Background

Agroforestry is a sustainable land use method with a long tradition in the Bolivian Andes. A better understanding of people’s knowledge and valuation of woody species can help to adjust actor-oriented agroforestry systems. In this case study, carried out in a peasant community of the Bolivian Andes, we aimed at calculating the cultural importance of selected agroforestry species, and at analysing the intracultural variation in the cultural importance and knowledge of plants according to peasants’ sex, age, and migration.

Methods

Data collection was based on semi-structured interviews and freelisting exercises. Two ethnobotanical indices (Composite Salience, Cultural Importance) were used for calculating the cultural importance of plants. Intracultural variation in the cultural importance and knowledge of plants was detected by using linear and generalised linear (mixed) models.

Results and discussion

The culturally most important woody species were mainly trees and exotic species (e.g. Schinus molle, Prosopis laevigata, Eucalyptus globulus). We found that knowledge and valuation of plants increased with age but that they were lower for migrants; sex, by contrast, played a minor role. The age effects possibly result from decreasing ecological apparency of valuable native species, and their substitution by exotic marketable trees, loss of traditional plant uses or the use of other materials (e.g. plastic) instead of wood. Decreasing dedication to traditional farming may have led to successive abandonment of traditional tool uses, and the overall transformation of woody plant use is possibly related to diminishing medicinal knowledge.

Conclusions

Age and migration affect how people value woody species and what they know about their uses. For this reason, we recommend paying particular attention to the potential of native species, which could open promising perspectives especially for the young migrating peasant generation and draw their interest in agroforestry. These native species should be ecologically sound and selected on their potential to provide subsistence and promising commercial uses. In addition to offering socio-economic and environmental services, agroforestry initiatives using native trees and shrubs can play a crucial role in recovering elements of the lost ancient landscape that still forms part of local people’s collective identity.

Adoptive therapy using T cells redirected to target tumor- or infection-associated antigens is a promising strategy that has curative potential and broad applicability. In order to accelerate the screening process for suitable antigen-specific T cell receptors (TCRs), we developed a new approach circumventing conventional in vitro expansion-based strategies. Direct isolation of paired full-length TCR sequences from non-expanded antigen-specific T cells was achieved by the establishment of a highly sensitive PCR-based T cell receptor single cell analysis method (TCR-SCAN). Using MHC multimer-labeled and single cell-sorted HCMV-specific T cells we demonstrate a high efficacy (approximately 25%) and target specificity of TCR-SCAN receptor identification. In combination with MHC-multimer based pre-enrichment steps, we were able to isolate TCRs specific for the oncogenes Her2/neu and WT1 even from very small populations (original precursor frequencies of down to 0.00005% of CD3+ T cells) without any cell culture step involved. Genetic re-expression of isolated receptors demonstrates their functionality and target specificity. We believe that this new strategy of TCR identification may provide broad access to specific TCRs for therapeutically relevant T cell epitopes.  相似文献

The potential natural distribution of deciduous forests in Central Asia is the subject of ongoing discussions. Ulmus pumila (Siberian elm) is the only tree species occurring in southern and south-eastern Mongolia. In the semi-arid Mongolian Gobi, the species is restricted to ravines and beds of semi-temporary rivers. Compared to zonal occurrences in moister northern Mongolia, elm trees in the Gobi were found to be larger, in spite of their slower growth. Recruitment was very rare in the field although germination studies revealed that seeds were viable, survived osmotic stress, and were tolerant of repeated cycles of moistening and drying. Thus, they should be capable of germination in episodically flooded river beds. Fingerprinting revealed that clonal growth is of negligible importance in the Gobi as almost all U. pumila individuals studied constituted separate genets. Given that many trees were <100 years old and must have become established under current climatic conditions, we infer that the current lack of recruitment is likely to be caused by grazing impact. Our data imply that Ulmus pumila could potentially be much more common in the drylands of southern Mongolia and northern China.  相似文献

The abundant centre hypothesis predicts that changing environmental conditions are detrimental to a species’ abundance and performance towards the periphery of its range. We tested these predictions for the perennial grass Stipa capillata, a species that is commonly found in steppes of Asia but is rare at its north‐western range edge, in central Europe. We compared 21 populations in dry grassland fragments in central Europe and 20 populations in steppe habitats of Kazakhstan. We studied 15 plant performance traits both in situ and under laboratory and common greenhouse conditions, including local density, plant size and biomass production, seed size, weight and viability. To assess environmental conditions, we assembled data on topography, soil properties and climatic parameters. Using variance components analysis and multivariate methods we analyzed whether plant performance and environmental attributes differed more, as predicted, between the core and peripheral regions or whether they differed more among their subregions or populations. Additionally, we tested whether performance was affected by the same set of environmental predictors in each region. Contrary to our expectations, plant performance traits were virtually unchanged between the range periphery and centre. As expected, macroclimatic conditions showed a significant difference between the two regions (annual mean temperature, annual precipitation). The only other measured environmental variable that differed significantly between the two regions was soil pH, which was lower in core‐range populations. Our findings thus do not support the notion of reduced performance at the range edge. Instead, our data lend support to earlier theories of relative habitat constancy, suggesting that peripheral populations can shift to other habitats through plasticity or adaptation.  相似文献

Temperature is an important selective agent in nature. Consequently, temperature-induced plasticity which may help buffering detrimental effects of temperature variation has received considerable attention over recent decades. Laboratory studies have almost exclusively used constant temperatures, while in nature, temperature typically shows pronounced daily fluctuations. Using a factorial design with constant versus fluctuating temperatures and a higher versus a lower mean temperature, we here investigate in the butterfly Lycaena tityrus whether the use of constant temperatures is justified. Fluctuating compared to constant temperatures caused shorter development times, increased heat but decreased cold stress resistance, decreased heat-shock protein expression, and increased immunocompetence. Thus, overall, fluctuating temperatures were more beneficial to the butterflies compared to constant ones. However, despite substantial variation across temperature regimes, the ranking of trait values among treatments remained largely unaffected (e.g. lower constant as well as fluctuating temperatures caused increased pupal mass). Thus, we tentatively conclude that there is no general reason for concern about using constant temperatures in studies investigating phenotypic plasticity, which seem to comprise a fair proxy. However, substantial differences in mean values as well as interactive effects suggest that one needs to be cautious. We further demonstrate negative effects of high temperatures on butterfly immune function, which seem to result from a trade-off between the latter and the heat shock response.  相似文献

Elevated levels of copper or silver ions in the environment are an immediate threat to many organisms. Escherichia coli is able to resist the toxic effects of these ions through strictly limiting intracellular levels of Cu(I) and Ag(I). The CusCFBA system is one system in E. coli responsible for copper/silver tolerance. A key component of this system is the periplasmic copper/silver-binding protein, CusF. Here the X-ray structure and XAS data on the CusF-Ag(I) and CusF-Cu(I) complexes, respectively, are reported. In the CusF-Ag(I) structure, Ag(I) is coordinated by two methionines and a histidine, with a nearby tryptophan capping the metal site. EXAFS measurements on the CusF-Cu(I) complex show a similar environment for Cu(I). The arrangement of ligands effectively sequesters the metal from its periplasmic environment and thus may play a role in protecting the cell from the toxic ion.  相似文献

With state‐of‐the‐art organic solar cells (OSCs) surpassing 16% efficiency, stability becomes critical for commercialization. In this work, the power of using photoluminescence (PL) measurements on plain films is demonstrated, as well as high‐performance liquid chromatography analysis to reveal the origin of UV instabilities in OSCs based on the most commonly used acceptors PC70BM ([6,6]‐phenyl‐C71‐butyric acid methyl ester), ITIC (3,9‐bis(2‐methylene‐(3‐(1,1‐dicyanomethylene)‐indanone))‐5,5,11,11‐tetrakis(4‐hexylphenyl)‐dithieno[2,3‐d:2′,3′‐d′]‐s‐indaceno[1,2‐b:5,6‐b′]dithiophene), and o‐IDTBR (indacenodithiophene‐based non‐fullerene acceptor). The UV dependent stability tests reveal instabilities in solar cells based on PC70BM and ITIC while devices based on o‐IDTBR are highly stable even under UV illumination. The analysis of solar cell devices based on charge extraction and sub‐bandgap external quantum efficiency only shows the UV‐dependent emergence of traps, while PL spectra of plain films on glass allows the disentanglement and identification of individual instabilities in multi‐component bulk‐heterojunction devices. In particular, the PL analysis demonstrates UV instabilities of PC70BM and ITIC toward the processing additive 1,8 diiodooctane (DIO). The chemical analysis reveals the in‐depth mechanism, by providing direct proof of photochemical reactions of PC70BM and ITIC with UV‐induced radicals of DIO. Based on this scientific understanding, it is shown how to stabilize PBQ‐QF:PC70BM devices.  相似文献

In several insect species, male mating success is higher in older than in younger males, although condition diminishes dramatically with age. Two hypotheses are under debate to explain the counterintuitive pattern of old male mating advantage: first, an increased eagerness of older males to mate, driven by their low residual reproductive value, and second female preference for older males based on chemical cues such as sex pheromones (female choice hypothesis). In a series of experiments, we manipulated female olfaction, male pheromone blend and female age to test whether old male mating advantage prevails when the influence of male sex pheromones is controlled for, using the tropical butterfly Bicyclus anynana as model. We found that older males had a higher mating success than younger ones irrespective of female scent‐sensitivity and irrespective of male pheromone blend. Interestingly, older males were found to court more often and for longer time bouts than younger males. These results were independent of female age, although younger males courted younger females more often and for longer bouts than older females. Taken together, our results indicate that male courtship activity (1) is higher in older compared to younger males and (2) increases the mating success of older males. Olfaction and sensing pheromones, in contrast, were not a necessary prerequisite for old male mating advantage to occur and may use other cues than pheromones to assess male quality.  相似文献

Ecological immunology examines the adaptive responses of animals to pathogens in relation to other environmental factors and explores the consequences of trade-offs between investment in immune function and other life-history traits. Among species of herbivorous insects, diet breadth may vary greatly, with generalists consuming a wide variety of plant families and specialists restricted to a few species. Generalists may thus be exposed to a wider range of pathogens exerting stronger selection on the innate immune system. To examine whether this produces an increase in the robustness of the immune response, we compared larvae of the generalist herbivore Heliothis virescens and the specialist Heliothis subflexa challenged by entomopathogenic and non-pathogenic bacteria. Heliothis virescens larvae showed lower mortality, a lower number of recoverable bacteria, lower proliferation of haemocytes and higher phagocytic activity. These results indicate a higher tolerance to entomopathogenic bacteria by the generalist, which is associated with a more efficient cell-mediated immune response by mechanisms that differ between these closely related species. Our findings provide novel insights into the consequences of diet breadth and related environmental factors, which may be significant in further studies to understand the ecological forces and investment trade-offs that shape the evolution of innate immunity.  相似文献
