In higher plants, histidine-aspartate phosphorelays (two-component system) are involved in hormone signaling and stress responses. In these systems, histidine-containing phosphotransfer (HPt) proteins mediate the signal transmission from sensory histidine kinases to response regulators, including integration of several signaling pathways or branching into different pathways. We have determined the crystal structure of a maize HPt protein, ZmHP2, at 2.2 A resolution. ZmHP2 has six alpha-helices with a four-helix bundle at the C-terminus, a feature commonly found in HPt domains. In ZmHP2, almost all of the conserved residues among plant HPt proteins surround this histidine, probably forming the docking interface for the receiver domain of histidine kinase or the response regulator. Arg102 of ZmHP2 is conserved as a basic residue in plant HPt proteins. In bacteria, it is replaced by glutamine or glutamate that form a hydrogen bond to Ndelta atoms of the phospho-accepting histidine. It may play a key role in the complex formation of ZmHP2 with receiver domains. 相似文献

Chattonella verruculosa Y. Hara et Chihara was re‐examined by molecular methods and microscopic examination. The 18S rDNA phylogenetic analysis clearly indicated that C. verruculosa is a member of the Dictyochophyceae, with a specific affinity to Florenciella parvula. The morphological features in C. verruculosa– namely the proximal helix with two gyres and many scattered DNA‐containing areas in the chloroplasts – display the evolutionary link to the Dictyochophyceae, instead of the Raphidophyceae. Similarly, unique pyrenoid morphologies are shared between C. verruculosa and the dictyochophycean algae. Combining the molecular data and morphological characteristics, C. verruculosa is transferred to Pseudochattonella gen. nov. of the class Dictyochophyceae as Pseudochattonella verruculosa (Y. Hara et Chihara) Hosoi‐Tanabe, Honda, Fukaya, Inagaki et Sako comb. nov. 相似文献

Improved inosine producers were found with a high frequency among the mutants resistant to a low concentration of 8-azaguanine derived from AMP deaminase negative adenine auxotrophs of Bacillus subtilis K strain. The best mutant accumulated 16~18 g/liter of inosine, 60~80% higher than the parent. PRPP amidotransferase and succino-AMP lyase of all of the improved inosine producers tested were not repressed by adenosine but still repressed by guanosine. Adenine permeability was suggested to be also altered in some of the mutants which produced inosine even in the presence of a high concentration of adenine. Adenine prototrophic revertants from all of the mutants tested accumulated a small amount of adenosine but not inosine. 相似文献

Mitochondrial nucleoids (mt-nucleoids) isolated from the yeastSaccharomyces cerevisiae were analyzed to identify the proteincomponents that are involved in the compact packaging of mtDNA.The isolated mt-nucleoids were disassembled by the additionof 2 M NaCl and the disassembled mt-nucleoids were reassembledonce again into compact structures by dialysis against a bufferthat contained NaCl at concentrations below 0.1 M, as monitoredby staining of the DNA with 4',6-diamidino-2-phenylindole. DNA-binding proteins with molecular masses of 67 kDa, 52 kDa,50 kDa, 38 kDa, 30 kDa and 20 kDa were separated from isolatedmt-nucleoids by column chromatography on DNA cellulose afterdigestion of mt-nucleoids by DNase I in the presence or absenceof 2 M NaCl. Purified mtDNA was compactly packaged into nucleoid-likestructures upon the addition of fractions that contained DNA-bindingproteins and subsequent dialysis to reduce the concentrationof NaCl. Five proteins, with molecular masses of 67 kDa, 52kDa, 50 kDa, 38 kDa and 30 kDa, respectively, had lower affinityfor double-stranded DNA than that of the 20-kDa protein. Thefraction that contained the five DNA-binding proteins otherthan the 20-kDa protein was also able to fold mtDNA compactlyinto nucleoid-like structures. By contrast, the combinationof the 20-kDa protein and mtDNA resulted in formation of lesstightly packed, string-of-bead structures. The bird's nest fungi (Basidiomycota, Agaricales) package millions of spores into peridioles that are splashed from their basidiomata by the impact of raindrops. In this study we report new information on the discharge mechanism in Crucibulum and Cyathus species revealed with high-speed video. Peridioles were ejected at speeds of 1–5 m per second utilizing less than 2 % of the kinetic energy in falling raindrops. Raindrops that hit the rim of the basidiome were most effective at ejecting peridioles. The mean angle of ejection varied from 67 to 73° and the peridioles travelled over an estimated maximum horizontal distance of 1 m. Each peridiole carried a cord or funiculus that remained in a condensed form during flight. The cord unravelled when its adhesive surface stuck to a surrounding obstacle and acted as a brake that quickly reduced the velocity of the projectile. In nature, this elaborate mechanism tethers peridioles to vegetation in a perfect location for browsing by herbivores. 相似文献

Interspecies somatic cell nucleus transfer (iSCNT) could be a useful bioassay system for assessing the ability of mammalian somatic cells to develop into embryos. To examine this possibility, we performed canine iSCNT using porcine oocytes, allowed to mature in vitro, as recipients. Canine fibroblasts from the tail tips and dewclaws of a female poodle (Fp) and a male poodle (Mp) were used as donors. We demonstrated that the use of porcine oocytes induced blastocyst formation in the iSCNT embryos cultured in porcine zygote medium-3. In Fp and Mp, the rate of blastocyst formation from cleaved embryos (Fp: 6.3% vs. 22.4%; and Mp: 26.1% vs. 52.4%) and the number of cells at the blastocyst stage (Fp: 30.7 vs. 60.0; and Mp: 27.2 vs. 40.1) were higher in the embryos derived from dewclaw cells than in those derived from tail-tip cells (P < 0.05). The use of donor cells of any type in later passages decreased the rate of blastocyst formation. Treatment with trichostatin-A did not improve the rate of blastocyst formation from cleaved dewclaw cell-derived embryos but did so in the embryos derived from the tail-tip cells of Fp. Only blastocysts derived from dewclaw cells of Mp developed outgrowths. However, outgrowth formation was retrieved in the embryos derived from dewclaw cells of Fp by aggregation at the 4-cell stage. We inferred that iSCNT performed using porcine oocytes as recipients could represent a novel bioassay system for evaluating the developmental competence of canine somatic cells. 相似文献

Rotenoids and related compounds were investigated for their effects on animal and plant viruses. Of 35 compounds examined, rotenone, rotenone norketone, acetylrotenone, acetylrotenone norketone, deguelin, deguelic acid, dehydrodeguelin, and isotubanol norketone, all used at low concentrations, suppressed the growth of Newcastle disease and herpes simplex viruses as determined by the agar diffusion, plaque inhibition method. Most of the compounds likewise decreased the number of necrotic spots on tobacco mosaic virus-infected leaf discs. Only derrisic acid completely inhibited the local lesion formation at subphytotoxic concentrations. Correlation of antiviral activity with respiratory inhibition of these compounds is discussed. 相似文献

Stable isotope-labeled precursors were synthesized for an analysis by liquid chromatography-tandem mass spectrometry (LC-MS/MS) to elucidate the biosynthetic flow of capsaicinoids, capsinoids, and capsiconinoids. [1'-(13)C][5-(2)H]-Vanillin was prepared by the condensation of guaiacol with [(13)C]-chloroform and a D(2)O treatment. Labeled vanillylamine, vanillyl alcohol, ferulic acid, and coniferyl alcohol were prepared from the labeled vanillin. The labeled vanillylamine was converted to labeled capsaicinoid in a crude enzyme solution extracted from pungent Capsicum fruits. 相似文献
