Hypoxia induces the expression of the pro-apoptotic gene BNIP3

It has been shown that oxygen deprivation results in apoptotic cell death, and that hypoxia inducible factor 1 (HIF1) and the tumor suppressor p53 play key roles in this process. However, the molecular mechanism through which hypoxia and HIF1 induce apoptosis is not clear. Here we show that the expression of pro-apoptotic gene BNIP3 is dramatically induced by hypoxia in various cell types, including primary rat neonatal cardiomyocytes. Overexpression of HIF1alpha, but not p53, induces the expression of BNIP3. Overexpression of BNIP3 leads to a rather unusual type of apoptosis, as no cytochrome c leakage from mitochondria was detected and inhibitors of caspases were unable to prevent cell death. Taken together, these data suggest that HIF1-dependent induction of BNIP3 may play a significant role during hypoxia-induced cell death.     

Suppression of Nonsense Mutations in the Dystrophin Gene by a Suppressor tRNA Gene

Nonsense mutations in the dystrophin gene are the cause of Duchenne muscular dystrophy (DMD) in 10–15% of patients. In such an event, one approach to gene therapy for DMD is the use of suppressor tRNAs to overcome the premature termination of translation of the mutant mRNA. We have carried out cotransfection of the HeLa cell culture with constructs containing a suptRNA gene (pcDNA3suptRNA) and a marker LacZ gene (pNTLacZhis) using their polymer VSST-525 complexes. It was found that the number of cells producing -galactosidase depends inversely on the dose of the suptRNA gene. A single in vivo injection of the construct providing for expression of the suptRNAochre gene into mdx mouse muscle resulted in the production of dystrophin in 2.5% of fibers. This suggests that suppressor tRNAs are applicable in gene therapy for hereditary diseases caused by nonsense mutations.     

Regulation of adenosine triphosphatase activity in mitochondria, chloroplasts and erythrocytes by anions, adenosine diphosphate and alcohols

The effects of sulfite, bicarbonate, thiocyanate, methanol, ethanol, glycerol, dimethy sulfoxide and ADP on the ATPase activity of the coupling factor from liver mitochondria (F1) and pea chloroplasts (CF1) and of the anion-sensitive ATPase from rat erythrocytes were investigated. Under steady-state conditions of ATP hydrolysis catalyzed by F1, CF1, and erythrocyte ATPase, three Km values for each of the enzymes, three activation constants for sulfite and three inhibition constants for thiocyanate were determined. The efficiency and direction of the effects of anions, alcohols and ADP strongly depend on temperature and substrate (Mg-ATP) concentration. The mechanisms of modification by anions and alcohols of the ATPase activities are discussed.     

Comamonas testosteroni Strain TI as a Potential Base for a Microbial Sensor Detecting Surfactants

Strain Comamonas testosteroni TI, capable of degrading the nonionic surfactant (NIS) nonylphenolethoxylate (OP-10), was used for constructing a pilot cellular biosensor. The lower NIS detection limit for the biosensor was 0.25 mg/l. We studied the substrate specificity of the biosensor with respect to a wide range of organic compounds: surfactants, polyaromatic compounds (PAC), carbohydrates, alcohols, etc. It was shown that the biosensor based on Comamonas testosteroni TI did not respond to glucose, which was an advantage over the formerly described biosensor based on Pseudomonas rathonis T. The amplitude of the sensor response remained stable for 10 days.     

Analysis of the Polymorphic Markers within the CFTR Gene in Cystic Fibrosis Patients and Healthy Donors from Bashkortostan

The differences in the polymorphic allele frequency distribution patterns of the biallelic (M470V and TUB20) and microsatellite (IVS6aGATT, IVS8CA, and IVS17CA) markers within the CFTR gene between normal and delF508 chromosomes have been established. For most of the marker loci similar distribution of the allele frequencies on normal and mutant chromosomes without delF508 was demonstrated. Certain polymorphic alleles displayed substantial linkage disequilibrium with the delF508 mutation. Analysis of the IVS6aGATT-IVS8CA-M470V-IVS17CA-TUB20 haplotypes association on normal and mutant chromosomes provided identification of the delF508 ancestral haplotype. It was suggested that delF508 mutant chromosomes were introduced into the modern Bashkir gene pool as a result of Slavic migrations from the Eastern Europe. The IVS6aGATT-IVS8CA-M470V-IVS17CA-TUB20major haplotype (77272) revealed was statistically significantly most frequently found on the mutant chromosomes without the delF508 mutation. This finding suggests that the Bashkir cystic fibrosis patients, mostly belonging to the Turkic-speaking families, possessed specific CFTR gene defect associated with the given haplotype.     

Hemispheric lateralization of visuo-spatial function in man

Two parts of a geometrical figure are consecutively presented to healthy adult subjects in the left and right visual fields; the subjects have to compare them mentally and to decide whether these parts form a standard figure or not. Correctness of the reaction is controlled by a computer which lights up on the screen the words "good" or "error". The number of correct decisions of this visual-spatial task does not depend on the hemisphere to which information is addressed. The reaction time is substantially shorter if the information comes "directly" to the right hemisphere. Due to better training in the left hemisphere interhemispheric difference in reaction time gradually disappears in repeated tests. Training to mental "constructing" takes place only in the tests following positive feedback stimulus. Analysis of amplitude-temporal parameters of P300 wave shows that at correct decision of the visual-spatial task the level of activation in the right hemisphere is higher than in the left one.     

Change in the number of epidermal growth factor receptors of hepatocytes during liver regeneration in rats

The number of epidermal growth factor (EGF) receptors was determined for rat's hepatocytes under condition of their regeneratory proliferation, following a partial hepatectomy. In the normal hepatocytes two classes of EGF-receptors are distinguished, with high and low affinity, respectively. As early as 2 hours after the partial hepatectomy the total number of EGF-receptors sharply decreases, those with high affinity being not determined at all. The patterns of autophosphorylation of EGF-receptors with low affinity, examined in regenerating liver hepatocytes, remained the same as in the intact liver: their kinase activity was EGF-dependent, tyrosine residues of receptor molecules, with molecular mass equal to 150-170 kDa, underwent phosphorylation.     

Learning to differentiate microintervals of time using verbal feedback

Reaction time (RT) and the number of correct estimations of time microintervals (10 and 180 ms) between two visual stimuli were recorded in healthy subjects. It has been shown that 10 ms interval is better estimated when the stimuli are presented in the right visual field, i.e. when they are addressed directly to the left hemisphere. At the same time the number of correct estimations of 180 ms interval is greater and their RT is less when the stimuli are addressed directly to the right hemisphere. This points to different hemispheric mechanisms of time microintervals estimation. Study of the influence of different forms of verbal reinforcement on this learning has shown that after positive reinforcement (the word "good") the number of correct estimations is on average by 10% greater than after negative reinforcement (the word "error"). This may be connected with such processes as isolation and identification of erroneous reaction.
