Although the AdhN/AdhN strain ofPeromyscus maniculatus (so-called ADH? deermouse) has been previously considered to be deficient in ADH, we found ADH isozymes of Classes II and III but not Class I in the liver of this strain. On the other hand, the AdhF/AdhF strain (so-called ADH+ deermouse), which has liver ADH activity, had Class I and III but not Class II ADH in the liver. In the stomach, Class III and IV ADHs were detected in both deermouse strains, as well as in the ddY mouse, which has the normal mammalian ADH system with four classes of ADH. These ADH isozymes were identified as electrophoretic phenotypes on the basis of their substrate specificity, pyrazole sensitivity, and immunoreactivity. Liver ADH activity of the ADH? strain was barely detectable in a conventional ADH assay using 15 mM ethanol as substrate; however, it increased markedly with high concentrations of ethanol (up to 3M) or hexenol (7 mM). Furthermore, in a hydrophobic reaction medium containing 1.0M t-butanol, liver ADH activity of this strain at low concentrations of ethanol (<100 mM) greatly increased (about sevenfold), to more than 50% that of ADH+ deermouse. These results were attributable to the presence of Class III ADH and the absence of Class I ADH in the liver of ADH? deermouse. It was also found that even the ADH+ strain has low liver ADH activity (<40% that of the ddY mouse) with 15 mM ethanol as substrate, probably due to low activity in Class I ADH. Consequently, liver ADH activity of this strain was lower than its stomach ADH activity, in contrast with the ddY mouse, whose ADH activity was much higher in the liver than in the stomach, as well as other mammals. Thus, the ADH systems in both ADH? and ADH+ deermouse were different not only from each other but also from that in the ddY mouse; the ADH? strain was deficient in only Class I ADH, and the ADH+ strain was deficient in Class II ADH and down-regulated in Class I ADH activity. Therefore, Class III ADH, which was found in both strains and activated allosterically, may participate in alcohol metabolism in deermouse, especially in the ADH? strain.  相似文献

Thermolysin (Bacillus thermoproteolyticus neutral proteinase, EC 3.4.24.4) has been immobilized by radiation polymerization of hydrophilic and hydrophobic monomers, and its properties, such as enzyme activity, thermal stability and durability, have been studied. The activity of the immobilized enzymes increased with an increase in the hydrophilicity of the polymer matrix and with a decrease in monomer concentration. Immobilization with hydrophilic monomers increased the thermal stability of the enzymes, but the thermal stability of the enzymes immobilized with hydrophobic monomers was comparable with that of native enzymes. The durability of the immobilized enzymes was examined by continuous hydrolysis of casein; enzymes immobilized with a high concentration (90%) of hydrophilic monomers appeared to be stabilized and could be used for long times.  相似文献

The three protein kinases of Lemna paucicostata that are separableby DEAE-Sephacel chromatography have been designated PI, PIIand PIII [Kato et al. (1983) Plant & Cell Physiol. 24: 841].The optimum pH for the PI and PII enzymes was 7.5 and for thePHI enzyme 7.0. The activities of these enzymes were stimulatedby divalent cations, the maximum stimulation being producedby 5 nw Mg2 $ for PI, by 3 mM Co2 $ for PII and by 1 mM Mn2$ for PIII. The cytokinins; benzyladenine, kinetin and zeatin,inhibited the activity of the PIII enzyme. The molecular weightsof the PI and PII enzymes did not change after incubation withcAMP even though their activities were regulated by this compound. (Received October 17, 1983; )  相似文献

Abstract Passive transfer between rates of protection against cholera toxin (CT) was studied. Extracts of various organs, obtained from CT-immunized rats, were injected intravenously into non-immunized recipient rats. The ability of the extracts to inhibit CT-induced secretion in ligated jejunal loop were tested. A significant inhibition of the response to CT was achieved by extracts from hypophysis, brain and jejunal mucosa. Extracts from pancreas, spleen or adrenal glands were without effect, as were all extracts obtained from control rats. The antisecretory effects of the hypophysis extracts became intensified with increasing numbers of immunizations, and the antisecretory effect was most pronounced when the extract was injected immediately before the CT challenge. The active component of the hypophysis extract was heat-labile and negatively charged, suggesting an acidic protein as the mediator of the protective effect against CT.  相似文献

The growth rates of young chicks were varied from 0 to 10% per day by manipulation of the adequacy of the amino acid and energy supply. The rates of protein synthesis in the white breast (pectoralis thoracica) muscle and the dark leg (gastrocnemius and peronaeus longus) muscles were estimated by feeding l-[U-14C]tyrosine in amino acid/agar-gel diets (`dietary infusion'). This treatment rapidly and consistently produced an isotopic equilibrium in the expired CO2 and in the free tyrosine of plasma and the muscles. Wholebody protein synthesis in 2-week-old chicks was estimated from the tyrosine flux and was 6.4g/day per 100g body wt. In 1-week-old chicks the rate of protein synthesis was more rapid in the breast muscles than in the leg muscles, but decreased until the rates were similar in 2-week-old birds. Synthesis was also more rapid in fast-growing Rock Cornish broilers than in medium-slow-growing New Hampshire×Single Comb White Leghorn chicks. No or barely significant decrease in the high rates of protein synthesis, in the protein/RNA ratio and in the activity of RNA for protein synthesis occurred in non- or slow-growing chicks fed on diets deficient in lysine, total nitrogen or energy. Thus the machinery of protein synthesis in the young chick seems to be relatively insensitive to dietary manipulation. In the leg muscles, there was a small but significant correlation between the fractional rate of growth and protein synthesis. A decrease in the fractional rate of degradation, however, appeared to account for much of the accumulation of muscle protein in rapidly growing birds. In addition, the rapid accumulation of breast-muscle protein in rapidly growing chicks appeared to be achieved almost entirely by a marked decrease in the fractional rate of degradation.  相似文献

To determine if alterations in collagen degradation may contribute to the pathogenesis of fibrosis and cirrhosis, we studied the hepatic collagenase activity of 20 baboons given alcohol containing diets or control diets under carefully controlled experimental conditions. We also studied 28 patients whose livers were biopsied for diagnostic purposes. Hepatic collagenase activity was significantly increased in baboons with fatty liver compared to levels in normal, control fed animals [(1.98 +/- 0.19 vs 1.20 +/- 0.08 units (microgram collagen digested/hour/mg liver protein) +/- S.E.M., p less than 0.001)]. The increase in hepatic collagenase activity persisted at the stage of fibrosis when compared to the activity in control baboons (2.16 +/- 0.07 vs 1.20 +/- 0.08 units +/- S.E.M., p less than 0.001). A single cirrhotic baboon available for study had an hepatic collagenase activity of 1.58 units. Patients with hepatic fibrosis had significantly higher hepatic collagenase activity than those with fatty livers [(9.12 +/- 0.94 (n =14) vs 4.52 +/- 0.54 (n = 7) units +/- S.E.M., p less than 0.001)]. However, in the group with cirrhosis, hepatic collagenase was lower [(3.92 +/- 0.61 (n = 7) units +/- S.E.M., p less than 0.001)] than in the group with fibrosis. Our findings suggest that the development of cirrhosis is coincident with, or favored by a failure of hepatic collagen degradative enzymes to keep pace with hepatic collagen synthesis.  相似文献

Summary A microbial sensor consisting of immobilized Clostridium butyricum, two gas permeable Teflon membranes and fuel cell type electrode was suitable for the determination of formic acid. When the sensor was inserted into the sample solution containing formic acid, the current increases to a steady state with a response time of 20 min. The relationship between the steady state current and the formic acid concentration is linear up to 1 000 mg l–1. The currents are reproducible with an average relative error of 5%. Selectivity of the sensor is satisfactory. Results obtained with this sensor and by gas chromatography were in good agreement (regression coefficient; 0.98) when the cultivation medium of Aeromonas formicans was employed. Immobilized Clostridium butyricum is stable for more than 20 days.  相似文献

Within three hours of the administration of hepatotoxic doses of carbon tetrachloride to rats there was a substantial increase in the ability of liver extracts to catalyze the accumulation of monoacetylspermidine when incubated with spermidine and acetyl-CoA. This increase was maximal by six hours and correlated with the period in which there was a pronounced fall in hepatic spermidine and concomitant increase in putrescine. During this time there is a large increase of the conversion of labeled spermidine into putrescine in the liver. These results therefore suggest that this conversion requires the prior acetylation of spermidine.  相似文献

Fluid and enzyme secretion from a number of mammalian exocrine glands is controlled by the action of neurotransmitters and hormones on acinar cell membranes. Sustained stimulation evoking sustained fluid and enzyme secretion also evokes sustained membrane depolarization and increase in conductance. Mouse and rat pancreatic fluid and enzyme secretion, as well as membrane depolarization and conductance increase evoked by sustained stimulation with acetylcholine or cholecystokinin-gastrin peptides, are acutely dependent on extracellular calcium. However, the initial stimulant-evoked conductance increase and secretion appear to be triggered by calcium released from inside the cells. Direct measurement of membrane current during sustained stimulation in voltage-clamp experiments with resolution of the total current into its Na, Cl and K components has allowed calculations of stimulant-evoked Na and Cl uptake into the acinar cells. The NaCl uptake is quantitatively sufficient to account for the stimulant-evoked fluid secretion. The role of the stimulant-evoked transmembrane ionic current appears to be the supply of salt for the fluid secretion. Calcium derived from intracellular sources in the initial phase of secretion, and from the extracellular fluid in the sustained phase, couples fluid and enzyme secretion to hormone-receptor interaction.  相似文献

A nitrogen-fixing bacterium, Azotobacter chroococcum, was immobilized in 2% agar gel. The optimum partial oxygen pressure, pO2, of immobilized cells was 0.2 atm, wherea s that of native cells was 0.05 atm. When continual nitrogen fixation was performed under aerobic conditions, the nitrogenase activity of immobilized cells increased with increasing time. On the other hand, the activity of native cells decreased rapidly. Increase of nitrogenase activity was attributed to growth of the bacteria in the gel matrix. The production rate of total nitrogen compounds by the immobilized bacteria was also increased during the first 4 days. Nitrogen compounds produced by the immobilized cells were mainly amino acids such as γ-aminobutyrate, glutamate and arginine.  相似文献
