Effect of antitubulin colcemide on polarization of mouse fibroblast-like cells on anisotropic substrate was studied. Such a substrate was obtained by scratching narrow strips in lipid films, adsorbed on the glass. The control cells were seen spread only along the strips, and in 4-6 hours they approached the length of120-150 mcm. In colcemide-containing media, the cells remain in an unspread state for a long time; they extrude their outgrowths both along the strip and perpendicularly to it. Due to frequent refractions of outgrowths, two thirds of colcemide-treated cells were detached from the substrate. Possible mechanisms of these effects of antitubulins are discussed. 相似文献

Effect of colcemide upon the spreading of mouse embryo fibroblast-like cells on the substrate was studied with the aid of time-lapse microcinematography and scanning electron microscopy. On the glass, colcemide did not prevent the transition of cells into a well-attached state, however, the time needed for this transition was seen considerably increased as compared with the control cultures. Intermediate stages of spreading on flat glass had the following abnormal features in colcemide-containing medium: a) shapes of cytoplasmic outgrowths formed by the cell were altered and their distribution along the cell border appeared less regular; b) partial detachments of the attached parts of cells occurred very frequently; c) the spreading of various parts of the cells was not correlated. Possible mechanisms of colcemide action on the cell spreading are discussed, and it is suggested that intracellular structures sensitive to colcemide are essential for coordination of reactions that occur in various parts of the cell in the course of spreading. 相似文献

Big gerbils, inhabitants of desert, were found to have indices of an increased functional activity of HHNS: low content of neurosecretory material all over the system, relatively large volumes of neurosecretory cell nuclei and nucleoli, hyperemia. Under the condition of a seven-day dehydration, the neurosecretory material reduction in all links of HHNS, enlargement of neurosecretory cell nuclei and nucleoli, intensification of hyperemia were found. With the increase in dehydration time to 23 days a progressive reduction of neurosecretory material in cell processes and hypophysis are seen. However, the decrease in nuclear size takes place under the condition of continuing increase in nucleolar size both in supraoptic and paraventricular nuclei. Extremely intensive hyperemia of all regions of HHNS is seen. The activation of HHNS under the water-salt regime change, seen in big gerbils, is not as pronounced as in albino rats. Parafollicular cells (PC) of the sheep thyroid gland are neural crest derivatives that synthesize and release the biogenic amine serotonin (5-HT) as well as the hormone calcitonin. The thyroid also contains a highly specific serotonin-binding protein (SBP). Separation of dissociated thyroid cells was done to study the cellular localization of SBP and to develop a means of isolating PC for study. Various methods were used to obtain an enriched and purified population of PC. Minced thyroid glands were enzymatically dissociated and the cells were layered on a Ficoll linear density gradient. Fractions obtained from the gradient were examined for cell number, viability, 5-HT concentration, SBP activity, and morphology by electron microscopy. One of the fractions was found to be enriched in PC. High levels of 5-HT and SBP were also found in this fraction, whereas these levels were low where the majority of cells were found. This PC-rich fraction, however, contained numerous follicular cells (FC); therefore, additional approaches to cell separation were used. FC can be stimulated in vitro with thyroid stimulating hormone (TSH) to become intensely phagocytic. When stimulated cells were incubated in the presence of silica microspheres, the FC engulfed the microspheres, which were toxic to them. PC did not become phagocytic and were unharmed by the microspheres. Suspended cells, after incubation with microspheres, were centrifuged on a discontinuous gradient, and a PC-rich fraction was obtained. Silica, however, interfered with analysis of SBP. Another method to take advantage of the phagocytic potential of FC was therefore used. TSH-stimulated cell suspensions were passed through a column of sepharose to which thyroglobulin had been coupled. Stimulated FC apparently adhered to the beads and were retained by the columns. Fractions eluting from the columns were greatly enriched with PC. These fractions contained high levels of 5-HT and SBP, and considerably reduced FC contamination was found by quantitative electron microscopy. It is concluded that SBP is localized to PC in the sheep thyroid. The idea that these cells resemble serotonergic neurons in their mechanisms of 5-HT storage is supported. 相似文献

Summary VIP-like immunoreactive material is present in the female reproductive tract, with a distinct pattern of distribution. The highest concentrations of extractable material and immunoreactive nerve fibres were found in the cervix and vagina. In the cervix these fibres were seen below the surface epithelium and around cervical glands as well as in association with blood vessels and smooth muscle bundles. In the vagina the nerve fibres were most abundant in the superficial regions of the lamina propria. Scattered fibres were also present in the rest of the uterus and in the fallopian tubes. Chromatographic evidence indicates that this VIP-like material is of a similar molecular size to that extracted from other organs. Possible roles for VIP in the regulation of myometrial activity and of cervical and vaginal dilation and secretion are proposed. 相似文献

Bean plants grown in a controlled temperature glasshouse athigh temperatures (33/28, 30/25, or 27/22 °C) during theperiod of seed development and maturation matured early andproduced small seeds. The seeds were of lower vigour than thosegrown at 21/16 or 18/13 °C. The detrimental effect of highmaturation temperatures was observed even on plants bearingwell-developed seeds (yellow, fleshy-pod stage). Seeds maturedat high temperatures were also more susceptible to deteriorationwith delay in harvest, and to mechanical damage. The level of catecholamines was measured in various brain parts in rats of August and Wistar lines, characterized by different stability of cardio-vascular functions in emotional stress. In norm, the August line has a higher content of dopamine (DA), in the mid-brain in particular; the Wistar line has a higher content of noradrenaline (NA) in the hypothalamus. In susceptible animals, which showed different cardio-vascular disorders during 30 hours of immobilization (48% of the Wistar line and 90% of the August), and in animals resistent against emotional stress, the changes in the initial level of catecholamines were observed, both common and specific for two lines. An assumption is made that the stability of cardio-vascular functions in the emotional stress is provided for by an intensive metabolism of the NA-synthesizing neurones in the brain isthmus, and by a moderate activity of the DA-synthesizing neurones of the mid-brain; the susceptibility to cardio-vascular disorders in the emotional stress is due to increase in the activity of the DA-synthesizing mid-brain neurones, the decrease in the activity of the DA-hydroxilase and the exhaustion of NA in the hypothalamus. 相似文献

As a result of histological investigations of seasonal dynamics in the anatomical-tissue organization of the litoral sponge Halichondria panicea (Pallas), morphogenetical processes during different periods of its life cycle are described in detail. Correlations between different phases of sexual reproduction and conditions of anatomical and tissue organization in the maternal organism are stated. It is noted that tissue reduction of the definitive sponge Halichondria panicea during autumn--winter season is a natural phenomenon, regularly repeating in the life of the litoral sponge investigated, as a result of the maternal organism's "exhaustion" in the period of sexual embryogenesis. 相似文献

The keeping of guinea pigs in a thermochamber at 37 degrees C for 3 days resulted in increased releasing of neurohormones of the posterior lobe of the hypophysis. By the end of the 3d day there was no substantial difference between the average logarhythm of the volume of cell nuclei of the supraoptic nucleus of experimental animals and that of intact animals. Injection of ACTH after a preliminary three-day presence of the animal in the thermochamber caused a pronounced inhibition of the hypothalamo-hypophyseal neurosecretory system (HHNS). The HHNS of animals subjected to 3-day-long continuous exposure to high temperature, in contrast to the HHNS of intact animals is not capable to increase its activity in responses to stress (immobilization). 相似文献
