We have evaluated the level of pp60c-src protein kinase activity in a variety of human tumor tissues and human tumor cell lines, and have estimated the abundance of the c-src protein in several of these tissues and cell lines. All cell lines derived from tumors of neuroectodermal origin that express a neural phenotype were found to possess c-src molecules with high levels of tyrosine-specific protein kinase activity. In contrast, cell lines derived from tumors of neuroectodermal origin that do not express neural characteristics, such as glioblastomas and melanomas, were found to have pp60c-src molecules with low levels of protein kinase activity. A similar pattern was observed when we analyzed the activity of c-src molecules extracted directly from corresponding tumor tissues. Analysis of human tumor cell lines derived from tissues other than those of neuroectodermal origin revealed that pp60c-src protein kinase activity was low in most cases. Exceptions to this observation were all rhabdomyosarcoma, osteogenic sarcoma, Ewing's sarcoma, and colon carcinoma lines tested. Comparison of pp60c-src kinase activity in normal skeletal muscle and rhabdomyosarcoma tissue and in normal breast tissue and breast adenocarcinoma tissue revealed that pp60c-src kinase activity was specifically elevated in the tumor tissues in both cases. However, the amount of pp60c-src protein in both normal and tumor tissues was found to be similar. These observations suggest that increases in the specific activity of the pp60c-src phosphotransferase in some rhabdomyosarcomas and breast carcinomas may be a characteristic acquired during the malignant transformation of the cells that is retained in cell lines established from these tumors.  相似文献

Cells from a strain of carrot which was prone to form deep-seated chlorophyll in its storage organ have been cultured in a manner that promoted them to organize into plantlets. Whereas the free cells contained only chloroplasts, the plantlets derived from these cells formed all types of plastids (“proplastids,” leucoplasts, chromoplasts, and chloroplasts) in accordance with the location of the cells in question in the developing plant body. The developmental history of the plastids has been traced with the electron microscope. The events of chloroplast development, previously described by Israel and Steward (1967) for cultured carrot explants, have been verified. The bearing of this new evidence upon the control of plastid development and biochemistry is discussed and related to other recent studies. The conclusion is that all totipotent carrot cells have plastids as essential organelles but that their final form and content are sharply defined by the factors inherent in the location of the cells in the plant body as it emerges.  相似文献

Glycolate synthesis was inhibited 40–50% in illuminated tobacco leaf disks, which have rapid rates of photorespiration, when floated on 20 mm potassium glycidate (2,3-epoxypropionate), an epoxide similar in structure to glycolate. The inhibitor also decreased the release of photorespiratory CO2 about 40%, and the specificity of glycidate was demonstrated by the 40–50% increase in rate of photosynthetic CO2 uptake observed in its presence. The importance of glycolate synthesis and metabolism in the production of photorespiratory CO2 and the role of glycolate in diminishing net photosynthesis in species with rapid rates of photorespiration was thus further confirmed. L-(or 2S)-Glycidate was slightly more active than DL-glycidate, but glycidate was more effective as a specific inhibitor in leaf tissue than several other epoxide analogs of glycolate examined. The products of photosynthetic 14O2 fixation after 3 or 4 min of uptake were proportionately altered in the presence of glycidate, and the specific radioactivity of the [14C]glycolate produced was closer to that of the 14CO2 supplied. Glycidate inhibited glycolate synthesis in tobacco leaf disks irreversibly, since the degree of inhibition was the same for at least 2 hr after the inhibitor solution was removed. Glycidate also blocked glycolate synthesis in maize leaf disks, tissue with low rates of photorespiration, but large increases in net photosynthesis were not observed in maize with glycidate, because glycolate synthesis is normally only about 10% as rapid in maize as in tobacco. The demonstration of increases in net photosynthesis of 40–50% when glycolate synthesis (and photorespiration) is blocked with glycidate indicates in an independent manner that the biochemical or genetic control of photorespiration should permit large increases in plant productivity in plant species possessing rapid rates of photorespiration.  相似文献

The effects of N-trifluoroacetyladriamycin-14-valerate on the uptake of [3H]thymidine and its incorporation into DNA of human P3HR-1 lymphoma cells were studied. In the absence of the drug, at 0 degrees C, [3H]thymidine was transported into the cells but not incorporated into DNA, as determined by both the trichloroacetic acid-soluble and -precipitable counts obtained with the cells. At 37 degrees C, [3H]thymidine was readily transported into the cells and incorporated into DNA. In the presence of the drug, both [3H]thymidine uptake (as shown by acid-soluble counts) and the amount of its incorporation into acid-precipitable materials were markedly reduced. However, the uptake of [3H]thymidine at 0 degrees C was found to be equally sensitive to drug inhibition as at 37 degrees C. The incorporation at 37 degrees C of [3H]thymidine into acid-precipitable materials of the cells, which had been prelabeled at 0 degrees C with [3H]thymidine, was found to be insensitive to inhibition by the drug. The in vitro activities of DNA polymerases alpha and beta purified from human P3HR-1 cells were also found not to be susceptible to inhibition. Nuclei purified from cells pretreated with the drug continued to synthesize DNA. The cytofluorograms of the cells treated with the drug indicated that the treated cells accumulated at the G2/M phase, whereas the S phase of the cells was not arrested. These results suggest that N-trifluoroacetyladriamycin-14-valerate inhibits [3H]thymidine uptake but not cellular DNA synthesis in human P3HR-1 lymphoma cells.  相似文献

The middle T antigen (MT Ag) encoded by polyoma virus has an associated protein kinase activity which transfers a phosphoryl group from ATP or GTP to a tyrosine residue on MT Ag in immunoprecipitates formed between polyoma virus-infected or transformed cell extracts and serum from animals bearing polyoma-induced tumors. Incubation of such immunoprecipitates or polyoma-transformed cell extracts prior to immunoprecipitation with the sulfhydryl reagent, N-ethylmaleimide (NEM), resulted in a significant inhibition of MT Ag-associated kinase activity. Inactivation of this enzyme activity by NEM was found to be dependent upon the incubation pH, time of incubation, and NEM concentration. ATP, GTP, and ADP in the presence of Mg2+ were found to decrease the rate of NEM-mediated inactivation of MT Ag-associated kinase activity, while CTP and UTP did not detectably alter the rate of enzyme inhibition by NEM. These results suggest that the MT Ag-associated kinase possesses at least one NEM-sensitive sulfhydryl group essential for phosphotransferase activity which may be present at or near the enzyme catalytic site.  相似文献

Today special emphasis is being placed upon the understanding of human aging and this study is an attempt to shed light on the craniofacial complex during the later years. Postcranial skeletal alteration is clear and it is now evident that cranial and facial structures are no exception to the aging process. The longitudinal information presented here indicates continuing overall growth from early adulthood to later life. The cranium thickens and the skull diameter increases. Endocranial dimensions enlarge as well. This suggests larger overall skull size and expansion of the cranial cavity. The visceral cranial structures also participate in the continuing growth process. Enlargement in all areas seems to be of similar magnitude except for skull thickness, sella turcica, and frontal sinus. The size increase in these three structures is greater than in other segments examined. In essence, the craniofacial complex is in a state of growth throughout life. The entire system is involved in a process of symmetrical enlargement.  相似文献

The two polyoma DNA fragments generated by cleavage with BamHI and EcoRI were cloned in pBR322, and their oncogenic potential was tested in vivo and in vitro. Only recombinant plasmid DNA containing a polyoma DNA fragment which extends clockwise from 58 to 0 map units and include approximately the 5'-proximal half of the early gene region produced tumors in newborn hamsters and transformed rat embryo cells in tissue culture. Southern blotting analysis indicated that the entire 2.2-kilobase polyoma BamHI-EcoRI fragment was intact in both a tumor cell line and a cell line transformed in culture which we examined. The presence of polyoma middle and small T antigen in these lines was demonstrated by immunoprecipitation and tryptic peptide mapping. DNA from a recombinant plasmid containing a polyoma genome deleted between 90 and 4 map units failed to induce tumors or transform cells.  相似文献

The use of the relative ureide content of xylem sap [(ureide-N/total N) × 100] as an indicator of N2 fixation in soybeans (Merr.) was examined under greenhouse conditions. Acetylene treatments to inhibit N2 fixation were imposed upon the root systems of plants totally dependent upon N2 fixation as their source of N and of plants dependent upon both N2 fixation and uptake of exogenous nitrate. Significant decreases in the total N concentration of xylem sap from plants of the former type were observed, but no significant decrease was observed in the total N concentration of sap from the latter type of plants. In both types of plants, acetylene treatment caused significant decreases in the relative ureide content of xylem sap. The results provided further support for a link between the presence of ureides in the xylem and the occurrence of N2 fixation in soybeans. The relative ureide content of xylem sap from plants totally dependent upon N2 fixation was shown to be insensitive to changes in the exudation rate and total N concentration of xylem sap brought about by diurnal changes in environmental factors. There was little evidence of soybean cultivars or nodulating strains affecting the relative ureide content of xylem sap. `Ransom' soybeans nodulated with Rhizobium japonicum strain USDA 110 were grown under conditions to obtain plants exhibiting a wide range of dependency upon N2 fixation. The relative ureide content of xylem sap was shown to indicate reliably the N2 fixation of these plants during vegetative growth using a 15N method to measure N2 fixation activity. The use of the relative ureide content of xylem sap for quantification of N2 fixation in soybeans should be evaluated further.  相似文献

To determine the stability of polyoma viral DNA in transformed rat cells during their growth in vivo, we compared the state and arrangement of polyoma virus DNA sequences in virus-transformed rat cell lines before and after their passage in vivo. In cell lines from 12 independent tumors induced by the inoculation of animals with three different transformed cell lines, we could detect no significant changes in the arrangement of viral DNA sequences associated with the in vivo passage of these cell lines. In 13 of 14 tumor cell lines examined, the pattern of polyoma virus tumor antigens, characterized by the presence of the polyoma virus large, middle, and small tumor antigens, was unchanged.  相似文献

Radiation is the primary therapeutic modality for children with medulloblastoma, a pediatric brain tumour. We examined the response of four medulloblastoma cell lines to ionising radiation. Our evaluation utilising flow cytometry, morphological analysis and terminal deoxynucleotidyl transferase assays demonstrated that medulloblastoma cells undergo radiation-induced apoptosis. p53 mediates radiation-induced apoptosis in many cell types, and p53 mutations have been associated with increased resistance to ionising radiation. p53 mutations are rare in medulloblastoma. We found that wildtype p53 is required for high levels of apoptosis in medulloblastoma, and cell lines in which p53 had been inactivated by mutation had very low levels of apoptosis. Inactivation of endogenous wildtype p53 in medulloblastoma cells by introduction of a dominant negative mutant of p53 decreased the level of radiation-induced apoptosis. Our results suggest that the sensitivity of medulloblastoma to irradiation involves p53-mediated apoptosis and that p53 gene status may be a predictor of response to radiation therapy.  相似文献
