. Over all 15·4% of the samples exceeding 100 CFU ml−1 were observed in a hospital setting.
Conclusions:  The prevalence of Legionella pollution in CTWs, especially in CTWs of subway stations and hospitals, is worrying, and the positive rate and CFU ml−1 of Legionellae in CTWs have a close relationship with air temperature.
Significance and Impact of the Study:  The study demonstrates pollution prevalence rates in different types of sites and various seasons, and provides a proportion of different serogroups of Legionellae . It illuminates an urgent need for dealing with the potential risk of legionellosis in Shanghai, through improved control and prevention strategies.  相似文献

以焦旱1号总DNA为材料,首先对影响RAPD-PCR反应的模板DNA、Mg~(2+)、dNTP、引物和Taq DNA聚合酶浓度等因素进行了初步优化,分析了各因素对RAPD-PCR扩增结果的影响.在此基础上对影响RAPD-PCR反应的Mg~(2+)、dNTP、引物和Taq DNA聚合酶浓度等4个主要因素进行正交优化,研究结果表明:在25 μl RAPD-PCR反应体系中,模板DNA 20 ng;Mg~(2+)浓度1.5mmol/L;dNTP的浓度0.2mmol/L;引物量15 pmol;Taq DNA聚合酶1.0 U.在此最佳条件下,利用引物B8对18个北方粳稻品种进行了成功的扩增.  相似文献

甘珀酸干预对大鼠脑缺血再灌注损伤的影响   总被引:1,自引:0,他引:1  
目的观察缝隙连接阻断剂甘珀酸对局灶性脑缺血/再灌注损伤的影响。方法采用大鼠大脑中动脉阻塞再灌流模型(MCAO),将动物随机分为脑缺血60min再灌注(MCAO)组,脑缺血再灌注加甘珀酸干预(MCAO+CBX)组和假手术组(sham)。采用尼氏染色显示脑梗死灶并计算梗死灶体积;应用免疫荧光与TUNEL染色法分别观察脑缺血后3d与7d不同时间点缺血边缘区胶质纤维酸性蛋白(GFAP)的表达和细胞凋亡情况。结果(1)缺血后3d、7d MCAO+CBX组大鼠梗死体积小于MCAO组,3d、7d MCAO+CBX组大鼠梗死体积较MCAO组分别缩小5%和4.6%;(2)缺血后3d、7d于缺血边缘区可见大量TUNEL阳性染色细胞,且MCAO组大鼠缺血边缘区细胞凋亡数目明显多于MCAO+CBX大鼠(P〈0.001);(3)缺血后3d和7d组缺血边缘区GFAP表达明显增强,3d的MCAO组与MCAO+CBX组大鼠缺血边缘区GFAP的表达均较假手术组强(P〈0.05),7d的MCAO+CBX组大鼠缺血边缘区GFAP的表达较假手术组强(P〈0.001),但明显弱于MCAO组大鼠(P〈0.01);结论缝隙连接阻断剂甘珀酸可减少大鼠大脑中动脉阻塞后脑梗死体积,其机制可能与阻断缝隙连接后缺血边缘区神经元凋亡降低有关,星型胶质细胞的反应性变化参与了该过程。  相似文献
