Phytostabilization is a promising option for the remediation of metal contaminated soils which requires the implementation of long-term monitoring programs. We here propose to incorporate the paradigm of “adaptive monitoring”, which enables monitoring programs to evolve iteratively as new information emerges and research questions change, to metal phytostabilization. Posing good questions that cover the chemical, toxicological and ecological concerns associated to metal contaminated soils is critical for an efficient long-term phytostabilization monitoring program. Regarding the ecological concerns, soil microbial parameters are most valuable indicators of the effectiveness of metal phytostabilization processes in terms of recovery of soil health. We suggest to group soil microbial parameters in higher-level categories such as “ecological attributes” (vigor, organization, stability) or “ecosystem services” in order to facilitate interpretation and, most importantly, to provide long-term phytostabilization monitoring programs with the required stability through time against changes in techniques, methods, interests, etc. that will inevitably occur during the monitoring program. Finally, a Phytostabilization Monitoring Card, based on both ecological attributes and ecosystem services, for soil microbial properties is provided.  相似文献

A study of the potential of autohydrolysis and alkaline extraction processes from corn stalks was performed for high purity hemicellulose extraction. The influence of process parameters on the purity of obtained hemicelluloses was analyzed. An experimental design was developed for the autohydrolysis treatments to determine the optimal conditions to solubilize the hemicelluloses with lowest content in contaminants. On the other hand, alkaline extraction, including raw material pretreatment (dewaxing and delignification step) was carried out analyzing the effectiveness of this processes for maximum pure hemicellulose recovery. The maximum yield (54% of the raw material hemicelluloses) and the best physicochemical properties (highest hemicellulose content free of lignin) were obtained with these pretreatments in alkaline extraction. Moreover, the effect of lignin removal by sulfuric acid from the autohydrolysis liquors before hemicellulose precipitation was studied. This purification step has allowed to obtain lignin-free autohydrolysis hemicellulose but with the presence of sulfur as predominant contaminant.  相似文献

A number of medium constituents were evaluated in an attempt to improve somatic embryo production in Medicago arborea ssp. arborea, using cotyledons, petioles and leaves as explants. Two culture steps were applied: in the first stage (2 months), Murashige–Skoog (MS) medium was used, containing 2,4 dichlorophenoxyacetic acid (9M 2,4-D) and kinetin (9 M KIN) together with different nitrogen sources (alanine, glutamine, proline or tryptophan (2.5 and 5 mM); casein hydrolysate (100, 500 and 1000 mg l–1; nitrate (4.69 and 9.39 mM) or casein hydrolysate (100 mg l–1) and nitrate (4.69 mM)), polyalcohols (mannitol at 164 and 328 mM or sorbitol at 219 and 438 mM), sucrose (43.8 and 175.4 mM) or calcium (1.5 and 6 mM). In the second stage (3 months of cultivation), calli were transferred to a kinetin-free MS medium with 2,4-D (2.25 M) only. The inclusion of proline (2.5 mM) was the most effective treatment for the induction of somatic embryos, with the petiole being the best explant. Treatment with casein hydrolysate (100 mg l–1) also improved the embryonic efficiency. The rest of the treatments neither affect nor inhibit the embryonic response.A special treatment with sorbitol (219 mM) in the second stage of cultivation produced a slight increase in embryogenesis, but less than that obtained with proline.  相似文献

TrwB is an integral membrane protein linking the relaxosome to the DNA transport apparatus in plasmid R388 conjugation. Native TrwB has been purified in monomeric and hexameric forms, in the presence of dodecylmaltoside from overexpressing bacterial cells. A truncated protein (TrwBDeltaN70) that lacked the transmembrane domain could be purified only in the monomeric form. Electron microscopy images revealed the hexameric structure and were in fact superimposable to the previously published atomic structure for TrwBDeltaN70. In addition, the electron micrographs showed an appendix, approximately 25 A wide, corresponding to the transmembrane region of TrwB. TrwB was located in the bacterial inner membrane in agreement with its proposed coupling role. Purified TrwB hexamers and monomers bound tightly the fluorescent ATP analogue TNP-ATP. A mutant in the Walker A motif, TrwB-K136T, was equally purified and found to bind TNP-ATP with a similar affinity to that of the wild type. However, the TNP-ATP affinity of TrwBDeltaN70 was significantly reduced in comparison with the TrwB hexamers. Competition experiments in which ATP was used to displace TNP-ATP gave an estimate of ATP binding by TrwB (K(d)((ATP)) = 0.48 mm for hexamers). The transmembrane domain appears to be involved in TrwB protein hexamerization and also influences its nucleotide binding properties.  相似文献

This paper deals with the syntaxonomy of the classThero-Salicornietea in the Iberian Peninsula, falling particularly upon the significance that taxonomy of the generaSalicornia andSuaeda has on it. Three alliances were differentiated, two Atlantic and one Mediterranean. The Atlantic ones wereSalicornion dolichostachyo-fragilis, with four associations grouping slikke communities with tetraploid glasswort (Salicornia) species, andSalicornion europaeo-ramosissimae, with three associations of schorre with generally diploid glasswort species. The Mediterranean allianceSalicornion patulae had six associations distributed both in coastal (southern and eastern coasts) and inland salt marshes. A complete syntaxonomy of the class, maps of the distribution of its associations and tables summarizing their floristic composition are also provided.  相似文献
