The aim of this study was to examine the effect of serotonin and epinephrine on ion transport of isolated frog skin. The addition of serotonin after incubation in Ringer solution (RH), bumetanide (BUME), and after initial incubation in amiloride and subsequently in RH, reduced hyperpolarization and did not effect the mechanosensitivity of frog skin. Following incubation of the frog skin with amiloride (AMI), serotonin did not affect the value of hyperpolarization and increased mechanosensitivity. The addition of epinephrine (EPI) on frog skin incubated in RH and AMI did not affect hyperpolarization, but repeated application of this compound after serotonin increased hyperpolarization. After incubation with bumetanide, addition of EPI before and after application of serotonin did not affect the value of the examined parameters of the frog skin. Initial incubation with AMI and later in RH caused a drop in reaction to EPI and no effect on mechanosensitivity. Repeated addition of epinephrine in this group did not affect the reaction value, while it decreased the reaction value during mechanical stimulation. The experimental data presented in this study indicate that serotonin inhibits the sodium ion current. Epinephrine inhibits the chloride ion current, however, after the application of serotonin, EPI stimulates sodium ion transport. 相似文献

Peripherally located parts of spider mechanosensory neurons are modulated by several neurotransmitters released from apposed
efferent fibers. Activities of acetylcholine (ACh) synthesizing enzyme choline acetyltransferase (ChAT) and ACh degrading
enzyme acetylcholine esterase (AChE) were previously found in some efferent fibers. ChAT activity was also present in all
the mechanosensory neurons, while AChE activity was only found in some. We show that spider mechanosensory neurons and probably
some efferent neurons are immunoreactive to a monoclonal antibody against muscarinic ACh receptors (mAChRs). However, application
of muscarinic agonists did not change the physiological responses or membrane potentials of neurons in the lyriform organ
VS-3. Similarly, the sensitivities of the neurons of trichobothria (filiform hairs) remained unchanged after application of
these agonists. Therefore, activation of mAChRs may only modulate the function of spider mechanosensory neurons indirectly,
for example, by affecting the release of other transmitter(s). However, a subgroup of VS-3 neurons was inhibited by ACh, which
also depolarized the membrane similar to these neurons’ responses to GABA, suggesting that ACh activates anion channels in
these neurons. Interestingly, all of the neurons responding to ACh were the rapidly adapting Type A neurons that were previously

High segregational stability of the streptococcal plasmid pSM19035 is achieved by the concerted action of systems involved in plasmid copy number control, multimer resolution, and postsegregational killing. In this study, we demonstrate the role of two genes, delta and omega, in plasmid stabilization by a partition mechanism. We show that these two genes can stabilize the native pSM19035 replicon as well as other theta- and sigma-type plasmids in Bacillus subtilis. In contrast to other known partition systems, in this case the two genes are transcribed separately; however, they are coregulated by the product of the parB-like gene omega. Analysis of mutants of the parA-like gene delta showed that the Walker A ATPase motif is necessary for plasmid stabilization. The ParB-like product of the omega gene binds to three regions containing repeated WATCACW heptamers, localized in the copS (regulation of plasmid copy number), delta, and omega promoter regions. We demonstrate that all three of these regions can cause partition-mediated incompatibility. Moreover, our data suggest that each of these could play the role of a centromere-like sequence. We conclude that delta and omega constitute a novel type of plasmid stabilization system. 相似文献

Progesterone in saliva was monitored using a new method called magnetic particle-based immuno supported liquid membrane assay (m-ISLMA) in a sequential injection (SI) setup, allowing automatic sample cleanup, analyte enrichment, and detection in a single analysis unit. Progesterone (Ag) diffuses from a continuous flowing sample - the donor - into a supported organic liquid membrane (SLM), based on analyte partitioning (solubility) between the aqueous donor and the organic phase. The Ag is re-extracted from the SLM into a second stagnant aqueous acceptor, containing antibodies (Ab) immobilized on magnetic beads, held at the bottom of the acceptor by a magnet. Due to the formation of strong Ag-Ab-bead complexes and a large excess of Ab-beads, the Ag is accumulated and selectively enriched in the acceptor. The extracted progesterone was quantified by injecting into the acceptor a horseradish peroxidase (HRP) labeled analyte tracer, the substrate (luminol, H(2)O(2), and p-iodophenol), and finally detection of the generated chemiluminescence by a photomultiplier tube. After optimization of experimental parameters (e.g., sample flow rate, extraction time, type of organic solvent and antibody-bead concentration in the acceptor), a detection limit of 8.50+/-0.17 fgL(-1) and a dynamic range between 35 fgL(-1) and 10 pgL(-1) was reached. The progesterone level of saliva for three subjects (women in different period of ovarian cycle) was investigated, and the corresponding progesterone concentrations detected with m-ISLMA coincided well with the expected values. 相似文献

Eighty soil-borne Bacillus cereus group isolates were collected from two neighbouring geographical sites in Belgium. Their genetic relationships and population structure were assessed using Multilocus sequence typing analysis of five chromosomal genes, while the contribution of extrachromosomal elements to the population dynamics was gauged by the presence, diversity and transfer capacity of pXO1- and pXO2-like plasmids. Globally, the bacterial population displayed a broad diversity, including an important subpopulation of psychrotolerant isolates related to Bacillus weihenstephanensis . pXO1- and pXO2-like replicons were present in 12% and 21% of the isolates, but no Bacillus anthracis -related toxin genes were found. Furthermore, only one of the isolates containing a pXO2-related plasmid was shown to be able to mobilize small non-self-conjugative plasmids. Interestingly, several B. cereus sensu lato isolates displaying the same sequence type were observed to have different plasmid contents, suggesting the occurrence of horizontal gene exchange. Similarly, a number of pXO2-like replicons with identical sequences were found in distinct bacterial isolates, therefore strongly arguing for lateral transfers among sympatric bacteria. 相似文献

The poultry red mite, Dermanyssus gallinae, is currently a significant pest in the poultry industry in Europe. Biological control by the introduction of predatory mites is one of the various options for controlling poultry red mites. Here, we present the first results of an attempt to identify potential predators by surveying the mite fauna of European starling (Sturnus vulgaris) nests, by assessing their ability to feed on poultry red mites and by testing for their inability to extract blood from bird hosts, i.e., newly hatched, young starlings and chickens. Two genuine predators of poultry red mites are identified: Hypoaspis aculeifer and Androlaelaps casalis. A review of the literature shows that some authors suspected the latter species to parasitize on the blood of birds and mammals, but they did not provide experimental evidence for these feeding habits and/or overlooked published evidence showing the reverse. We advocate careful analysis of the trophic structure of arthropods inhabiting bird nests as a basis for identifying candidate predators for control of poultry red mites. 相似文献

Cul3 (Cullin3)-based E3 ubiquitin ligases recently emerged as critical regulators of mitosis. In this study, we identify two mammalian BTB (Bric-a-brac–Tramtrack–Broad complex)-Kelch proteins, KLHL21 and KLHL22, that interact with Cul3 and are required for efficient chromosome alignment. Interestingly, KLHL21 but not KLHL22 is necessary for cytokinesis and regulates translocation of the chromosomal passenger complex (CPC) from chromosomes to the spindle midzone in anaphase, similar to the previously described BTB-Kelch proteins KLHL9 and KLHL13. KLHL21 directly binds to Aurora B and mediates ubiquitination of Aurora B in vitro. In contrast to KLHL9 and KLHL13, KLHL21 localizes to midzone microtubules in anaphase and recruits Aurora B and Cul3 to this region. Together, our results suggest that different Cul3 adaptors nonredundantly regulate Aurora B during mitosis, possibly by ubiquitinating different pools of Aurora B at distinct subcellular localizations. 相似文献
