Chick embryo fibroblasts constitute a useful model for investigating cell surface differentiation using Ricinus lectin as a marker. Fibroblasts from 8-day chick embryos had two classes of Ricinus lectin binding sites, whereas those from 16-day embryos displayed only one class. Hyaluronidase treatment of fibroblasts from 8-day embryos had no effect on their capacity to bind Ricinus lectin; however after this treatment, 16-day cells resembled 8-day cells since the former also exhibited two classes of lectin-binding sites. Treatment with hyaluronidase released 2-5 times more hyaluronic acid from the older cells than from the younger cells. The same hyaluronidase treatment did not change the number of 8-day cells detached by trypsin from the substrate, but increased the number of detached 16-day cells. These observations suggest (i) that the greater adhesiveness to the substrate of the 16-day cells might be due to the presence on the cell surface of a larger amount of glycosaminoglycans at 16 days than at 8 days, and (ii) that the increased accumulation of hyaluronic acid on the cell surface might be involved in an alteration in the cell membrane during differentiation.  相似文献

Recombinant human procollagen C-proteinase enhancer (rPCPE) was expressed using a baculovirus system and purified to homogeneity using a three-step procedure including heparin affinity chromatography. Heparin binding was dependent on the C-terminal netrin-like domain. The recombinant protein was found to be active, increasing the activity of procollagen C-proteinase/bone morphogenetic protein-1 on type I procollagen in a manner comparable to the native protein. Enhancing activity was dependent on intact disulfide bonding within the protein. By circular dichroism, the observed secondary structure of rPCPE was consistent with the known three-dimensional structures of proteins containing homologous domains.  相似文献

The influence of rehydration conditions on the recovery of 16 species of freeze-dried lactic acid bacteria was investigated. The survival of dried cultures during reconstitution to the wet state was increased by rehydration with small volumes of the medium used for that purpose (0.3 and 0.5 ml). Comparison of rehydration at several times of exposure showed best survival at 10 min for the majority of the species analyzed.  相似文献

Ricinus communis lectin RCA I-binding sites on the surface of cultured human liver cells have been studied with a view to establishing a relationship between the aging of the cells in vitro and the age of the donors. Fetal and new-born liver cells exhibited two classes of lectin-binding sites in the early in vitro passages, whereas adult cells in the early passages and fetal and new-born cells in the late passages had only one class of binding site. The modifications of the cell surface RCA I-binding sites would appear to occur either following masking of one of the classes of lectin-binding site during aging and/or be caused by a post-translational event rather than by glycoprotein mis-synthesis. A striking relationship was found between the in vitro cellular aging and human age in vivo, which would suggest that fetal human liver cells could prove to be a convenient model for studying the changes occurring during aging in vivo.  相似文献

An automatic flow injection analysis (FIA) system for on-line determination of esterase activity has been developed. It is based on a colorimetric method using p-nitrophenyl propionate as substrate. The system permits a linear range analysis up to 0.18 U ml–1, although the range can be extended up to 1 U ml–1 without external dilution of the sample. The sampling frequency is of 4 samples per h with a relative standard deviation of 0.9%.  相似文献

AIMS: The objective of this work was to evaluate the fermentation pattern of and the exopolysaccharide (EPS) production by Lactobacillus helveticus ATCC 15807 in milk batch cultures under controlled pH (4.5, 5.0 and 6.2). METHODS AND RESULTS: EPS concentration was estimated by the phenol/sulphuric acid method and the chemical composition of purified EPS by HPLC. Fermentation products and residual sugars were determined by HPLC and enzymatic methods. The micro-organism shifted from a homofermentative to a heterofermentative pattern, producing acetate (9.5 and 5.8 mmol l(-1)) at pH 5.0 and 6.2, respectively, and acetate (7.1 mmol l(-1)) plus succinate (1.2 mmol l(-1)) at pH 4.5. At pH 5.0 and 6.2, acetate derived from citrate while at pH 4.5 it came from both citrate and pyruvate splitting. The EPS has a MW of 10(5)-10(6) and contains phosphate (81% in average), rhamnose (traces), and glucose and galactose in a ratio of 1 : 1 (pH 6.2) and 2 : 1 (pH 4.5 and 5.0). The highest production (549 mg l(-1)) corresponded to pH 5.0 and the lowest (49 mg l(-1)) to pH 6.2. CONCLUSIONS: The heterofermentative pattern of Lact. helveticus ATCC 15807 was linked to alternative pyruvate pathways and/or citrate metabolism according to the environmental pH. The EPS production was improved under low environmental pH conditions. SIGNIFICANCE AND IMPACT OF THE STUDY: This work provides relevant information of the effect of pH on the metabolism of citrate and EPS production by Lact. helveticus. It may contribute to improve technological aspects of ropy and citrate-utilizing lactic acid bacteria.  相似文献
