Using a fluorescent probe for superoxide, hydroethidine, we have demonstrated that glucose deprivation (GD) activates production of reactive oxygen species (ROS) in cultured cerebellar granule neurons. ROS production was insensitive to the blockade of ionotropic glutamate channels by MK-801 (10 microM) and NBQX (10 microM). Inhibitors of mitochondrial electron transport, i.e. rotenone (complex I), antimycin A (complex III), or sodium azide (complex IV), an inhibitor of mitochondrial ATP synthase--oligomycin, an uncoupler of oxidative phosphorylation--CCCP, a chelator of intracellular Ca2+--BAPTA, an inhibitor of electrogenic mitochondrial Ca2+ transport--ruthenium red, as well as pyruvate significantly decreased neuronal ROS production induced by GD. GD was accompanied by a progressive decrease in the mitochondrial membrane potential and an increase in free cytosolic calcium ions, [Ca2+](i). Pyruvate, BAPTA, and ruthenium red lowered the GD-induced calcium overload, while pyruvate and ruthenium red also prevented mitochondrial membrane potential changes induced by GD. We conclude that GD-induced ROS production in neurons is related to potential-dependent mitochondrial Ca2+ overload. GD-induced mitochondrial Ca2+ overload in neurons in combination with depletion of energy substrates may result in the decrease of the membrane potential in these organelles. 相似文献

The kinetic and pharmacological properties of low voltage-activated (LVA) Ca2+ channels were studied in neurons of the laterodorsal (LD) thalamic nucleus in brain slices from 12-day-old rats. A homogeneous
population of LVA Ca2+ channels was found in the tested neurons. LVA Ca2+ current evoked by a step depolarization from a holding potential more negative than −70 mV was found to be sensitive to nifedipine
(K
d=2.6 (M). This current gained its maximum at −55 mV and demonstrated fast monoexponential decay with the time constant of
32.3±4.0 msec (n=15). Lanthanum (1 μM) effectively blocked LVA Ca2+ current, while nickel (25 μM) did not affect this current. It is concluded that the channels that, according to their pharmacological

The trophic status of the eastern Gulf of Finland, where the largest Baltic metropolis St. Petersburg sits at the mouth of
the largest Baltic river Neva, is elevated but existing recommendations on water protection measures are controversial. In
this study, the effects of nutrient load reductions on this ecosystem were estimated with the aid of a three-dimensional coupled
hydrodynamic-biogeochemical model. As a reference, the contemporary seasonal dynamics were simulated with nutrient inputs
corresponding to the recent estimates of point and riverine sources. In order to eliminate the effects of natural inter-annual
variations, the computations were run under recurrent annual forcing for 3 years, until quasi steady-state seasonal dynamics
were reached. Reasonable comparability of simulated concentrations and biogeochemical fluxes to available field estimates
provides credibility to scenario simulations. These simulations show that substantial reductions of nutrient point sources
in St. Petersburg would affect only the Neva Bay as the immediate receptor of treated sewage waters, where primary production
could decrease by up to 20%. Eutrophication in the other parts of the Neva Estuary and in the entire eastern Gulf of Finland
would change insignificantly owing to increased nutrient import from the offshore waters. Therefore, more significant changes
can occur only via a reduction in nutrient pools in the open Gulf of Finland and the Baltic Proper, which would require a
longer time.

The structure of the neutral glycosphingolipids of the Ehrlich ascite carcinoma (EAC) cells was studied. The main four components were identified as glycosylceramide, lastosylceramide, N-acetylgalactosyllactosylceramide and galactosyl-N-acetyllactosylceramide (asialo-GM1). The neutral glycolipid pattern of the cells was found to depend on their density. Dilution of the cell suspension resulted in an increased content of asia-lo-GM1, whereas the content of the other neutral glycolipids remained unchanged. The possible connection between these changes and the earlier disclosed cell density dependence of the gangliosides in EAC cells is discussed. 相似文献
