Microcrustacean species (Cladocera, Copepoda) as a significant component of the plankton assembly play diverse roles in floodplain ecosystems and cope with the heterogenic environment differently. Since alterations in the hydro regime are considered disturbance events for zooplankton development, we assessed whether and to what extent are Cladocera and Copepoda exposed to disturbances caused by hydro regime alterations in a floodplain lake. To evaluate the disturbance levels for individual microcrustacean assemblage in different water layers and between different hydrological phases, we applied the abundance/biomass comparison (ABC) method. Recorded microcrustacean assemblages highly depended on the hydrological state of the waterbody and diverged accordingly. Cladocera demonstrated to be more sensitive to change in specific environmental parameters like conductivity, temperature and transparency, while Copepoda exhibited no limitation by ecological variables. Although both groups were influenced by the bottom–up and top–down control, different disturbance intensity was recorded for the individual microcrustacean class. An undisturbed condition during isolation of the Lake allowed Cladocera to reach equilibrium while both microcrustacean groups exhibited moderate to high levels of disturbance following inundation. Regarding disturbance intensity, Cladocera expressed a weak distinction following hydro regime and water layer replacement. On the other hand, Copepoda disturbance levels notably depended on the position in the water column within a specific hydrological phase. Results of the ABC analyses gave a good representation of a real community dispersal in the studied Lake. However, more research is needed to re-evaluate here given levels of disturbance properly.  相似文献

A comparative study of amyloid properties of the aggregates of smooth muscle titin (SMT) from chicken gizzard was carried out. These aggregates were formed in two solutions: 0.15 M glycine-KOH, pH 7.2–7.4 (SMT(Gly)) and 0.2 M KCl, 10 mM imidazole, pH 7.0 (SMT(KCl)). Electron microscopy data showed that SMT aggregates has an amorphous structure in both cases. The results of atomic-force microscopy demonstrated slight differences in morphology in two types of aggregates. The SMT(Gly) aggregates were represented as branching chains, composed of spherical aggregates approximately 300–500 nm in diameter and up to 35 nm in height. The SMT(KCl) aggregates formed sponge-like structures with strands of 8–10 nm in height. Structural analysis of SMT aggregates by X-ray diffraction revealed the presence of cross-β-sheet structure in the samples under study. In the presence of SMT(Gly) aggregates, thioflavine T fluorescence intensity was higher (~3-fold times) compared with that in the presence of SMT(KCl) aggregates. Congo red-stained SMT(Gly) aggregates had yellow to apple-green birefringence under polarized light, which was not observed for SMT(KCl) aggregates. Dynamic light scattering data showed the similar rate of aggregation for both types of aggregates, though SMT(KCl) aggregates were able to partially disaggregate under increased ionic strength of the solution. The ability of SMT to aggregation followed by disaggregation may be functionally significant in the cell.  相似文献

The beet armyworm Spodoptera exigua (Hübner) (Lepidoptera: Noctuidae) is a polyphagous insect that is distributed worldwide and was recently reported as an important pest on African indigenous vegetables. Cotesia icipe Fernandez‐Triana & Fiaboe (Hymenoptera: Braconidae) is a recently described parasitoid, reported from various Afrotropical countries. This work investigated the performance of C. icipe on S. exigua infesting Amaranthus dubius Mart. ex Thell. under laboratory conditions. Cotesia icipe was aggressive on the host and successfully oviposited on S. exigua with 70% of parasitoid females ovipositing after 2 hr of exposure. Parasitoid densities significantly affected the parasitism rate and the nonreproductive larval mortality. Parasitism rate was 9.7 ± 0.8% and 59.5 ± 3.1% for a single and cohort of five females released, respectively, when offered 50 host larvae. The cohort female release resulted in significantly higher larval nonreproductive mortality than the single release. However, there was no significant difference between parasitoid release densities in regard to pupal nonreproductive mortality. The larval and pupal mortalities in the presence of C. icipe were significantly higher than the natural mortalities at both parasitoid release densities. The parasitoid sex ratio was female‐biased for the cohort females but balanced when a single female was released. The hind tibia and forewing lengths were not affected by the density of female parasitoids but there were variations according to sex. The implication of these findings on the potential use of C. icipe for biological control of S. exigua in amaranth production systems is discussed.  相似文献

The amiloride-sensitive epithelial Nachannel (ENaC) is a heteromultimeric channel made of three αβγ subunits. The structures involved in the ion permeation pathway have only been partially identified, and the respective contributions of each subunit in the formation of the conduction pore has not yet been established. Using a site-directed mutagenesis approach, we have identified in a short segment preceding the second membrane-spanning domain (the pre-M2 segment) amino acid residues involved in ion permeation and critical for channel block by amiloride. Cys substitutions of Gly residues in β and γ subunits at position βG525 and γG537 increased the apparent inhibitory constant (K i) for amiloride by >1,000-fold and decreased channel unitary current without affecting ion selectivity. The corresponding mutation S583 to C in the α subunit increased amiloride K i by 20-fold, without changing channel conducting properties. Coexpression of these mutated αβγ subunits resulted in a nonconducting channel expressed at the cell surface. Finally, these Cys substitutions increased channel affinity for block by externalZn2+ ions, in particular the αS583C mutant showing a K i for Zn2+of 29 μM. Mutations of residues αW582L or βG522D also increased amiloride K i, the later mutation generating a Ca2+blocking site located 15% within the membrane electric field. These experiments provide strong evidence that αβγ ENaCs are pore-forming subunits involved in ion permeation through the channel. The pre-M2 segment of αβγ subunits may form a pore loop structure at the extracellular face of the channel, where amiloride binds within the channel lumen. We propose that amiloride interacts with Na+ions at an external Na+binding site preventing ion permeation through the channel pore.  相似文献

Focal adhesions are sites for integrin-mediated attachment of cultured cells to the extracellular matrix. Localization studies have shown that focal adhesions can be stained by antiphosphotyrosine antibodies, but the role of tyrosine-phosphorylated proteins in focal adhesions is not known. By using ventral plasma membranes prepared from chicken embryo fibroblasts spread on the substrate, we present evidence for the preferential localization of a minor pool of tyrosine-phosphorylated paxillin in focal adhesions. Ventral plasma membranes showed an enrichment in β1-integrins, and in several tyrosine-phosphorylated polypeptides, while focal adhesion proteins like vinculin and paxillin, although localized to focal adhesions in ventral plasma membranes, were not particularly enriched in these preparations compared to whole cell lysates. Biochemical and morphological analysis of ventral plasma membranes showed a dramatic increase in the level of tyrosine-phosphorylation of the pool of paxillin localized to the adhesive sites, when compared to the paxillin present in whole cell lysates. The observed preferential localization of tyrosine-phosphorylated paxillin to focal adhesions may represent a general mechanism to compartmentalize focal adhesion components from large non-phosphorylated, cytosolic pools.  相似文献
