We describe for the first time the sexual behavior and the courtship song of males of the African fly Zaprionus indianus (Gupta), a recent invader of South America. The male courtship song is formed by monocyclic pulses and the courtship behavior is simple when compared to that of species of Drosophila. Two interpulse interval (IPI) distributions were observed: pre-mounting and mounting. No significant difference was observed between the pre-mounting IPIs of males that descended from three geographical populations from South America. We also observed the songs produced by females and the homosexual behavior exhibited by males. A sequence of bursts is produced by females as a refusal signal against males, while males emit a characteristic song that identifies sex genus, which differs from the courtship song. The short courtship and mating latencies recorded reveal vigorous males and receptive females, respectively.  相似文献

A multiconformational study of substrates of cytochrome P450 3A4 has been carried out within the BiS/MC algorithm. The method allowed one to create a pseudoatomic model of the cytochrome and to find the substrate conformers responsible for the interaction with the cytochrome. It has been found that, in most cases, the geometry of the acting conformer is much different from the geometry of the global minimum conformer. It has been shown that the mirror conformational antipodes ("enantioconformers") are characterized as a rule by different Michaelis constants. A quantitative relationship between the Michaelis constants and the parameters of interactions in "model 3A4 isoform-substrate" complexes has been determined. The relationship describes the experimental value of Michaelis constant with the squared cross-validation correlation coefficient of 0.88.  相似文献

The structural and functional diversity of the main ecological trophic groups of soil microorganisms in meadow soils of the Central Urals anthropogenically contaminated with heavy metals was studied. The increase in the total numbers of these microorganisms in technozems, in comparison with those in agrozems, is due to the higher abundance of iron-reducing, denitrifying, nitrogen-fixing, and sulfate-reducing bacteria, an increase in cellulolytic activity, and the dependence of these characteristics on the toxic load of the soil. A reductive structure of the microbial community with the predominance of r-strategists, which reflects earlier stages of microbiocenoses succession under soil contamination, is formed under soil pollution with heavy metals.  相似文献

Biotreatment of petroleum-contaminated water in a fluidized-bed bioreactor (FBB) is a relatively new and promising technology, which efficiency is strongly affected by the biocatalyst used. Our laboratory experiments involved biotreatment of the water contaminated with a synthetic petroleum mixture consisting of aliphatic and polyaromatic hydrocarbons (PAHs) using a continuous column bioreactor with recycle. Different type biocatalysts were tested, including Rhodococcus bacteria immobilized in hydrophobized carriers such as sawdust, poly(vinyl alcohol) cryogel (cryoPVA) and poly(acrylamide) cryogel (cryoPAAG). Biocatalyst abilities to oxidize petroleum hydrocarbons were evaluated using the Columbus Micro-Oxymax® respirometer. The hydrophobized sawdust-supported biocatalyst demonstrated substantially higher metabolic activity than C12-cryoPAAG-based biocatalyst due to larger number of immobilized Rhodococcus cells and, therefore, had benefits for application in FBBs. The obtained results showed that designed FBB process is successful, providing 70–100% removal of n-alkanes (C10–C19) and 66–70% removal of 2–3-ring PAHs from contaminated water after 2–3 weeks.  相似文献

A simple biosurfactant-based hydrophobization procedure for poly(vinyl alcohol) (PVA) cryogels was developed allowing effective immobilization of hydrocarbon-oxidizing bacteria. The resulting partially hydrophobized PVA cryogel granules (granule volume 5 microl) contained sufficient number (6.5 x 10(3)) of viable bacterial cells per granule, possessed high mechanical strength and spontaneously located at the interface in water-hydrocarbon system. Such interfacial location of PVA granules allowed high contact of immobilized biocatalyst with hydrophobic substrate and water phase, thus providing bacterial cells with mineral and organic nutrients. As a result, n-hexadecane oxidation efficiency of 51% after 10-day incubation was achieved using immobilized biocatalyst. PVA cryogels with increased hydrophobicity can be used for immobilization of bacterial cultures performing oxidative transformations of water-immiscible organic compounds. Immobilization of in situ biosurfactant producing Rhodococcus bacteria into PVA cryogel is discussed. PVA cryogel granules with entrapped alkanotrophic rhodococcal cells were stable after 10-month storage at room temperature.  相似文献

Experiments on the biodegradation of unusable medicines containing a phenolic hydroxy group by actinobacteria of the genus Rhodococcus were performed. Six species and sixty-four strains were tested. It was found that rhodococci could degrade paracetamol, and some R. ruber strains showed high levels of its degradation. An efficient method for the identification and quantification of paracetamol and the products of its conversion (p-aminophenol, pyrocatechol, and hydroquinone) immediately in the culture liquid was developed. Conditions for the complete biodegradation of paracetamol dosage forms (pills) were optimized. The experimental results can be applied for the development of biotechnological methods for degrading medicines: faked, rejected, or those that are expired.  相似文献

In geographically distant populations of ladybird beetle Adalia bipunctata from Eurasia mitotypes and infection with symbiotic bacteria Spiroplasma and Rickettsia were determined. All populations examined demonstrated mtDNA polymorphism and striking differences in prevalence of bacteria (from about 50% of individuals infected with Spiroplasma in St.-Petersburg population and 50% of the Rickettsia prevalence in Kem' population to complete absence of bacteria in the population from Archangelsk). In the populations studied a total of 14 mitotypes were discovered, including two mitotypes that were remarkably different from the others in nucleotide composition. Mitotype 10, which was the most different from all the others, was found in all populations from Germany to Transbaikalia, excluding the population from Tashkent. Linkage disequilibrium between mitotype 10 and the Rickettsia infection was confirmed. Infection with the Spiroplasma bacteria was typical of the individuals with haplotype 1 and relative to it. The results obtained supported the conclusion on the association between infection with Spiroplasma and Rickettsia and certain mitotype of A. bipunctata, which was the consequence of either absence or rare horizontal transfer of symbionts and ancientness of the first contact between the bacteria and A. bipunctata ladybird beetles.  相似文献

Introduction

Osteopontin (OPN) has been implicated as a mediator of Th17 regulation via type I interferon (IFN) receptor signaling and in macrophage activity at sites of tissue repair. This study assessed whether increased circulating plasma OPN (cOPN) precedes development of organ damage in pediatric systemic lupus erythematosus (pSLE) and compared it to circulating plasma neutrophil gelatinase-associated lipocalin (cNGAL), a predictor of increased SLE disease activity.

Methods

cOPN and cNGAL were measured in prospectively followed pSLE (n = 42) and adult SLE (aSLE; n = 23) patients and age-matched controls. Time-adjusted cumulative disease activity and disease damage were respectively assessed using adjusted-mean SLE disease activity index (SLEDAI) (AMS) and SLICC/ACR damage index (SDI).

Results

Compared to controls, elevated cOPN and cNGAL were observed in pSLE and aSLE. cNGAL preceded worsening SLEDAI by 3-6 months (P = 0.04), but was not associated with increased 6-month AMS. High baseline cOPN, which was associated with high IFNalpha activity and expression of autoantibodies to nucleic acids, positively correlated with 6-month AMS (r = 0.51 and 0.52, P = 0.001 and 0.01 in pSLE and aSLE, respectively) and was associated with SDI increase at 12 months in pSLE (P = 0.001). Risk factors for change in SDI in pSLE were cOPN (OR 7.5, 95% CI [2.9-20], P = 0.03), but not cNGAL, cumulative prednisone, disease duration, immunosuppression use, gender or ancestry using univariate and multivariate logistic regression. The area under the curve (AUC) when generating the receiver-operating characteristic (ROC) of baseline cOPN sensitivity and specificity for the indication of SLE patients with an increase of SDI over a 12 month period is 0.543 (95% CI 0.347-0.738; positive predictive value 95% and negative predictive value 38%).

Conclusion

High circulating OPN levels preceded increased cumulative disease activity and organ damage in SLE patients, especially in pSLE, and its value as a predictor of poor outcome should be further validated in large longitudinal cohorts.  相似文献
