Background
Venous thromboembolism (VTE) is a common disease where known genetic risk factors explain only a small portion of the genetic variance. Then, the analysis of intermediate phenotypes, such as thrombin generation assay, can be used to identify novel genetic risk factors that contribute to VTE.Objectives
To investigate the genetic basis of distinct quantitative phenotypes of thrombin generation and its relationship to the risk of VTE.Patients/Methods
Lag time, thrombin peak and endogenous thrombin potential (ETP) were measured in the families of the Genetic Analysis of Idiopathic Thrombophilia 2 (GAIT-2) Project. This sample consisted of 935 individuals in 35 extended families selected through a proband with idiopathic thrombophilia. We performed also genome wide association studies (GWAS) with thrombin generation phenotypes.Results
The results showed that 67% of the variation in the risk of VTE is attributable to genetic factors. The heritabilities of lag time, thrombin peak and ETP were 49%, 54% and 52%, respectively. More importantly, we demonstrated also the existence of positive genetic correlations between thrombin peak or ETP and the risk of VTE. Moreover, the major genetic determinant of thrombin generation was the F2 gene. However, other suggestive signals were observed.Conclusions
The thrombin generation phenotypes are strongly genetically determined. The thrombin peak and ETP are significantly genetically correlated with the risk of VTE. In addition, F2 was identified as a major determinant of thrombin generation. We reported suggestive signals that might increase our knowledge to explain the variability of this important phenotype. The dewlap is an extendible flap of skin ordinarily folded under the throat. Lizards, particularly those in the genus Anolis, extend their dewlaps during interactions with conspecifics, other lizards, and potential predators. Dewlap extension is effected by movements of elements of the hyoid apparatus. This paper describes the anatomy of the hyoid and associated musculature in Anolis equestris, a large arboreal lizard with a prominent dewlap. A mechanism for dewlap extension is proposed based on results of morphological and experimental techniques. Specializations of the hyoid skeleton for dewlap extension include elongated second ceratobranchials and highly movable joints between the ceratohyals and the hypohyals and between the first ceratobranchials and the body of the hyoid. A well developed M. ceratohyoideus extends between the ceratohyals and the first ceratobranchials of the hyoid apparatus. During dewlap extension, the hyoid apparatus acts as a first order lever. Contraction of M. ceratohyoideus pulls the ceratohyals posteriorly causing the hypohyals and the body of the hyoid to rotate dorsally around the first ceratobranchial/body joints. This movement results in the second ceratobranchials swinging forward and down, unfolding the dewlap. The relative immobility of the first ceratobranchials provides stability to the hyoid apparatus during dewlap extension. A comparison is made of dewlap extension and other hyoid displays. 相似文献

Dual-specificity tyrosine phosphorylation-related kinase 1A (DYRK1A) is a dual-specificity protein kinase that catalyses phosphorylation and autophosphorylation. Higher DYRK1A expression correlates with cancer, in particular glioblastoma present within the brain. We report here the synthesis and biological evaluation of new heterocyclic diphenolic derivatives designed as novel DYRK1A inhibitors. The generation of these heterocycles such as benzimidazole, imidazole, naphthyridine, pyrazole-pyridines, bipyridine, and triazolopyrazines was made based on the structural modification of the lead DANDY and tested for their ability to inhibit DYRK1A. None of these derivatives showed significant DYRK1A inhibition but provide valuable knowledge around the importance of the 7-azaindole moiety. Endozoochory is usually involved in seed dispersal mutualisms, whereas ectozoochory is non‐rewarding, and therefore neutral (or even negative) for the animal vector. Synzoochory is an intermediate dispersal type between endo and ectozoochory in which propagules are deliberately transported (usually in the mouth) but with no ingestion or gut passage involved. We present empirical evidence of synzoochoric mutualism between the hummingbird Sephanoides sephaniodes and cryptogams (one fern and seven moss species). Two species (Lophosoria quadripinnata and Ancistrodes genuflexa) constituted the bulk of nest biomass, and another six moss species were present in lesser quantity. The hummingbird was selective when collecting nest material so that the nests contained a higher density of reproductive structures (that could be dispersed further) than natural patches of the cryptogam species. Even after one year, the nests maintained half of the original reproductive structures (sporangia, sporophytes) and biomass, constituting an important dispersal source. These results show a new type of mutualism in which mosses could be dispersed throughout longer distances (several km) by hummingbirds and to higher positions (particularly for ground‐living species, promoting dispersal potential). The hummingbird benefits from collecting cryptogam material for nest building, and cryptogams benefit from the concentration and relocation of diaspore sources into more effective recruiting sites. Questions: Does fuzzy clustering provide an appropriate numerical framework to manage vegetation classifications? What is the best fuzzy clustering method to achieve this? Material: We used 531 relevés from Catalonia (Spain), belonging to two syntaxonomic alliances of mesophytic and xerophytic montane pastures, and originally classified by experts into nine and 13 associations, respectively. Methods: We compared the performance of fuzzy C‐means (FCM), noise clustering (NC) and possibilistic C‐means (PCM) on four different management tasks: (1) assigning new relevé data to existing types; (2) updating types incorporating new data; (3) defining new types with unclassified relevés; and (4) reviewing traditional vegetation classifications. Results: As fuzzy classifiers, FCM fails to indicate when a given relevé does not belong to any of the existing types; NC might leave too many relevés unclassified; and PCM membership values cannot be compared. As unsupervised clustering methods, FCM is more sensitive than NC to transitional relevés and therefore produces fuzzier classifications. PCM looks for dense regions in the space of species composition, but these are scarce when vegetation data contain many transitional relevés. Conclusions: All three models have advantages and disadvantages, although the NC model may be a good compromise between the restricted FCM model and the robust but impractical PCM model. In our opinion, fuzzy clustering might provide a suitable framework to manage vegetation classifications using a consistent operational definition of vegetation type. Aims: To evaluate the efficiency of the vitamin B12- producing Lactobacillus reuteri CRL1098 strain in preventing the symptoms caused by a nutritional cobalamin-deficient diet in pregnant female mice and their weaned offspring.
Methods and Results: Pregnant female mice were divided into three groups: animals fed with a B12 -deficient diet (DD), animals fed with DD plus L. reuteri CRL1098 and animals fed with a B12 -sufficient diet. The animals received the different feedings from the end of gestation up to weaning. At the end of the trials, they and their corresponding offspring were bled to determine haematological, immunological and histological parameters. The administration of the pseudovitamin B12 -producing strain prevented the symptoms observed in female and weaned young animals fed with a nutritional B12 -deficient diet.
Conclusions: Our data suggest that the pseudovitamin B12 produced by L. reuteri CRL1098 is biologically active and effective in preventing the pathologies caused by the nutritional deficiency of B12 both in pregnant mice and their offspring.
Significance and Impact of the Study: The ability of L. reuteri CRL1098 to prevent a nutritional vitamin deficiency was demonstrated for the first time. The addition of a GRAS micro-organism to complement the B12 content in deficient foods is an interesting biotechnological alternative. 相似文献
