Patients with cirrhosis of the liver often have insulin resistance and elevated circulating growth hormone levels. This study was undertaken (a) to evaluate glucose intolerance, insulin resistance and abnormal growth hormone secretion and (b) to determine if GH suppression improves insulin resistance. Glucose tolerance tests (GTT), intravenous insulin tolerance tests (IVITT), arginine stimulation tests (AST) and glucose clamp studies before and during GH suppression with somatostatin were performed in a group of patients with alcohol-induced liver cirrhosis. During GTT cirrhotic subjects had a 2-hour plasma glucose of 200 +/- 9.8 ng/dl (N = 14) compared to 128 +/- 8.0 ng/dl in normal controls (N = 15), P less than 0.001. Basal GH was elevated in cirrhotic patients and in response to arginine stimulation reached a peak of 17.0 +/- 5.4 ng/ml (N = 7), compared to a peak of 11.3 +/- 1.8 ng/ml in 5 normal controls (P = NS). During IVITT patients with cirrhosis had a glucose nadir of 60.0 +/- 4.0 mg/dl (N = 9), compared to 29.0 +/- 7.0 mg/dl in controls (N = 5), P less than 0.001. Peak GH levels during IVITT were not significantly different in cirrhotics and controls. Glucose utilization rates in 4 patients with cirrhosis of the liver before somatostatin mediated GH suppression was 3.1 +/- 0.5 mg/kg/min and 6.5 +/- 1.5 mg/kg/min during somatostatin infusion, P less than 0.025. We conclude that patients with alcohol induced cirrhosis have sustained GH elevations resulting in insulin resistance which improves after GH suppression.  相似文献

Summary Rhizobium meliloti infective on Medicago, Melilotus and Trigonella plants has three copies of the nodulation regulatory gene nodD. Strains containing mutations in nodD1 exhibited a delayed and/or decreased nodulation on Melilotus albus (Ma), Medicago sativa (Ms), Medicago quasifalcata (Mqu) and Trigonella coerulea (Tc), while on Medicago truncatula (Mt) they nodulated similarly to the wild-type R. meliloti. Delayed nodulation was observed also when nodD2 mutants were inoculated onto Ms, Mt and Tc, but not on Ma and Mqu. A nodD3 mutant exhibited delayed nodulation on Ms and Ma. Using a nodC-lacZ fusion and cloned nodD genes on plasmids, high induction levels were detected in R. meliloti when nodD1 was present with seed exudates from Ms, Ma and Mqu, nodD2 with those from Ms and Mt, and nodD3 with those from Ms, Ma and Mqu. NOne of the nodD copies exhibited high levels of nodC-lacZ induction when present with seed exudate from Tc. Only nodD1 induced nodC-lacZ expression in conjunction with the flavone, luteolin. The plant hosts used in this study exude different flavonoids and correlation between nodulation and nodC-lacZ induction abilities of the host exudates was observed. We concluded that all the three nodD copies of R. meliloti have common nod-promoter activating but diverged flavonoid-recognizing abilities. Thus, the three nodD alleles contribute to the activation of nodulation genes in a host-dependent manner.  相似文献

The bacterial host ranges of the conjugatively self-transmissible IncN group plasmid pCU1 and its mobilizable miniderivative, pCU785, were examined. Species of the gram-negative purple bacteria were chosen for this study. Conjugative mobilization of pCU785 into a wide variety of bacteria was facilitated by the presence of oriT of the broad-host-range plasmid RK2 in pCU785. Although the host range of the IncN tra system of pCU1 is broad, the host range of its replicon is limited. However, the pCU1 replicon can be maintained in Agrobacterium, Bradyrhizobium, and Rhizobium species under conditions that select for plasmid maintenance. It is lost efficiently from these populations on release of selection.  相似文献

The hypothesis that the accumulation of prostaglandin (PG)E2 during reperfusion of severely ischemic tissue contributes to a breakdown in the blood-brain barrier (BBB) was expanded to include a parallel role for levuglandins(LGs), gamma-ketoaldehydes produced by rearrangement of PGH2. LGE2 was shown to be more potent than PGE2 in causing breakdown of the BBB when injected intrahemispherically. Brain tissue necrosis was clearly evident with total doses of levuglandin as low as 100 nmole.  相似文献

Investigation of the genetic system of six species ofPlantago has revealed striking differences in their breeding and meiotic systems.Plantago patagonica is an inbreeder on account of preanthesis cleistogamy, whereasP. lanceolata is an obligate outbreeder, as it is self-incompatible.Plantago drummondii, P. lagopus, P. ovata, andP. major show mixed mating but in varying proportions. In terms of their energy budgets, outbreeding species invest more in floral advertisement and male function, while inbreeders invest more in female function. The contribution of the meiotic system to genetic variability, as revealed through recombination index, is more important in the inbreeding species.  相似文献

The specificity between the sym-2 gene bred into certain cultivars of pea (Pisum sativum L.) and the nodX gene, present only rarely in isolates of Rhizobium leguminosarum, can be exploited to preempt competition or nodulation blocking by a Rhizobium strain indigenous to a soil environment. The principle is to isolate an R. leguminosarum strain prevalent in a locale, convert it into a strain that will nodulate a desirable pea cultivar carrying sym-2 by establishing nodX in it, and then use the resulting Rhizobium strain with the pea cultivar carrying sym-2. To accomplish this, we first constructed a transposon Tn5 derivative called Tn5nodX and an efficient delivery vehicle that is suicidal in R. leguminosarum. We tested the potential utility of the system in greenhouse experiments. The results are encouraging enough to warrant extensive experiments under field conditions.  相似文献

Summary A novel cyclic dipeptide —cyclo[(S)-His-(S)-NorArg] — has been prepared which catalyzes an enantioselective version of the Strecker amino acid synthesis. The catalyst, when present in 2 mol % quantity in methanol solution, catalyzes the addition of hydrogen cyanide toN-alkylimines to afford-amino nitriles in high yield and high enantiomeric excess. Furthermore, acid hydrolysis ofN-benzhydryl--amino nitriles afforded the corresponding-amino acids directly. This methodology affords a variety of arylglycines in exceptionally high enantiomeric excess, but aliphatic amino acids were obtained with low enantioselectivity. Current efforts are underway to expand the scope of this reaction, as well as to elucidate the mechanism of catalysis and the roles played by substrate and catalyst in determining the stereochemical outcome of the reaction.A preliminary communication on this work has recently appeared in: Iyer MS, Gigstad KM, Namdev ND, Lipton MA (1996) J Am Chem Soc 118: 4910–4911.  相似文献

A modified and rapid technique has been developed for activity staining of a fungal poly(3-hydroxybutyrate) depoly-merase by incorporating poly(3-hydroxybutyrate) (2.5% w/v) in the polyacrylamide gel under polymerisation and incubating in 100 mM Tris/HCl (pH 8.0) containing 1 mM CaCl at 30°C. This decreased the time required for detection to 4 h from the conventional 12–18 h using solid-plate method. The enzyme preparation failed to penetrate the gel on conventional electrophoresis but treatment with 0.5% (v/v) Triton X-100 facilitated enzyme migration into the gel without its activity being affected.  相似文献

Fifteen red pigmented trisomics were isolated in the F2 generation from the cross Corchorus olitorius L. x C. capsularis L. In the F3 generation a few green trisomics were obtained; more of these were isolated from the backcross generation. A detailed morphological and cytological analysis of the trisomic hybrid populations derived from the F3 and F4 generations is presented. The trisomics were scored for several morphologically differentiating characters and most of them were intermediate between the parental species, a few resembling the olitorius parent more. Cytological studies showed the formation of abnormal sporads in trisomics with different frequencies indicating a misdivision at meiosis. This imbalance at meiosis results in a higher percentage of pollen sterility in the trisomics as compared with the parents. Analysis of M1 of meiosis showed that there were: differences in the frequencies of the various chromosome configurations between the two categories of trisomics; significantly higher trivalent frequencies per PMC in the green trisomics; in contrast significantly hihger univalent frequencies per PMC in the red trisomics. No significant difference in chiasma formation was observed between red and green trisomics, nor between trisomics and their parental species. It appears that segmental homology in the parental chromosomes has probably resulted in varying degrees of preferential pairing in the trisomic hybrid.  相似文献
