Mating type (MAT)-specific fragments of the two idiomorphs ofGibberella fujikuroi (anamorph,Fusarium moniliforme) were obtained by PCR amplification using primers to conserved regions ofMAT homologs from other fungal species and used to assign mating type by molecular criteria rather than the arbitrary historical designation. Mating type—strains of mating populations A-E and a mating type+strain of mating population F carry an α-box motif and should therefore be designatedMAT-1. Mating type+strains of mating populations A-E and a mating type—strain of mating population F carry an HMG-box motif and should be designatedMAT-2. Thus, assessment of mating type ofG. fujikurol strains can be easily achieved usingMAT-specific primers.  相似文献

Five male and 9 female house musk shrews (Suncus murinus) captured in Koralawella, Sri Lanka were acquired by our laboratory. One male and 3 females alone contributed to the development of a new laboratory line (SRI line). The SRI line has been maintained as a closed breeding colony consisting of more than 40 shrews. Gestation period and litter size at birth averaged 31.2 days (43 cases) and 2.4 (43 litters), respectively. The SRI line shrews were uniformly covered with light gray fur. The body weight and total length of adult shrews averaged 72.9 g and 23.9 cm in males, and 49.6 g and 21.0 cm in females. These data indicate a body size intermediate between BAN line shrews originating from Bangladesh and Nag or OKI line shrews from Japan. The SRI shrews are also characterized by having 30 chromosomes in diploid, being the smallest number in the entire species, whereas the otherlines (Nag, OKI and BAN) all have 40 chromosomes.  相似文献

Dark accumulation of chlorophyll was demonstrated in calluscells of Nicotiana glutinosa. The chlorophyll content of dark-growncallus cells was dependent on the agar concentration in theculture medium: the content was significantly higher at lowerconcentrations of agar. A cytopathogenic agent was isolated in monkey kidney (MK) cell cultures from the stool specimen of a 3-month-old Filipina hospitalized with lower respiratory disease. The agent was designated the Drilon strain. It was characterized as an enterovirus on the basis of electron microscopic morphology, nucleic acid type (RNA), resistance to ether and acid (pH 3.0) treatments, stabilization by molar MgCl2 against heat inactivation, and buoyant density in CsCl. The strain caused mild febrile illness in experimentally inoculated cynomolgus monkeys, but not in suckling mice. In addition to its effect in primary MK cells, the virus was cytopathogenic in primary and secondary human amnion or embryonic lung cell cultures and in WI-38 or HEp-2 cell lines, but not in primary bovine kidney, primary porcine kidney, primary embryonic mouse or primary embryonic chick cell cultures. The Drilon strain was not neutralized by reference antisera against the known enterovirus serotypes, and the antiserum prepared with the Drilon strain did not neutralize any of the recognized prototype enterovirus strains. Although the patient's sera were not available, antibodies against the Drilon strain were prevalent in normal Filipinos and Indonesians, but not in Japanese people. The Drilon strain fulfilled the criteria of human enterovirus and is considered a candidate for designation as a new type.  相似文献
