Here we describe a simple high-performance liquid chromatography (HPLC) procedure for the simultaneous detection and quantitation in standard solutions of 13 important metabolites of cellular energy metabolism, including 9 tricarboxylic acid (TCA) cycle components and 4 additional metabolites. The metabolites are detected by their absorbance at 210 nm. The procedure does not require prior derivatization, and an analysis can be carried out at ambient temperature within 15 min. The significance of the current work is that the current HPLC procedure should motivate the development of simplified TCA cycle enzyme assays, isotopomer analysis, and determination of selected TCA metabolite levels in plasma/tissues.  相似文献

Degradation of immunoactive peptide splenopentin and its analogue N-acetylsplenopentin in human serum has been investigated by 1H NMR spectroscopy. It is shown that degradation of splenopentin occurs due to hydrolysis of peptide bonds in its N-terminal part, whereas in N-acetylsplenopentin peptide bonds in C-terminal part of the molecule are cleaved. Degradation pathways and life times of these peptides in human serum are established.  相似文献

The blends of polylactide with ethyl cellulose and chitosan are obtained in a Brabender mixer under conditions of high temperature shear deformation at different initial ratios of the reagents. The investigation of physicomechanical properties of the compositions has shown that the systems have high rigidity. Polyethylene glycol (PEG) and high-molecular-weight polyolefin polydecene have been added to the compositions in order to improve their elasticity. It has turned out that polydecene scarcely affects the mechanical characteristics, while PEG leads to a noticeable increase in elongation at break. Biodegradability of the compositions is investigated by the mass loss of samples after holding in soil, fungus resistance tests, and the analysis of the film morphology by scanning electron microscopy after holding in soil. It is found that the introduction of the third component (PEG) leads to an increase in biodegradability of the compositions.  相似文献

The content of 11-hydroxycorticosteroids, free and bound hydroxyproline in the peripheral blood, as well as the level of free and net hydroxyproline in the aortic wall and myocardium of rabbits were studied in chronic prolonged electrical stimulation of the mesencephalon reticular system. The experiments have shown that electrical stimulation leads to the activation of adrenal cortex function and is accompanied by alterations in collagen metabolism, accumulation of collagen in the aorta and decreased collagen level in the myocardium.  相似文献

The complex preparations (CP) containing yeast mannan, diacetyl-2,4-dioxihexahydro-1,3,5-triasyne, cyanoguanidine, alcan-sulfo-acids, or secondary metabolites of Pseudomonas sp. manifest in the inhibitory activity against tumors induced by A. tumefaciens in the parenchymatous tissue of potato tubers. As a rule, antitumor activity of CP is higher in the prophylactic application compared to the therapeutic application of the preparations.  相似文献

The Afanasievo Quarry, approximately 90 km southeast of Moscow and about 5 km southwest of Voskresensk, starts with Late Moscovian limestone of the Peski Formation, which is overlain by shallow-water carbonates of the Krevyakinian Horizon (Substage) (Suvorovo and Voskresensk formations) and the lower part of the Khamovnikian Horizon (Substage) (Ratmirovo and Neverovo formations). These rocks were formed under the strong influence of glacio-eustatic sea-level fluctuations and are separated by the palaeosol horizons and minor stratigraphic gaps. The Moscovian–Kasimovian transition interval contains fusulinids, brachiopods, bryozoans, corals, and conodonts. It was recently proposed [Villa, E., Task Group, 2005. Report of the Task Group to establish GSSPs at the Moscovian–Kasimovian and Kasimovian-Gzhelian boundaries. Newsletter on Carboniferous Stratigraphy 23, 9–10] that the appearance of the conodont Idiognathodus sagittalis Kozitskaya is one of the best markers for definition of the base of the Kasimovian on the global chronostratigraphic scale. The first appearance of this species is 2 m above the base of Neverovo Formation, and is close to the first appearance of the fusulinid Montiparus in the section. The possible ancestor of I. sagittalis occurs in the lower Suvorovo Formation, but is more abundant and more advanced in the middle to upper parts of the Voskresensk Formation. This lineage has potential for defining a GSSP at the first appearance of I. sagittalis.  相似文献

Mitochondria of the yeast Endomyces magnusii were examined for the presence of a Ca2+- and phosphate-induced permeability of the inner mitochondrial membrane (pore). For this purpose, coupled mitochondria were incubated under conditions known to induce the permeability transition pore in animal mitochondria, i.e., in the presence of high concentrations of Ca2+ and P(i), prooxidants (t-butylhydroperoxide), oxaloacetate, atractyloside (an inhibitor of ADP/ATP translocator), SH-reagents, by depletion of adenine nucleotide pools, and deenergization of the mitochondria. Large amplitude swelling, collapse of the membrane potential, and efflux of the accumulated Ca2+ were used as parameters for demonstrating pore induction. E. magnusii mitochondria were highly resistant to the above-mentioned substances. Deenergization of mitochondria or depletion of adenine nucleotide pools have no effect on low-amplitude swelling or the other parameters. Cyclosporin A, a specific inhibitor of the nonspecific permeability transition in animal mitochondria, did not affect the parameters measured. It is thus evident that E. magnusii mitochondria lack a functional Ca2+-dependent pore, or possess a pore differently regulated as compared to that of mammalian mitochondria.  相似文献

The influence of low-intensity electromagnetic irradiation and exposure to cadmium on survival and reproduction rates of daphnia in the process of their ontogenesis is investigated. The effects of the irradiation are clearly manifested in the toxicological experiments at coactions of the irradiation and cadmium and are weakly exhibited at action of the irradiation itself. At all stages of development, the irradiation does not influence the survival rate of daphnia. However, the irradiation increases their sensitivity to cadmium effects.  相似文献

Dynamics of the reproduction in the trematode Echinostoma caproni parthenites (Echinostomatidae) was observed. Early laying and maturation of the generative cells are for the first time shown to be characteristic for all parthenogenetic generations. Really the process of reproduction had been finishing to the beginning of the generating of new age by parthenites. Mother sporocysts, as well as redia of different generations, in fact stop producing new generative cells with the beginning of the generating of new age, and assume the function of a brood pouch. This feature was considered previously as peculiar mainly to mother sporocysts. Data on the autotomy of the anterior body end in mother sporocysts are verified. In our opinion, these data are an evidence of an early manifestation of the evolutionary trend to the morpho-functional regress and disintegration of the parasitic stage of mother sporocyst.  相似文献

Being devoted to studies in structural peculiarities of chromatin distribution of interphase nuclei from eucaryote cells, the paper embraces literature data and the authors' results in this field. Main tendencies in development of the method, based on the qualitative and quantitative estimation of hetero- and euchromatin nucleus substance are considered. Basic successes achieved while estimating experimental and clinical materials are indicated. It is determined as promising to put the method into practice.  相似文献
