The A mating factor of Coprinus cinereus determines compatibility in mating by regulating part of a developmental sequence that leads to dikaryon formation. The A genes that trigger development encode two different classes of homeodomain proteins, and for a successful mating, a protein of one class, HD 1, must interact with a protein of the other class, HD 2. In this report we show that C. cinereus A genes that encode HD 2 proteins, a2-1 and b2-1, can elicit A-regulated development in the heterologous host C. bilanatus. Transformation rates were very low, suggesting that the genes were poorly transcribed. The fact that the HD 2 genes are functionally expressed implies successful heteromultimeric association of putative DNA-binding proteins coded by the two Coprinus species. This interaction was sufficient to satisfy the need for different A factors in the formation of a fertile C. bilanatus dikaryon, but fertile dikaryons were more readily produced in matings with the a2-1 gene transformants. The C. cinereus A genes, b1-1 and d1-1, which encode HD1 proteins, were either not expressed or their proteins were non-functional in C. bilanatus. Anuran males emit advertisement calls for the purpose of attracting females and repelling conspecific males. Call transmission is influenced by the acoustics of the propagation environment, including vegetation. Thus, forestry monocultures of non-native trees represent artificial environments that could modify call transmission. These monocultures have substituted large areas of Atlantic Forest in southern Brazil, representing a conservation challenge. Considering this context, we hypothesized that anurans have calls that are less attenuated in their native environment than in forest plantations. To test it, we performed sound transmission experiments using calls of three anuran species native to southern Brazil: Boana bischoffi, B. leptolineata, and Hylodes meridionalis. We compared sound attenuation between the native forest and forestry monocultures (Eucalyptus sp. and Pinus sp. forests), and included distance from the sound source, air temperature, humidity, and vegetation density as cofactors in linear mixed models. Our results show that the advertisement calls of the two tree frogs (Boana) attenuate less in tree plantations than in native forests, while the third species shows either no differences or less attenuation in native forests. Our results can be understood according to differences in natural history and microhabitat use among the studied species. The two tree frog species vocalize perched from vegetation so their calls become better transmitted in forestry monocultures, which have fewer trees than the native forest. On the other hand, H. meridionalis calls from rocks on streams and the propagation is finely tuned to this peculiar microhabitat. Isolates of Phytophthora infestans were collected from all potato growing regions of Poland during the blight seasons of 1987—1989. All 1987 isolates were of Al mating type and were sensitive to metalaxyl. In 1988 and 1989, 46.5 % and 55.3 % of the isolates were sensitive to metalaxyl, respectively. The percentage of highly resistant (R) isolates increased from 25.6 % in 1988 to 39.5 % in 1989; however the percentage of intermediately resistant (I) isolates decreased during that period from 27.9 % to 5.3 %. A significant association was observed between the A1 compatibility type and metalaxyl resistance. The A2 mating type first appeared in 1988, and its frequency increased from 4.7 % of the population in 1988 to 47.6 % in 1989. Coincident with this change in mating type frequency, changes in ploidy levels of isolates were observed. Whereas 3 % of the 1988 isolates were diploid, 90 % of the 1989 A2 isolates and 28.6 % of the 1989 Al isolates were diploid. The approximate 1:1 ratio of the two mating types encountered in 1989, and the predominance of diploidy, indicates that the Polish population of P. infestans has the potential to become sexual. 相似文献

Experiments were performed with the mycorrhizal fungus Suillus granulatus to define the parameters for production and regeneration of protoplasts. Protoplasts were released at frequencies between 1 and 3×107/ml from mycelium 3 to 7 days old. The best osmotic stabilizer for protoplast release was MgSO4 (0.7 m). To optimize protoplast release and regeneration an enzyme (Novozym 234) concentration 1.7 mg/ml was chosen, with a digestion time of 1 to 2 h. The activation of subcomponents C1r and C1s in the first component of complement, C1, when bound to antibody-antigen complexes was investigated. Activation was followed both by the splitting of the peptide chains of subcomponents C1r and C1s and by the development of proteolytic activity. For the maximum rate of activation to occur, all components must be present in approximate molar proportions of antibody: C1q:C1r:C1s of 13:1:5:5. For activation of subcomponent C1s, subcomponents C1r or C1r, but not C1r inactivated with iPr2P-F (di-isopropyl phosphorofluorideate), are effective. For activation of subcomponent C1r, subcomponents C1s, C1s or C1s inactivated with iPr2P-F are effective. Subcomponent C1s is activated by C1r, and C1r is activated autocatalytically, probably through the formation of an intermediary C1r. in which the peptide chain is unsplit but a conformational change caused by interaction with the other components has led to the formation of a catalytic site able to split subcomponent C1r to C1r. 相似文献

By using the techniques of ligation of the larvae (brain and endocrine glands extirpation) and salivary gland implantation, the hormonal dependence of the activity of certain puffs of Rhynchosciara was investigated. Our results have shown that the puffing behaviour — activation and deactivation — varies according to the developmental stage in which the larvae were ligated. When the larvae were ligated just before the drastic changes in the puffing pattern, which occur prior to pupation, these changes fail to occur. When the larvae were ligated after the onset of these changes we have observed: a) some of the puffs active at the time of the ligature regress promptly, earlier than their normal timing observed in controls; b) others remain active indefinitely and c) there are still some which regress accordingly to the normal timing.The puff B2 which behaves as those in b was double checked by means of implantation experiments. Salivary glands which had puff B2 at its maximum expansion were implanted into younger larvae and that puff also remained active in the body cavity of these larvae. Caffeine causes severe retardation of growth and development and high mortality of larvae Telmatoscopus albipunctatus. In general, sensitivity to caffeine is inversely proportional to the age of the treated larvae. Adults originating from treated larvae show lower fertility. Treatment of pupae results in elongated abdomens, abnormal wings and reduced fertility in adults. Caffeine also affects adversely polytenization of chromosomes. It is suggested that effects on larval growth and polytenization of chromosomes are due to retardation of DNA synthesis. Other possible mechanisms of action of caffeine are discussed.
Zusammenfassung Koffein verursacht starke Verzögerung von Wachstum und Entwicklung sowie hohe Mortalität der Larven von Telmatoscopus albipunctatus. Im allgemeinen ist die Empfindlichkeit genenüber Koffein umgekehrt proportional zum Alter der behandelten Larven. Adulte, die als Larven damit behandelt wurden, zeigen verminderte Fertilität. Behandlung von Puppen ergibt Adulte mit verlängertem Abdomen, anormalen Flügeln und verminderter Fertilität. Koffein beeinflußt auch die Polytänisierung der Chromosomen ungünstig. Es wird angenommen, daß die Beeinflussung des Larvenwachstums und der Polytänisierung der Chromosomen zurückzuführen ist auf verzögerte DNS-Synthese. Adriamycin and daunorubicin bound to covalently closed circular DNA nick the latter when reduced by sodium borohydride as demonstrated using an ethidium bromide fluorescence assay. The degradation, dependent on oxygen, is strongly inhibited by (i) superoxide dismutase (ii) catalase and (iii) sodium benzoate indicating the intermediacy in the cleavage of superoxide radical anion, hydrogen peroxide and hydroxyl radicals respectively. Less nicking of the DNA is observed by the reduced aglycones, so binding to the DNA by the aminosugar moiety assists the cleavage process. Adriamycin, daunorubicin and both ring C reduced forms bind intercalatively and completely relax supercoiled DNA. The results provide a possible rationale for the degradation of DNA which accompanies anthracycline administration. 相似文献

Summary The applicability of the Electro-Ultra-Filtration (EUF) method in soil analyses was studied. The reproducibilities of the
amounts of soil extracts, of ion concentrations in the extracts and of the distribution of cations and anions over the cathode
and anode extracts by use of fully automatic EUF equipment were tested.
The degree of variability among replicates was expressed as coefficient of variation (CV) and as the highest percentual divergence
of an individual analytical measurement from the mean (L).
The extraction volumes of five replicates of six different soils were found to vary between 1.1–7.1% with an average of 3.8%,
as CV and between 1.5–11.3% as L. The reproducibility of desorbed P in the anode extract varied between 2.7–31.7% with an
average of 8.7%, as CV and between 3.2–37.9% as L. Corresponding values for CV and L of K desorbed varied between 1.3–13.9%
and 1.6–23.8%, respectively.
Variations among replicates of desorbed P were especially high in the first 1–2 sub-fractions of a total of seven fractions
in a single extraction run. Low K concentrations in the extract had a slightly negative influence on the reproducibility of
K desorption.
Furthermore, it was found that a portion of the cations is collected in the anode extract and a portion of the anions in the
cathode extract, especially at the beginning of an extraction run. Pooling of anode and cathode extracts before analysis is

Wood samples, infested by fungi during storage, were shown to contain, besides the known 5-methyl-mellein, additional (3R)-8-hydroxy-3-methyl-3,4-dihydroisocoumarins substituted by 7-methyl, 5-formyl, 5-carboxy, 5-hydroxy, 5-methoxy, 6-methoxy-5-methyl and 6,7-dimethoxy-5-methyl groups, as well as 6-formyl-7-hydroxy-5-methoxy-4-methylphthalide. Several 2-methylchromanones were synthesized in order to show that this class of compounds can be distinguished from 3-methyl-3,4-dihydroisocoumarins by MS. 相似文献
