Exogenous nitric oxide triggers classic ischemic preconditioning by preventing intracellular Ca2+ overload in cardiomyocytes 总被引:1,自引:0,他引:1
The involvement of nitric oxide (NO) in the late phase of ischemic preconditioning is well established. However, the role of NO as a trigger or mediator of "classic preconditioning" remains to be determined. The present study was designed to investigate the effects of NO on calcium homeostasis in cultured newborn rat cardiomyocytes in normoxia and hypoxia. We found that treatment with the NO donor, sodium nitroprusside (SNP) induced a sustained elevation of intracellular calcium level ([Ca(2+)](i)) followed by a decrease to control levels. Elevation of extracellular calcium, which generally occurs during ischemia, caused an immediate increase in [Ca(2+)](i) and arrhythmia in cultures of newborn cardiomyocytes. Treatment with SNP decreased [Ca(2+)](i) to control levels and re-established synchronized beating of cardiomyocytes. A decrease in extracellular [Na(+)], which inhibits the Na(+)/Ca(2+) exchanger, did not prevent [Ca(2+)](i) reduction by SNP. In contrast, application of thapsigargin, an inhibitor of sarcoplasmic reticulum Ca(2+)-ATPase (SERCA2a), increased [Ca(2+)](i), and in its presence, SNP did not reduce [Ca(2+)](i), indicating that Ca(2+) reduction is achieved via activation of SERCA2a. The results obtained suggest that activation of SERCA2a by SNP increases Ca(2+) uptake into the sarcoplasmic reticulum (SR) and prevents cytosolic Ca(2+) overload, which might explain the protective effect of SNP from hypoxic damage. 相似文献

Most studies of European genetic diversity have focused on large-scale variation and interpretations based on events in prehistory, but migrations and invasions in historical times could also have had profound effects on the genetic landscape. The Iberian Peninsula provides a suitable region for examination of the demographic impact of such recent events, because its complex recent history has involved the long-term residence of two very different populations with distinct geographical origins and their own particular cultural and religious characteristics—North African Muslims and Sephardic Jews. To address this issue, we analyzed Y chromosome haplotypes, which provide the necessary phylogeographic resolution, in 1140 males from the Iberian Peninsula and Balearic Islands. Admixture analysis based on binary and Y-STR haplotypes indicates a high mean proportion of ancestry from North African (10.6%) and Sephardic Jewish (19.8%) sources. Despite alternative possible sources for lineages ascribed a Sephardic Jewish origin, these proportions attest to a high level of religious conversion (whether voluntary or enforced), driven by historical episodes of social and religious intolerance, that ultimately led to the integration of descendants. In agreement with the historical record, analysis of haplotype sharing and diversity within specific haplogroups suggests that the Sephardic Jewish component is the more ancient. The geographical distribution of North African ancestry in the peninsula does not reflect the initial colonization and subsequent withdrawal and is likely to result from later enforced population movement—more marked in some regions than in others—plus the effects of genetic drift. 相似文献

The continuity of abiotically formed bilayer membraneswith similar structures in contemporary cellular life,and the requirement for microenvironments in whichlarge and small molecules could be compartmentalized, support the idea that amphiphilic boundary structurescontributed to the emergence of life. As an extensionof this notion, we propose here a `Lipid World'scenario as an early evolutionary step in theemergence of cellular life on Earth. This conceptcombines the potential chemical activities of lipidsand other amphiphiles, with their capacity to undergospontaneous self-organization into supramolecularstructures such as micelles and bilayers. Inparticular, the documented chemical rate enhancementswithin lipid assemblies suggest that energy-dependentsynthetic reactions could lead to the growth andincreased abundance of certain amphiphilic assemblies.We further propose that selective processes might acton such assemblies, as suggested by our computersimulations of mutual catalysis among amphiphiles. Asdemonstrated also by other researchers, such mutualcatalysis within random molecular assemblies couldhave led to a primordial homeostatic system displayingrudimentary life-like properties. Ethylene involvement in germination of Striga hermonthica (Del.) Benth., an important root parasitic weed on poaceous crops, was investigated at the physiological and molecular levels. Seeds, conditioned at 30°C for 14 days, were treated with ethylene, ethephon or 1-aminocyclopropane-1-carboxylic acid (ACC). Ethylene consistently induced low germination. Ethephon and ACC effectively stimulated germination at concentrations of 0.01 and 1 m M , respectively. In contrast to ethylene, both ethephon and ACC acted in a concentration-dependent manner. Germination induced by the synthetic strigolactone GR24 was inhibited by aminoethoxyvinylglycine (AVG) and 1-methylcyclopropene. ACC reversed the inhibition caused by AVG. When seeds were treated with GR24 in sealed vials, ethylene concentration in headspace gas increased prior to the onset of germination. Total RNA extracted from germinating seeds 12 h after GR24 treatment was used for PCR-based amplification of cDNA fragments encoding the ACC synthase- and oxidase-active site domains. Two distinct cDNA fragments encoding ACC synthase ( SHACS1 and SHACS2 ) and one encoding ACC oxidase ( SHACO1 ) were cloned and sequenced. Southern analysis suggested that each of the cloned genes was present as a single copy in the genome of S. hermonthica . Northern analyses showed that SHACS1 exhibited a temporal change in expression peaking at 10 h after GR24 treatment, which coincided with a steady increase in ethylene concentration. SHACS2 was expressed at a low level with a similar trend. SHACO1 exhibited a temporal change in expression peaking at 15 days during conditioning, when seed response to GR24 was maximal. In summary, expression of ACC synthase and ACC oxidase genes was found to be responsive to a germination stimulant and to conditioning, respectively. The implications of these findings with respect to germination of S. hermonthica under field conditions are discussed. 相似文献

Arabidopsis thaliana is frequently grown on semisolid medium in Petri dishes, for various experiments that usually consist of two stages on two distinct growth media. Seedlings are germinated under favorable conditions followed by their transfer to another medium containing the given treatment(s). This often causes secondary effects on seedlings due to root shock, or direct and unavoidable contact of the shoot with the second medium. We have developed a simple and efficient method for the transfer of seedlings grown on semisolid medium with minimal damage. In this double-agar-layer method, seeds are germinated on a thin growth-medium-containing agar layer. Subsequently, medium blocks containing the embedded seedlings are excised and placed on the second semisolid medium supplemented with the treatment agent. Differential agar concentrations allow easy penetration of the roots into the second medium, but do not allow the shoots to come into contact with it. This unique method offers several advantages over others that are in common use, in which the seedlings are individually transferred to the second medium or alternatively grown on transfer-carrier matrices, such as filter paper, mesh and cellophane. In the presented method, the entire root system faces the growth medium, the shoots are surrounded by air at all growth stages and transfer of the seedlings is much easier. In addition, a large number of seedlings can be transferred in a single step, without stressing the plants or damaging the delicate root system. The major function of the Haptoglobin (Hp) protein is to control trafficking of extracorpuscular hemoglobin (Hb) thru the macrophage CD163 receptor with degradation of the Hb in the lysosome. There is a common copy number polymorphism in the Hp gene (Hp 2 allele) that has been associated with a severalfold increased incidence of atherothrombosis in multiple longitudinal studies. Increased plaque oxidation and apoptotic markers have been observed in Hp 2-2 atherosclerotic plaques, but the mechanism responsible for this finding has not been determined. We proposed that the increased oxidative injury in Hp 2-2 plaques is due to an impaired processing of Hp 2-2-Hb complexes within macrophage lysosomes, thereby resulting in redox active iron accumulation, lysosomal membrane oxidative injury, and macrophage apoptosis. We sought to test this hypothesis in vitro using purified Hp-Hb complex and cells genetically manipulated to express CD163. CD163-mediated endocytosis and lysosomal degradation of Hp-Hb were decreased for Hp 2-2-Hb complexes. Confocal microscopy using lysotropic pH indicator dyes demonstrated that uptake of Hp 2-2-Hb complexes disrupted the lysosomal pH gradient. Cellular fractionation studies of lysosomes isolated from macrophages incubated with Hp 2-2-Hb complexes demonstrated increased lysosomal membrane oxidation and a loss of lysosomal membrane integrity leading to lysosomal enzyme leakage into the cytoplasm. Additionally, markers of apoptosis, DNA fragmentation, and active caspase 3 were increased in macrophages that had endocytosed Hp 2-2-Hb complexes. These data provide novel mechanistic insights into how the Hp genotype regulates lysosomal oxidative stress within macrophages after receptor-mediated endocytosis of Hb. 相似文献
