Participation of nitrilotriacetic acid degrading bacterial strain NTA-1 in the continuous-cultivated mixed culture was studied under different conditions including predation pressure of the ciliate Dexiostoma campyla (STOKES , 1886). From the viewpoint of dispersed/flocculated biomass distribution, significant relationships between NTA-1 and total bacteria ratio, and dispersed and total biomass ratio were proved in the systems without high concentrations of ciliates. The ciliate concentrations reaching 104 ml−1 stabilized flocculated biomass growth without directly affecting NTA-1 portion. Using fluorescently labelled NTA-1 bacteria, filter feeding rates of ciliates were evaluated (maximum individual uptake rate upon NTA-1 bacteria as a number of bacteria per ciliate per hour being 120 h−1 and 260 h−1 under ciliate division rate of 0.3 day−1 and 1 day−1, respectively). Biomass balance showed that dispersed NTA-1 bacteria should not serve as the sole feeding source for these free-swimming ciliates. The role of diversity of mixed bacterial diet in ciliate growth and the role of ciliate predation in stabilizing bacterial assemblage structure was proved.  相似文献

Poly(ADP-ribose) glycohydrolase (PARG) is the major enzyme responsible for the catabolism of poly(ADP-ribose), a reversible covalent-modifier of chromosomal proteins. Purification of PARG from many tissues revealed heterogeneity in activity and structure of this enzyme. To investigate PARG structure and localization, we developed a highly sensitive one-dimensional zymogram allowing us to analyze PARG activity in crude extracts of Cos-7, Jurkat, HL-60, and Molt-3 cells. In all extracts, a single PARG activity band corresponding to a protein of about 110 kDa was detected. This 110-kDa PARG activity was found mainly in cytoplasmic rather than in nuclear extracts of Cos-7 cells.  相似文献

Although cytokinins (CKs) affect a number of processes connected with chloroplasts, it has never been rigorously proven that chloroplasts contain CKs. We isolated intact chloroplasts from tobacco (Nicotiana tabacum L. cv SR1) and wheat (Triticum aestivum L. cv Ritmo) leaves and determined their CKs by liquid chromatography/tandem mass spectroscopy. Chloroplasts from both species contained a whole spectrum of CKs, including free bases (zeatin and isopentenyladenine), ribosides (zeatin riboside, and isopentenyladenosine), ribotides (isopentenyladenosine-5'-monophosphate, zeatin riboside-5'-monophosphate, and dihydrozeatin riboside-5'-monophosphate), and N-glucosides (zeatin-N(9)-glucoside, dihydrozeatin-N(9)-glucoside, zeatin-N(7)-glucoside, and isopentenyladenine-N-glucosides). In chloroplasts there was a moderately higher relative amount of bases, ribosides, and ribotides than in leaves, and a significantly increased level of N(9)-glucosides of zeatin and dihydrozeatin. Tobacco and wheat chloroplasts were prepared from leaves at the end of either a dark or light period. After a dark period, chloroplasts accumulated more CKs than after a light period. The differences were moderate for free bases and ribosides, but highly significant for glucosides. Tobacco chloroplasts from dark-treated leaves contained zeatin riboside-O-glucoside and dihydrozeatin riboside-O-glucoside, as well as a relatively high CK oxidase activity. These data show that chloroplasts contain a whole spectrum of CKs and the enzymatic activity necessary for their metabolism.  相似文献

Durable tumor cell eradication by chemotherapy is challenged by the development of multidrug-resistance (MDR) and the failure to induce immunogenic cell death. The aim of this work was to investigate whether MDR and immunogenic cell death share a common biochemical pathway eventually amenable to therapeutic intervention. We found that mevalonate pathway activity, Ras and RhoA protein isoprenylation, Ras- and RhoA-downstream signalling pathway activities, Hypoxia Inducible Factor-1alpha activation were significantly higher in MDR+ compared with MDR− human cancer cells, leading to increased P-glycoprotein expression, and protection from doxorubicin-induced cytotoxicity and immunogenic cell death. Zoledronic acid, a potent aminobisphosphonate targeting the mevalonate pathway, interrupted Ras- and RhoA-dependent downstream signalling pathways, abrogated the Hypoxia Inducible Factor-1alpha-driven P-glycoprotein expression, and restored doxorubicin-induced cytotoxicity and immunogenic cell death in MDR+ cells. Immunogenic cell death recovery was documented by the ability of dendritic cells to phagocytise MDR+ cells treated with zoledronic acid plus doxorubicin, and to recruit anti-tumor cytotoxic CD8+ T lymphocytes. These data indicate that MDR+ cells have an hyper-active mevalonate pathway which is targetable with zoledronic acid to antagonize their ability to withstand chemotherapy-induced cytotoxicity and escape immunogenic cell death.  相似文献

Mutations in the gene for polycystin 2 (Pkd2) lead to polycystic kidney disease, however the main cause of mortality in humans is cardiac related. We previously showed that 5 month old Pkd2+/- mice have altered calcium-contractile activity in cardiomyocytes, but have preserved cardiac function. Here, we examined 1 and 9 month old Pkd2+/- mice to determine if decreased amounts of functional polycystin 2 leads to impaired cardiac function with aging. We observed changes in calcium handling proteins in 1 month old Pkd2+/- mice, and these changes were exacerbated in 9 month old Pkd2+/- mice. Anatomically, the 9 month old Pkd2+/- mice had thinner left ventricular walls, consistent with dilated cardiomyopathy, and the left ventricular ejection fraction was decreased. Intriguingly, in response to acute isoproterenol stimulation to examine β-adrenergic responses, the 9 month old Pkd2+/- mice exhibited a stronger contractile response, which also coincided with preserved localization of the β2 adrenergic receptor. Importantly, the Pkd2+/- mice did not have any renal impairment. We conclude that the cardiac-related impact of decreased polycystin 2 progresses over time towards cardiac dysfunction and altered adrenergic signaling. These results provide further evidence that polycystin 2 provides a critical function in the heart, independent of renal involvement.  相似文献

The 1999 NATO bombing of the oil refinery in Novi Sad (Yugoslavia) has heavily contaminated the Danube River and its sediments, as well as the surrounding soil and groundwater. The destruction of the factories released 73,569 tons of crude oil of which 90% was incinerated, 560 tons reached the Danube River, and the remainder was spilled onto the soil. The contents of oil and oil derivatives in the soil were in the range of 3 to 42,000?mg/kg. The first soil layer contained an average of 67,000?mg/kg of crude oil and oil derivatives. The layers beneath it, above the groundwater table, contained 56?ml/l of free oil derivatives in the drained water. The spreading of this pollution could imperil the groundwater quality in the water supply zone because the refinery is located in the hinterland of the zone. The quality of water and sediment samples was monitored from April 1999 to November 2000 by measuring concentrations of hydrocarbons and polyaromatic hydrocarbons (PAH). The hydrocarbon content in the Danube River water in October 2000 was about 20% of the value measured at the time of the accident. Immediately after the accident the concentration of mineral oil in the surface sediment was in the range of 0.11 to 0.29?g/kg. At the same time PAH concentrations in the river sediment were up to 160?mg/kg, depending on the sampling site location. The values showed a decrease in the course of further monitoring.  相似文献

A novel method for initiating somatic embryogenesis in grapevine, based on immature whole flower culture, is presented. The embryogenic competence of flowers was compared to that of anthers and ovaries, the most widely used explant types, for five grapevine cultivars. Both the genotype and the explant source affected the differentiation of somatic embryos. The highest percentages of embryogenesis were obtained in ovary-derived calli from all cultivars tested with the exception of Brachetto a grappolo lungo. Whole flowers proved to be suitable material for initiating embryogenic cultures for most tested cultivars, and for 110 R, Chardonnay, and Grignolino they gave similar or better results than anthers. Collection of whole flowers from the inflorescence is easier and faster than excision of anthers and ovaries from the flower itself; it can be done without the use of a stereomicroscope and damage to the explant is unlikely. No morphological difference was noted among embryogenic cultures originated from ovaries, flowers, or anthers.  相似文献
