In 13 cystic fibrosis (CF) patients of 5 to 23 years of age with a known mutation spectrum of gene CFTR, sweat chloride values and nasal-potential differences (NPD) were measured and localization characteristics of the protein product of gene CFTR in the cells of nasal epithelium were studied. Sweat Chloride values were normal or boundary (24 to 62 mM/l) in six CF patients. In seven CF patients, these values were significantly above the estimates for the control group. On average, the NPD values were -44.7 +/- 2.2 mV (from -32.5 to -68.9 mV) and -17.2 +/- 1.8 mV (from -6.8 to -30.2 mV) in CF patients and the control group, respectively. Histochemical studies clearly revealed the localization of the CFTR protein on the apical membrane of the nasal epithelium. Depending on the type of mutation, the protein product of gene CFTR was either absent or regularly distributed in the cytoplasm in CF patients; it was not detected in the apical membrane. Thus, NPD measurements and the analysis of the localization of the protein product of gene CFTR in scrapes of nasal epithelium were shown to be additional, highly informative methods of CF diagnostics. 相似文献

The ratio between the normal (+) and null (0) alleles of the genes encoding glutatione S-transferases M1 (GSTM1) and T1 (GSTT1) were studied in normal individuals from northwestern Russia (control group) and in patients with bronchial asthma (BA). The frequency of the GSTM1 0/0 genotype in the population sample was statistically significantly lower (37.8%) than in the BA patients (82.1%; chi 2 = 16.8; P < 0.001; w chi 2 = 15.7; alpha = 0.01). For the GSTT1 gene, similar data were obtained. The frequency of the GSTT1 0/0 genotype in healthy donors was statistically significantly higher (16.3%) than in the BA patients (73.7%; chi 2 = 28.5; P < 0.001; w chi 2 = 23.22; alpha = 0.01). A significant preponderance of the compound homozygotes for the GSTM1 and GSTT1 null alleles among the BA patients was observed. The frequency of the GSTM1 0/0, GSTT1 0/0 individuals among the patients was 57.9%, while it was only 4.7% among the controls (chi 2 = 27.4; P < 0.001). 相似文献

A simple and effective method for typing of CAG repeats in the IT-15 gene has been suggested. This method was applied for examination of the CAG allele distribution in Huntington's disease (HD) patients in five different populations from the Commonwealth of Independent States. A total of 21 normal alleles with the sizes ranging from 9 to 32 triplet repeat units were revealed. Moreover, alleles with the size ranging from 16 to 20 repeats predominated constituting from 54.4 to 74.6% of all alleles in different populations. The number of repeats in one allele in HD patients exceeded 38 units (43 triplets on average). In two families an increase in the CAG repeat units number in the mutant allele upon its paternal transmission was recorded. 相似文献

Effect of NaF and AlCl3 the activity of the sarcoplasmic reticulum Ca-ATP-ase has been investigated. NaF (mM) completely inhibits the Ca-ATP-ase activity in presence of 0.02% tween-20. The inhibition is time- and NaF-concentration-dependent and increases as affected by AlCl3 (microM). The potentiated action of AlCl3 depends on the NaF concentration. AlCl3 without NaF does not change the Ca-ATP-ase activity. NaF inhibits the Ca-ATP-ase competitively with respect to ATP, but NaF plus AlCl3 make the inhibition combined. The affinity of the Ca-ATP-ase to the NaF + AlCl3 complex, but not to NaF decreases by 5 mM with an increase of the Pi concentration. NaF probably interacts with the ATP-binding site and the NaF + AlCl3 complex interacts with the phosphate-binding site of the ATP-ase. 相似文献

Recognition of short time intervals (10, 60, and 180 ms) between visual stimuli presented to the left or right hemisphere was studied in adult healthy people. The interval of 180 ms is recognized better than that of 10 or 60 ms. Learning with repeated tests with 180 ms intervals proceeds better than that with short intervals. The predominance of the left hemisphere has been revealed only for perception of 10 ms interval. The other time intervals asymmetry is not observed. It is suggested that the left hemisphere is predominant in estimation of short (less than 60 ms) time intervals. In formation of time nervous model a significant role is played by local activation of the cortical zone where the standard stimulus is addressed. 相似文献

Using the non-ionic detergent Triton X-100 and gel-chromatography, an anion-sensitive ATPase was isolated from rat and rabbit erythrocyte membranes. The ATPase preparations possess no Na, K- or Mg, Ca-ATPase activities. The properties of anion-sensitive ATPase of rat heart mitochondria were studied. Na2CO3, NaHCO3 and Na2SO3 stimualted ATPase activity by 69, 41 and 110%, respectively. Azide, tiocinate and perchlorate inhibited bicarbonate-stimulated ATPase. Bivalent cations increased ATPase activity in such a sequence: Zn2+ greater than or equal to Cd2+ greater than or equal to Co2+ greater than or equal to Mg2+ greater than or equal to Mn2+ greater than Ni2+. In the presence of bicarbonate and sulfite. ATPase activity was maximally stimulated with magnesium. Ni2+ and Ca2+-ions inhibited Mg2+-dependent activity of bicarbonate-stimulated ATPase. AMP uninhibited ATPase activity. The 4 mM concentration of ADP inhibited activity of HCO-3-ATPase. Activity of ATPases decreased at lower temperatures. The properties of anion-sensitive ATPase of rat heart mitochondria and that of HCO-2-ATPase of other cells are discussed. 相似文献
