Seedlings of barley were grown either in continuous darkness or under a diurnal 12 h light/12 h dark cycle and the effects on NADPH-protochlorophyllide oxidoreductase were followed at two different levels. Firstly, the relative content of the mRNA encoding the NADPH-protochlorophyllide oxidoreductase was measured by dot-blot hybridization. Secondly, changes in the enzyme polypeptide were monitored either by the method of immunoblotting or by immunogold labelling of ultrathin sections of Lowicryl-embedded leaf tissue. Our results demonstrate that drastic diurnal changes in the level of mRNA sequences and the enzyme protein are unlikely to occur in plants which have been grown under natural light/dark conditions. In the dark, protein and mRNA accumulation occurs at an early developmental stage. These results are difficult to reconcile with the suggestion that the massive accumulation of mRNA and enzyme protein in dark-grown seedlings is primarily the consequence of an artificially extended darkperiod. In addition to the plastid-specific NADPH-protochlorophyllide oxidoreductase a closely related polypeptide has been detected outside the plastid in the surrounding cytoplasm (Dehseh et al. 1986b, Planta 169, 172–183). During the diurnal light/dark treatment of seedlings the concentrations of the two protein populations did not show any variation indicative of an exchange between the two protein populations across the plastid envelope.Abbreviation poly(A)+RNA polyadenylated RNA  相似文献

Cooperative hydrogen bond interactions in the streptavidin-biotin system   总被引:1,自引:0,他引:1  
The thermodynamic and structural cooperativity between the Ser45- and D128-biotin hydrogen bonds was measured by calorimetric and X-ray crystallographic studies of the S45A/D128A double mutant of streptavidin. The double mutant exhibits a binding affinity approximately 2x10(7) times lower than that of wild-type streptavidin at 25 degrees C. The corresponding reduction in binding free energy (DeltaDeltaG) of 10.1 kcal/mol was nearly completely due to binding enthalpy losses at this temperature. The loss of binding affinity is 11-fold greater than that predicted by a linear combination of the single-mutant energetic perturbations (8.7 kcal/mol), indicating that these two mutations interact cooperatively. Crystallographic characterization of the double mutant and comparison with the two single mutant structures suggest that structural rearrangements at the S45 position, when the D128 carboxylate is removed, mask the true energetic contribution of the D128-biotin interaction. Taken together, the thermodynamic and structural analyses support the conclusion that the wild-type hydrogen bond between D128-OD and biotin-N2 is thermodynamically stronger than that between S45-OG and biotin-N1.  相似文献

Spatial variability of methane emissions from Swiss alpine fens   总被引:1,自引:0,他引:1  
Wetland ecosystems are a major natural source of the important greenhouse gas methane (CH4). Among these ecosystems, fens have been shown to release high quantities of CH4. Data on CH4 emissions from alpine fens are scarce and mainly limited to the United States and China. Therefore, static chambers were used to quantify CH4 emissions from 14 fens located in the Swiss Alps. The aims of this study were to determine the spatial variability of the emissions and to identify potential key factors which influence CH4 turnover. The fens were located at altitudes between 1,800 and 2,600 m a.s.l., the pore water varied from acidic to slightly acidic (pH 4.5–6.4) and the vegetation was dominated by plants of the genus Carex. In addition, the underlying bedrock was either siliceous or calcareous. Methane emissions ranged from 74 ± 43 to 711 ± 212 mg CH4 m?2 day?1. The type of bedrock, the plant biomass above the water table and the CH4 pore water concentrations at depths from 0 to 20 cm were the main factors influencing CH4 emissions. Detailed measurements in three selected fens suggested that more than 98 % of the total CH4 emissions are due to plant-mediated transport.  相似文献

The morphology, altitudinal distribution and phytocoenology ofHypericum empetrifolium Willd., var.oliganthum Rech. fil. and var.tortuosum Rech. fil. are reassessed and as a result, the taxa are treated at subspecific rank. Summary Regulation of the paracellular pathway in rabbit distal colon by the hormone aldosterone was investigated in vitro in Ussing chambers by means of transepithelial and microelectrode techniques. To evaluate the cellular and paracellular resistances an equivalent circuit analysis was used. For the analysis the apical membrane resistance was altered using the antibiotic nystatin. Under control conditions two groups of epithelia were found, each clearly dependent on the light: dark regime. Low-transporting epithelia (LT) were observed in the morning and high-transporting epithelia (HT) in the afternoon. Na+ transport was about 3-fold higher in HT than in LT epithelia. Incubating epithelia of both groups with 0.1 mol·1-1 aldosterone on the serosal side nearly doubled in LT epithelia the short circuit current and transepithelial voltage but the transepithelial resistance was not influenced. Maximal values were reached after 4–5 h of aldosterone treatment. In HT epithelia due to the effect of aldosterone all three transepithelial parameters remained constant over time. Evaluation of the paracellular resistance revealed a significant increase after aldosterone stimulation in both epithelial groups. This increase suggests that tight junctions might have been regulated by aldosterone. The hormonal effect on electrolyte transport was also dependent on the physiological state of the rabbit colon. Monoclonal antibodies against partially purified adenylyl cyclase from bovine brain cortex were raised in mice. Three types of antibody were obtained. Type 1 was specific for the calmodulin-sensitive enzyme. Type II also recognized this enzyme, but recognized the calmodulin-insensitive enzymes from a variety of species and tissues as well. Type I antibodies precipitated their antigens in both the native and denatured forms, while type II strongly favored the denatured forms. Type III antibodies precipitated adenylyl cyclase activity, but as shown by Western blot analysis, were directed against 38-kDa and 45-kDa glycoproteins. The 38-kDa protein was identified as synaptophysin.  相似文献

The three species of banded newt (genus Ommatotriton) are endemic to the Near East. Recently an introduced banded newt population was discovered in Catalonia, Spain. To determine the species involved and the geographical source, we genotyped 11 individuals for one mitochondrial and two nuclear genetic markers, and compared the observed haplotypes to a range-wide phylogeography of Ommatotriton. All haplotypes identified in Spain are identical to haplotypes known from the native range. The mitochondrial haplotypes derive from O. ophryticus and were originally recorded in northeast Turkey. The nuclear haplotypes reveal that all individuals are genetically admixed between O. ophryticus and O. nesterovi. While the geographical resolution for the nuclear markers is low, the source of the O. nesterovi ancestry must be Turkey, as this species is a Turkish endemic. Species distribution models suggest a large potential distribution for the two Ommatotriton species, extending over northern Iberia and southern France. The ecology of hybrids can differ substantially from that of the parent species, making the impact of the Spanish hybrid banded newt population difficult to predict.  相似文献

Abstract. Phenological development of the colonizer Bunias orientalis L. was investigated in permanent plots in herbaceous vegetation, using photography and image analysis. In all habitats but one, B. orientalis showed a two-phased rosette growth behaviour, with a first peak cover reached in spring, roughly coinciding with an early peak cover of the matrix vegetation, and a second autumn growth flush occurring in a phase of reduced matrix vegetation cover. This seasonally bimodal growth pattern of B. orientalis appears to partly compensate for its competitive inferiority in crowded herbaceous vegetation. Small or overshadowed individuals of this species particularly profit from higher above-ground performance or release from overlap in autumn. The significance of temporal niche separation for survival and growth of B. orientalis individuals varies with habitat conditions, being most apparent in occasionally disturbed habitats with a relatively low frequency of gap formation. Despite some limitations, image analysis proved to be useful for phenological investigations within herbaceous plant stands.  相似文献

Prolonged treatment of human platelets with the prostacyclin analog iloprost led to desensitization of the response to various prostaglandin derivatives. However, basal adenylyl cyclase activity and stimulation by agents acting directly via Gs, the stimulatory guanine-nucleotide-binding regulatory protein of adenylyl cyclase, were likewise decreased. Reconstitution of desensitized membranes with purified Gs from turkey erythrocytes indicated no alteration in the catalyst itself. However, the function of Gs (in cholate extracts) appeared to be severely impaired when reconstituted with adenylyl cyclase catalyst. Modification of Gs was also indicated by its altered sedimentation in sucrose density gradients. From Western blots, the alpha subunit of Gs, alpha s, from control platelets sedimented as a 5.6S species, while that from desensitized cells appeared at higher S values (in a polydisperse distribution). Activation by guanosine 5'-[gamma-thio]triphosphate of Gs from control platelets shifted alpha s to 3.5-3.7S, while activation of Gs from desensitized platelets induced such shift only for a minor portion of alpha s. This small fraction alone appeared to be susceptible to ADP-ribosylation by cholera toxin/[32P]NAD. Furthermore, an antibody directed against the C-terminal hexadecapeptide of alpha s precipitated much less alpha s from cholate extracts derived from desensitized platelets. Modification of alpha s during desensitization was also suggested from cross-linking experiments using the homobifunctional agent bismaleimidohexane: alpha s from desensitized platelets formed a single product of 80 kDa, while that from untreated platelets yielded a doublet (100 kDa and 110 kDa).  相似文献
