Molecular cytogenetic study of Robertsonian translocation 13;14 and Down syndrome in a 3-year-old infant

We describe here a rare case of Robertsonian translocation 13;14 of maternal origin combined with regular trisomy 21 (46,XX,der(13;14)(q10;q10)mat,+21) with Down syndrome phenotype. Molecular cytogenetic studies allowed us to determine the maternal origin of additional chromosome 21 and the non-disjunction of chromosome 21 to occur in meiosis I. On the basis of data obtained we discuss the possible involvement of structural alterations of chromosomes 13 and 14 in the chromosome 21 non-disjunction.     

Structural analysis of the Alu-containing DNA fragment with disperse distribution in human chromosomes

Nucleotide sequencing of two terminal subfragments of a cloned human DNA fragment has been done. The fragment cloned hybridized dispersively along all chromosomes except for Y chromosome and C heterochromatic chromosome regions. Both subfragments sequenced contain Alu repeats, whose structures differ partially from those of accurately determined sequences of human Alu repeats. The results obtained are discussed in respect of possible usage of Alu repeats containing sequences for construction of special polymorphic molecular markers of human chromosomes.     

Mutagenic effect on Escherichia coli bacteria of 8-hydroxy-2'-deoxyguanosine--a DNA base damage product induced by oxygen radicals and ionizing radiation]

The effect of 8-oxo-2'-deoxyguanosine (8-oxo-dG) (8-hydroxydeoxyguanosine)--a DNA base damage product induced by oxygen radicals and irradiation on survival and mutagenesis in Escherichia coli strains C-600 and P-687 was investigated. Survival and mutagenesis curves, in dependence of 8-oxo-dG concentrations in the medium, ranging from 0.2 through 10 mM, were obtained. Bacterial survival at all 8-oxo-dG concentrations tested was shown to be no lesser than in the control. The mutagenic effect of 8-oxo-dG was tested by frequency of reversions in the absence of leucine and threonine. A non-linear dependence of mutagenesis on the concentration was observed. Linear increase in the amount of revertants took place at concentrations of 8-oxo-dG lower than 1 mM, and being kept constant at higher concentrations. Induction of SOS repair under the action of 8-oxo-dG in E. coli PQ37 strain was estimated according to alteration of activity of beta-galactosidase in the SOS chromotest. Weak induction of the SOS response was observed within the wide range of 8-oxo-dG concentration values, which points to a lack of genotoxicity and independence of mutagenesis on SOS repair.     

Biological activity of gamma-irradiated serum albumin

Gamma-irradiated serum albumin activated L-tyrosine oxidation to 3,4-dioxyphenyl alanine (DOPA) and forms adducts with DOPA oxidation products. These adducts are more resistant to proteolysis and have bactericidal and mutagenic capacity. A possible role of such adducts in a radiation damage to the organism is discussed.     

The nature of the toxic properties of the blood plasma of gamma irradiated rats

In addition to the growth of low molecular weight quinoid radiotoxins the toxic effects of albumin were registered in rat plasma 4-24 h following gamma-irradiation. It is assumed that adducts of albumin with quinoid radiotoxins are formed and contribute to the development of radiation injury to critical organism tissues.     

Serotonin type 2a (5-HTR2A) receptor gene polymorphism and personality traits in patients with endogenous psychoses]

Genetic polymorphism of the serotonin receptor (5-HTR2A) gene has been reported to be associated with the expression of clinical signs characteristic of major psychoses, including schizophrenia and affective disorders. In this study, personality traits of patients with these diseases and the associations of these traits with 5-HTR2A allelic polymorphisms were studied. It was demonstrated that schizophrenic and affective patients with the 2/2 genotype of serotonin receptor had lower scores on the anxiety scale and on the anxiety-related hypochondriasis and neuroticism scales than subjects with the 1/1 and 1/2 genotypes.     

Comparative characteristics of systemic and pulmonary hemodynamics during changes in the heart preloading

A greater degree of relative shifts in the systemic arterial pressure in enhancing the right heart pre-load as compared with its diminishing. A primary role of the compensation mechanisms of the enhanced systemic arterial pressure level. The main role in the compensation of integral shifts of the arterial pressure induced by changes in the heart pre-load was shown to belong to the vascular resistance both in the major and the minor circulation circles. An idea of a greater involvement of the capacity function of the vascular bed in the minor circulation circle as compared with that in the major circulation circle in systemic haemodynamic shifts in changes of the heart pre-load, has been advanced.     

Use of a cloned alphoid repetitive sequence of human DNA in studying the polymorphism of heterochromatin regions of chromosomes

Chromosomal location of the cloned fragment pHS05 of alphoid DNA from the collection of human PstI restricts has been studied in 38 individuals by in situ hybridization. Pericentromeric localization of the DNA fraction studied was found in practically all chromosomes of the set. Significant interchromosomal and poorly expressed interindividual differences were detected in a number of the copies of the sequence class investigated. The majority of the label (approx. 27%) was observed over the pericentromeric region of chromosome 3. No relationship was discovered between hybridization results and the pattern of Q-polymorphism.
