Sphingosine-1-phosphate lyase (SGPL1) is the last enzyme in the catabolism of sphingolipids. It catalyzes the retroaldolic cleavage of long chain base phosphates into phosphoethanolamine and a fatty aldehyde. In this article we report on an easy and sensitive procedure to determine SPL activity. The assays uses C17-sphinganine-1-phosphate as substrate and the aldehyde product, pentadecanal, is quantified as its pentafluorobenzyloxime derivative by GC/MS. Derivatization of pentadecanal is performed as a one-step reaction, and the oxime product is directly injected for GC/MS analysis without any further purification. Acquisition in selected ion monitoring mode allows very high sensitivity, with a limit of detection of 281fmol. The assay is linear with both protein concentration and incubation time up to 20μg and 40min, respectively. The K(m) value obtained (6μM) is similar to that for the natural substrate sphingosine-1-phosphate. Using this method, FTY720 and deoxypyridoxine phosphate inhibited SPL with similar potencies to those reported.  相似文献

After several years of research, there is now a consensus that America was populated from Asia through Beringia, probably at the end of the Pleistocene. But many details such as the timing, route(s), and origin of the first settlers remain uncertain. In the last decade genetic evidence has taken on a major role in elucidating the peopling of the Americas. To study the early peopling of South America, we sequenced the control region of mitochondrial DNA from 300 individuals belonging to indigenous populations of Chile and Argentina, and also obtained seven complete mitochondrial DNA sequences. We identified two novel mtDNA monophyletic clades, preliminarily designated B2l and C1b13, which together with the recently described D1g sub-haplogroup have locally high frequencies and are basically restricted to populations from the extreme south of South America. The estimated ages of D1g and B2l, about ~15,000 years BP, together with their similar population dynamics and the high haplotype diversity shown by the networks, suggests that they probably appeared soon after the arrival of the first settlers and agrees with the dating of the earliest archaeological sites in South America (Monte Verde, Chile, 14,500 BP). One further sub-haplogroup, D4h3a5, appears to be restricted to Fuegian-Patagonian populations and reinforces our hypothesis of the continuity of the current Patagonian populations with the initial founders. Our results indicate that the extant native populations inhabiting South Chile and Argentina are a group which had a common origin, and suggest a population break between the extreme south of South America and the more northern part of the continent. Thus the early colonization process was not just an expansion from north to south, but also included movements across the Andes.  相似文献

The spill from the oil tanker Prestige (NW Spain, November 2002) was perhaps the biggest ecological disaster that happened worldwide in the last decades. As a consequence of this catastrophe a general concern led to a huge mobilization of human and technical resources. Given that no information was reported in the scientific literature regarding to the chronic repercussions to human health of exposure to oil spills, a pilot study was performed by our group revealing some increased genotoxic effects in the subjects exposed to the oil during cleaning activities. Due to the seriousness of the results, we extended our study comprising a larger population and including an extensive evaluation of the main polymorphic sites in metabolizing and DNA-repair genes. General increases in micronucleus (MN) frequency and decreases in the proliferation index were observed in individuals with longer time of exposure. Age was a significant predictor of MN frequency. CYP1A1 3'-UTR, EPHX1 codons 113 and 139, GSTP1, GSTM1 and GSTT1 metabolic polymorphisms, and XRCC3 codon 241 and XPD codon 751 repair polymorphisms influenced cytogenetic damage levels. In view of these results, it seems essential to pay more attention to the chronic human health effects of exposure to oil and to focus new studies on such a relevant but overlooked public health field that involves a large number of people all over the world.  相似文献

Area‐wide environmentally friendly pest control methods such as the sterile insect technique (SIT) are being developed and improved to contribute in managing agricultural, environmental and public health problems. A key aspect to evaluate performance of sterile males is to directly measure sterility induction in the field. Sterility induction has been estimated for tephritid fruit flies by recovering egg from host fruit in the field, the method is, however, impractical, and past efforts to develop artificial egg‐laying devices have not prospered. Here, we evaluated response of wild gravid Ceratitis capitata (Medfly) females to long‐distance fruit‐based chemical attractants, visual and tactile stimuli to develop an artificial egg‐laying device. The device combining the most attractive features was further tested under two deployment schemes. Finally, devices and deployment tactics were used to compare fertility levels between feral Medfly females under conventional management and under SIT. Agar spheres wrapped in plastic film, baited with pressed peach juice and visually enhanced with yellow discs received more egg than other combinations of attractive features. Such devices also received more eggs when deployed on fruitless trees and when placed on the orchard perimeter. The egg hatch in an orchard under conventional management was estimated at 86%, whilst egg hatch in an area under SIT was reduced to 31%. The egg‐laying devices are therefore useful to measure sterility induction and can be further improved by refining long‐distance attraction and deployment schemes.  相似文献

Genetic Bit Analysis (GBA) is a relatively new technique developed to score single-nucleotide polymorphisms among alleles. It is currently being used as a genetic diagnostic tool in human paternity tests as well as in pedigree analysis of farm animals. GBA relies on the polymerase chain reaction and enzyme-linked colorimetry to distinguish differences among genotypes. The identification of a single-nucleotide polymorphism which distinguished the plastome of cytoplasmic male-sterile onion varieties from the plastome of fertile lines provided a model system for testing the utility of GBA in plants. Our results demonstrate that GBA permits rapid and accurate allele determination in onion breeding lines, resulting in accurate prediction of sterility at the seedling stage. The use of 96-well microtiter plates and automated liquid-handlers allowed semi-automation of Genetic Bit Analysis.  相似文献

Using fluorescence and SH-groups titration techniques we have proved existence of an inverse relationship betweenthe rate constants of bimolecular deactivation of phosphorylase b coenzyme by iodide ion in the presence of AMP and its analogues and the shielding rate constants of slow cysteine, C-108, by the diverse nucleotides, The greater the shielding rate constants, the smaller the rate constant of deactivation by iodied ion. The analogues we have used were AMP, IMP, 2′dAMP, 2′AMP and 3′AMP.  相似文献

Two irrigation systems were used to compare nitrogen uptake efficiency in citrus trees and to evaluate the NO3 runoff in «Navelina» orange trees [Citrus sinensis (L.) Osbeck] on Carrizo citrange rootstock (Citrus sinensis × Poncirus trifoliata Raf.). These were fertilized with 125 g N as labelled K15NO3 and grown outdoors in containers filled with a sand-loamy soil. Two groups of 3 trees received this N dose either in five equally split applications by a flooding irrigation system or in 66 applications by drip. Trees were harvested at the end of the vegetative cycle (December) and the isotopic ratios of 15N/14N were measured in the soil-plant system. The N uptake efficiency of the whole tree was higher with drip irrigation (75 percnt;) than with flooding system (64 percnt;). In the 0-90 cm soil profile, the N immobilized in the organic fraction was similar for both irrigation methods (around 13 percnt;), whereas the N retained as NO3 was 1 percnt; of the N applied under drip and 10 percnt; under flooding. In the last case, most of NO3 remained under root system and it could be lost to leaching either by heavy rainfalls or excessive water applications. These results showed that a drip irrigation system was more efficient for improving water use and N uptake from fertilizer, in addition to potentially reduced leaching losses.  相似文献
