In the present study we provide the first evidence supporting the fact that the Kp43 NK-associated cell-surface dimer may be involved in regulating MHC-unrestricted cytotoxicity. Our results indicated that incubation of IL-2-activated NK cells in a 51Cr-release assay with either the Kp43-specific mAb or its F(ab')2 fragments induced a significant cytolytic activity directed against normal autologous and allogeneic T cell blasts, which are relatively resistant to NK cell-mediated lysis. The cytotoxic effect was not observed in fresh CD3- CD16+ CD56+ Kp43+ lymphocytes and was only substantiated in IL-2-preactivated NK cells. Although stimulation with the Kp43-specific mAb did not significantly change the intracellular Ca2+ concentration, both Ca2+ and Mg2+ were required for the induction of cytotoxicity. The anti-Kp43-mediated activation of cytolysis was inhibited by anti-CD18 and CD11a mAb, whereas it was not significantly altered by either CD11b, CD11c, CD2, or LFA-3-specific mAb, rendering unlikely the participation of the latter. In contrast to these results the Kp43-specific mAb did not enhance the high levels of spontaneous cytotoxicity mediated by IL-2-activated NK cells against a panel of different tumor cell lines. An inhibitory effect mediated by anti-Kp43 mAb on the IL-2-dependent proliferation of NK cells was previously reported and appears, at least partially, secondary to the induction of an autolytic mechanism that is synergistically enhanced by anti-CD16 mAb. Altogether our results point out that interaction of the Kp43 dimer with its specific mAb is capable of inducing cytolytic activity and suggest that the molecule may play an important functional role in lymphokine-activated NK cells.  相似文献

The functional feeding groups and diversity of macroinvertebrate communities associated with duckweed mats in the New Years River (two sites) and Bloukrans River (two sites), Eastern Cape province, South Africa, were assessed. Duckweed (Lemnaceae) is a ubiquitous family of floating macrophytes. A total of 41 macroinvertebrate families were collected monthly over a six-month period from February to July 2014. Duckweed biomass in both rivers was highly variable both temporally and spatially. The majority of identified macroinvertebrate taxa were predators and detritivores, with a small percentage of herbivores. An average of approximately 26% of the macroinvertebrate taxa found were from families that include species from more than one functional feeding group. Although overall measures of diversity and ecosystem health (Fisher’s α and Simpson’s index) remained constant over time in the New Years River, significant differences in macroinvertebrate community structure were seen between sites and months on both rivers, with dissimilarity being driven by a larger number of species in the New Years River. This high variability within macroinvertebrate assemblages probably reflects a combination of heterogeneous duckweed distribution, variation in physico-chemistry, opportunistic behaviours of macroinvertebrate predators and/or successional colonisation of duckweed mats.  相似文献

This study compares a non-linear (neural network) and a linear (linear regression) power mapping using a set of features of the surface electromyogram recorded from the vastus medialis and lateralis muscles. Fifteen healthy participants performed 5 sets of 10 repetitions leg press using the individual maximum load corresponding what they could perform 10 times (10RM) with 120 s of rest between them. The following sEMG variables were computed from each extension contraction and used as inputs to both approaches: mean average value (MAV), median frequency (Fmed), the spectral parameter proposed by Dimitrov (FInsm5), average (over the observation interval) of the instantaneous mean frequency obtained from a Choi–Williams distribution (MFM), and wavelet indices ratio between moments at different scales (WIRM1551, WIRM1M51, WIRM1522, WIRE51, and WIRW51). The non-linear mapping (neural network) provided higher correlation coefficients and signal-to-noise ratios values (although not significantly different) between the actual and the estimated changes of power compared to linear mapping (linear regression) using the sEMG variables alone and a combination of WIRW51 and MFM (obtained by a stepwise multiple linear regression). In conclusion, non-linear mapping of force loss during dynamic knee extension exercise showed higher signal-to-noise ratio and correlation coefficients between the actual and estimated power output compared to linear mapping. However, since no significant differences were observed between linear and non-linear approaches, both were equally valid to estimate changes in peak power during fatiguing repetitive leg extension exercise.  相似文献

Motile organisms actively detect environmental signals and migrate to a preferable environment. Especially, small animals convert subtle spatial difference in sensory input into orientation behavioral output for directly steering toward a destination, but the neural mechanisms underlying steering behavior remain elusive. Here, we analyze a C. elegans thermotactic behavior in which a small number of neurons are shown to mediate steering toward a destination temperature. We construct a neuroanatomical model and use an evolutionary algorithm to find configurations of the model that reproduce empirical thermotactic behavior. We find that, in all the evolved models, steering curvature are modulated by temporally persistent thermal signals sensed beyond the time scale of sinusoidal locomotion of C. elegans. Persistent rise in temperature decreases steering curvature resulting in straight movement of model worms, whereas fall in temperature increases curvature resulting in crooked movement. This relation between temperature change and steering curvature reproduces the empirical thermotactic migration up thermal gradients and steering bias toward higher temperature. Further, spectrum decomposition of neural activities in model worms show that thermal signals are transmitted from a sensory neuron to motor neurons on the longer time scale than sinusoidal locomotion of C. elegans. Our results suggest that employments of temporally persistent sensory signals enable small animals to steer toward a destination in natural environment with variable, noisy, and subtle cues.  相似文献

Aim We test whether species of western Mediterranean aquatic Coleoptera of the ‘Haenydra’ lineage (Hydraenidae, Hydraena) originated through: (1) successive periods of dispersal and speciation, (2) range fragmentation by random vicariance, or (3) range fragmentation by geographic isolation owing to a general reduction of population density. Location Europe. Methods To discriminate between scenarios we use contrasting predictions of the relationship between phylogenetic and geographic distance. The phylogeny was based on 3 kb of four mitochondrial and two nuclear gene fragments of about half of the known species of ‘Haenydra’, including most western Mediterranean taxa. Divergences were estimated using a molecular clock. The relationship between phylogenetic and geographic distance was tested using bivariate plots, Mantel tests and comparison of the observed phylogeny with the one minimizing geographic distances between species, as measured using Euclidean minimum spanning trees (EMSTs). Results The monophyly of ‘Haenydra’ was strongly supported, although its phylogenetic placement was not resolved. ‘Haenydra’ was estimated to be of late Miocene age, with most species originating during the Pleistocene. In two clades (Hydraena tatii and Hydraena emarginata clades) there was a significant association between geographic and phylogenetic distance, and the reconstructed phylogeny was identical to that obtained through the EMST, demonstrating a strong non‐randomness of the geographic distribution of the species. In two other clades (Hydraena iberica and Hydraena bitruncata clades) there was no association between geographic and phylogenetic distance, and the observed phylogeny was not the one minimizing geographic distances. In one of the clades this seems to be due to a secondary, recent range expansion of one species (H. iberica), which erased the geographic signal of their distributions. Main conclusions We show that it is possible to obtain strong evidence of stasis of the geographic ranges of narrow‐range endemic species through the study of their phylogenetic relationships and current distributions. In at least two of the studied clades, current species seem to have originated through the fragmentation of a more widely distributed species, without further range movements. A process of range expansion and fragmentation may have occurred repeatedly within the ‘Haenydra’ lineage, contributing to the accumulation of narrow‐range endemics in Mediterranean Pleistocene refugia.  相似文献

The purpose of the present study was to evaluate the possible effect of melanin-concentrating hormone (MCH) on learning and memory by using the one-trial step-down inhibitory avoidance test in rats. The peptide was infused into hippocampus, amygdala, and entorhinal cortex. MCH caused retrograde facilitation when given at 0 or 4 h post-training into hippocampus, but only at 0 h into amygdala. From these results, it seems that MCH modulates memory early after training by acting on both the amygdala and hippocampus and, 4 h after training, on the hippocampus.  相似文献

6-Methyl-3'-bromoflavone inhibited [(3)H]flunitrazepam binding to the benzodiazepine binding site of the GABA(A) receptor (BDZ-bs) with Ki values between 10 and 50 nM in different brain regions.The GABA ratio of 1.03 for [(3)H]flunitrazepam binding to cerebral cortex, 0.76 for cerebellum, 0.7 for hippocampus, 0.7 for striatum, and 0.8 for spinal cord indicated an antagonistic or weak inverse agonistic profile of 6-methyl-3'-bromoflavone on BDZ-bs. Unlike classical benzodiazepines, it had no anticonvulsant, anxiolytic, myorelaxant, sedative, amnestic or motor incoordination effects. However, it antagonized the muscle relaxant, the sedative effect, and the changes in locomotor activity induced by diazepam. Taken together, these findings suggest that 6-methyl-3'-bromoflavone has an antagonistic profile on the BDZ-bs.  相似文献

We have recently reported that activation of protein kinase C (PKC) plays a negative role in CD95-mediated apoptosis in human T cell lines. Here we present data indicating that although the PKC-induced mitogen-activated protein kinase pathway could be partially implicated in the abrogation of CD95-mediated apoptosis by phorbol esters in Jurkat T cells, the major inhibitory effect is exerted through a PKC-dependent, mitogen-activated protein kinase-independent signaling pathway. Furthermore, we demonstrate that activation of PKC diminishes CD95 receptor aggregation elicited by agonistic CD95 Abs. On the other hand, it has been reported that UV radiation-induced apoptosis is mediated at least in part by the induction of CD95 oligomerization at the cell surface. Here we show that activation of PKC also inhibits UVB light-induced CD95 aggregation and apoptosis in Jurkat T cells. These results reveal a novel mechanism by which T cells may restrain their sensitivity to CD95-induced cell death through PKC-mediated regulation of CD95 receptor oligomerization at the cell membrane.  相似文献
