The gene for a pectate lyase of E. chrysanthemi ENA49 cloned in a recombinant plasmid pPTL1 (a derivative of RSF1010) was transferred into E. carotovora. The pectate lyase determined by the cloned gene was secreted into the cultural medium from the cells of E. crysanthemi EC16. Partial secretion of the enzyme was registered for E. carotovora cells. The major part of EC1 E. chrysanthemi pectate lyase synthesized by E. carotovora cells is accumulated in periplasmic and cytoplasmic fractions. The obtained results suggest the different specificity or efficiency of pectate lyase secretion systems in the studied Erwinia strains.  相似文献

The prohibition of copulations among near relatives may raise the fitness of population. This effect being irregular and insignificant for a distinct generation, becomes apparent in evolutionary time intervals through the natural selection of populations with incest-taboo. The "characteristic selection time" theta depends on typical population size, genetic damage and the mean rate of population growth. The estimation obtained for theta permit us to assert that the model describes the phenomenon of "socio-cultural selection" in prehistory. The model shows the demographic specificity of small populations. The problem of the number of consanguineous marriages is considered in detail. New explanation for deviation of the observed frequency of consanguineous marriages from classical estimations is proposed.  相似文献

The biosynthesis of riboflavin precursor 6,7-dimethyl-8-ribityllumazine was studied in extracts of Pichia guilliermondii yeast mutants of rib5 and rib6 genotypes with impaired synthesis of proteins P1 and P2, respectively. It was shown that synthesis of 6,7-dimethyl-8-ribityllumazine took place in extracts of rib5 mutant (active P1 protein) in the presence of 2,4-dihydroxy-5-amino-6-ribitylaminopyrimidine and the compound formed from ribose-5-phosphate by extracts of rib6 mutant (active P2 protein). No lumazine was formed in extracts of rib6 mutant from pyrimidine substrate and ribose-5-phosphate preincubated with extracts of rib5 mutant. Hence, P1 protein (the product of RIB5 gene) participates in the biosynthesis of 6,7-dimethyl-8-ribityllumazine from 2,4-dihydroxy-5-amino-6-ribitylaminopyrimidine and aliphatic intermediate which is formed from ribose-5-phosphate, under the action of P2 protein (the product of RIB6 gene).  相似文献

Dynamics of allele frequencies for the B-locus of blood groups in cattle populations and of a number of phenotype indexes in these animals was studied. It is determined that natural stabilizing selection and prezygotic selection, leading to a change in genetic structure, act in every population. Mutants of Streptomyces tenebrarius with the blocked synthesis of 3'-deoxykanamycin B were obtained by treating the producer with NTG and chloramphenicol, or after gamma-irradiation. These mutants (idiotrophs) were distributed into three groups by means of the cosynthesis experiments on agar plates: convertors, secretors and "neutral" strains. Five idiotrophs represented five complementation groups for biosynthesis of the antibiotic. Three of these were defective in 2-deoxystreptamine synthesis, the fourth was defined as neamine-negative, and the fifth was probably blocked in regulation of enzymes responsible for conversion of neamine or paromamine into kanamycins. Localization of mutations has been shown on the scheme of kanamycins' biosynthesis.  相似文献

The mutagenic activity of two known carcinogens (benzo(a) pyrene and 2-acetylaminofluorene) and that of two structurally closely related but not carcinogenic compounds (pyrene and 4-acetylaminofluorene) was examined by the Muller-5 test for sex-linked recessive lethals (SRL). The chemicals tested were applied to the food medium for larvae of Canton-S Drosophila melanogaster. No statistically significant differences in frequencies of induced SRL were found either within pairs of chemicals or between treated and untreated animals.  相似文献

This study deals with the analysis of body weight and length augmentation in the group of newborns survived and died during the prenatal period. The effect of stabilizing selection for these characters was demonstrated. The analysis of dependence of the innate characters on the order number of pregnancy and parturition is accomplished. The advantages of the use of generalized characters of variability for revealing the stabilizing selection and of the possibility of using anthropometric characters of newborns in view of genetic monitoring are discussed.  相似文献

Substance P (10 and 100 ng/ml) stimulates the proliferation of basal cells from rat's tongue epithelium in primary cultures with 2.5% fetal calf serum. In serum-free medium substance P have no effect on the epithelial cell growth. This neuropeptide secreted by afferent nerve fibers of the tongue epithelium is suggested to have a neurotrophic influence on epithelial cells controlling their proliferation.  相似文献

The dynamics of cAMP and cGMP content of the thymus homogenate from developing chick embryos and chickens was studied during ante- and postnatal development. Changes in the content of cyclic nucleotides bear an oscillatory pattern. At the early stages of embryogenesis (9 to 11 days of incubation) the content of cyclic nucleotides was low and gradually increased by 13 days of incubation. As the development proceeded, the quantitative and qualitative rearrangement of the thymus cellular composition reflected in changes in the content of cyclic nucleotides. At the same time the curves of cyclic nucleotide content became antiphasic. These reciprocal cAMP to cGMP ratios might reflect the cyclic and synchronous reproduction and functional development of the main bulk of the thymus cellular elements. The maximum content of cAMP and the minimum content of cGMP were recorded on the 17th day of embryogenesis.  相似文献

The results obtained mainly by the author and coworkers are summarized. One efficient method to detect free radicals in biological samples is chemiluminescence (CL). In the absence of activators CL of membraneous systems is due to lipid peroxide free radicals, whereas in the presence of luminol it is initiated by oxygen radicals. Low levels of free radicals in the cells and blood plasma are maintained by antioxidants, enzymes included. Ferrous ions increase free radical concentrations in the cells and tissues. Deleterious action of hydroxyl radicals is the result of the breakage of DNA strains and of lipid peroxidation (LPO). The latter reaction brings about the damage of the membrane barriers due to a decrease of the electrical stability of the membrane lipid bilayer and "self-breakdown" of the membranes by potential differences produced in the living cells.  相似文献
