The possible role of antibody as a selective pressure on antigenic mutants of poliovirus in nature was investigated in vitro. A mutant resistant to a monospecific antibody with a definite specificity was readily obtained by several cycles of neutralization of Mahoney strain with a monospecific antibody and multiplication in monkey kidney (MS) cells. Mutants resistant to more than two different monospecific antibodies were also readily obtained in a similar manner. Studies on the antigenicity of these mutants by kinetic neutralization tests revealed that the Mahoney strain underwent a progressive serological variation as it became successively resistant to one to five different monospecific antibodies isolated from anti-Mahoney serum. 相似文献

Gene conversion and natural selection in the evolution of X-linked color vision genes in higher primates 总被引:2,自引:1,他引:1
During higher primate evolution, gene conversion seems to have occurred
often between the red and green photo-pigment genes, which are tandemly
linked on the X chromosome. To understand this phenomenon better, intron 4
sequences of the red and green pigment genes of a male human (an Asian
Indian), a male chimpanzee, and a male baboon were amplified by PCR and
sequenced. The data show that the intron 4 sequences between the two genes
have been strongly or completely homogenized in the three species studied.
Apparently recent gene conversion events have occurred in introns 4 of the
red and green pigment genes in humans and chimpanzees. Two or more
conversion events may have occurred at different times in introns 4 of the
two pigment genes in baboons. The divergence between the two genes is
significantly lower in intron 4 than in exons 4 and 5 in each species,
contrary to the usual situation that introns evolve faster than exons. It
is most likely that strong natural selection for maintaining the distinct
functions of exons 4 and 5 of the red and green pigment genes has acted
against sequence homogenization of these exons.

Thrombin has been shown to increase expression of chemokines such as monocyte chemoattractant protein 1 (MCP-1) in endothelial cells, leading to the development of atherosclerosis. However, the precise mechanism of this induction remains unknown. In the present study, we investigated whether the small G protein RhoA, and its effector, Rho-kinase are involved in MCP-1 induction by thrombin in endothelial cells. Y-27632, a specific Rho-kinase inhibitor, potently inhibited MCP-1 induction by thrombin. Y-27632 significantly decreased the chemotactic activity of thrombin-stimulated supernatants of endothelial cells on monocytes. Importantly, fasudil, a specific Rho-kinase inhibitor, attenuated MCP-1 gene expression in the aorta of db/db mice. Y-27632 attenuated thrombin-mediated phosphorylation of p38MAPK and p65, indicating that Rho-kinase mediates thrombin-induced MCP-1 expression through p38MAPK and NF-κB activation. Our findings demonstrate that the Rho/Rho-kinase signaling pathway plays a critical role in thrombin-mediated MCP-1 expression and function, and suggest that Rho/Rho-kinase may be an important target in the development of new therapeutic strategies for atherosclerosis. 相似文献

Iron is an essential trace metal for most organisms. However, excess iron causes oxidative stress through production of highly toxic hydroxyl radicals via the Fenton/Haber-Weiss reaction. Iron storage in the body is reported to be associated with fat accumulation and type 2 diabetes mellitus. We investigated the role of iron in adiposity by using KKAy mice and obese and diabetic model mice. Eight-week-old KKAy mice were divided into two groups and treated with deferoxamine (DFO), an iron chelator agent, or a vehicle for 2 wk. DFO treatment diminished fat iron concentration and serum ferritin levels in KKAy mice. Fat weight and adipocyte size were reduced significantly in DFO-treated mice compared with vehicle-treated mice. Macrophage infiltration into fat was also decreased in DFO-treated mice compared with vehicle-treated mice. Superoxide production and NADPH oxidase activity in fat, as well as urinary 8-hydroxy-2'-deoxyguanosine excretion, were decreased in KKAy mice after DFO treatment while p22(phox) expression in adipose tissue was diminished in such mice. Ferritin expression in the fat of DFO-treated KKAy mice was decreased. In addition, F4/80-positive cells also presented through both p22(phox) and ferritin expression. The mRNA expression levels of inflammatory cytokines were also reduced in fat tissue of DFO-treated mice. These findings suggest that reduction of iron levels ameliorates adipocyte hypertrophy via suppression of oxidative stress, inflammatory cytokines, and macrophage infiltration, thereby breaking a vicious cycle in obesity. 相似文献
