In Tenebrio molitor, as well as in other biological systems, there are indications that differences in leucyl-tRNA synthetase activity may play a role in translational control. However, it has not been clear whether the difference in activity is due to the appearance of a multiplicity of enzymes during development or to the alteration of a single enzyme.The purification of leucyl-tRNA synthetase from day 1 and day 7 after the larval pupal molt of Tenebrio molitor is described. The enzyme from both developmental stages was purified over a 1000-fold. The two enzyme preparations are identical in molecular weight (99,000). They show the same characteristics after aging. The pH optimum, heat inactivation behavior, and dependency on divalent cations are the same for both enzymes. They also show identical kinetics with similar values of Km for leucine, ATP, Mg2+, and tRNA day 1. However, leucyl-tRNA synthetase purified from day 7 exhibits an additional function in recognizing a new species of isoaccepting tRNA in day 7 tRNA. We have tentatively concluded that the two enzymes are probably different forms of the same enzyme and the additional activity is due to alteration of the enzyme at the macromolecular level during development.  相似文献

Barnacle adhesion strength was used to screen seventy-seven polydimethylsiloxane elastomeric coatings for fouling-release properties. The test coatings were designed to investigate the effect on barnacle adhesion strength of silicone fluid additive type, additive location, additive molecular weight, additive loading level, mixtures of additives, coating matrix type and coating fillers. The type of silicone fluid additive was the primary controlling factor in barnacle fouling-release. The type of silicone matrix in which the fluid resided was found to alter the effect on fouling-release. Two PDMS fluids, DMSC15 and DBE224, significantly reduced the adhesion strength of barnacles compared to unmodified elastomers. Optimum fouling-release performance was dependent on the interaction of fluid type and elastomeric matrix.  相似文献

A cross between two different races (race 7xrace 25) of the soybean root and stem rot pathogen Phytophthora sojae was analyzed to characterize the genomic region flanking two cosegregating avirulence genes, Avr4 and Avr6. Both genes cosegregated in the ratio of 82:17 (avirulent:virulent) in an F(2) population, suggestive of a single locus controlling both phenotypes. A chromosome walk was commenced from RAPD marker OPE7.1C, 2.0cM distant from the Avr4/6 locus. Three overlapping cosmids were isolated which included genetic markers that flank the Avr4/6 locus. The chromosome walk spanned a physical distance of 67kb which represented a genetic map distance of 22.3cM, an average recombination frequency of 3.0kb/cM and 11.7-fold greater than the predicted average recombination frequency of 35.3kb/cM for the entire P. sojae genome. Six genes (cDNA clones) expressed from the Avr4/6 genomic region encompassed by the cosmid contig were identified. Single nucleotide polymorphisms and restriction fragment length polymorphisms showed these six genes were closely linked to the Avr4/6 locus. Physical mapping of the cDNA clones within the cosmid contig made it possible to deduce the precise linkage order of the cDNAs. None of the six cDNA clones appear to be candidates for Avr4/6. We conclude that two of these cDNA clones flank a physical region of approximately 24kb and 4.3cM that appears to include the Avr4/6 locus.  相似文献

Objective: To describe the determinants, specifically age, body mass index, percentage of body fat, and physical activity (PA) level, associated with over‐ and underestimation of energy expenditure (EE) using PA records and the Stanford Seven‐Day Physical Activity Recall (7DR) compared with doubly labeled water (DLW). Research Methods and Procedures: We collected PA measures on 24 males eating a controlled diet designed to maintain body weight, and we determined EE from DLW and estimated EE from PA records and 7DR. Results: Absolute differences in the estimation of EE between DLW and PA assessment methods were greater for the 7DR (30.6 ± 9.9%) than PA records (7.9 ± 3.2%). In PA records, overestimation of EE was greater with older age and higher body fatness; EE was overestimated by 16.7% among men 50 years and older compared with only 5.3% among men <40 years of age. For percentage of body fat, EE was overestimated by 19.7% among men with a percentage of body fat ≥30% compared with only 5.6% among men with a percentage of body fat <25%. A trend for less overestimation of EE with higher levels of PA (measured by DLW/basal metabolic rate [BMR]) also was observed in the PA records. In the 7DR, the estimates of EE varied widely and no trends were observed by age, percentage of body fat, and PA levels. Discussion: Estimation of EE from the 7DR is considerably more variable than from PA records. Factors related to age and percentage of body fat influenced the accuracy of estimated EE in the PA record. Additional studies are needed to understand factors related to accurate reporting of PA behaviors, which are used to estimate EE in free‐living adults.  相似文献

An extensive search resulted in the identification of pamoic acid as an inhibitor of superoxide dismutases. Pamoic acid appeared to rapidly and reversibly inhibit all types of superoxide dismutases and did so in both the cytochrome c reduction and in the dianisidine photooxidation assays, used to measure this activity. It could nevertheless be shown that pamoic acid did not at all inhibit superoxide dismutase but rather diminished the sensitivity of the assays. The mechanism proposed to account for this effect involved oxidation of pamoate, by O2?, to yield a pamoate radical which can then reduce cytochrome c or oxidize pyrogallol. Pamoate thus competes with superoxide dismutase for the available O2?, without affecting the observable effects of that O2? upon cytochrome c or upon pyrogallol. It consequently makes these assays less responsive to superoxide dismutase, while appearing to be without effect in the absence of superoxide dismutase. Several of the predicted consequences of this proposal were affirmed. Other workers, interested in finding inhibitors for superoxide dismutases, are hereby forwarned of this subtle snare.  相似文献

Entry into mitosis requires the activation of cdk1/cyclin B, while mitotic exit is achieved when the same kinase activity decreases, as cyclin B is degraded. Cyclin B proteolysis is mediated by the anaphase promoting complex, or APC, an E3 ligase that is active at anaphase in mitosis through G1. We have identified a G1 substrate of the APC that we have termed Tome-1, for trigger of mitotic entry. Tome-1 is a cytosolic protein required for proper activation of cdk1/cyclin B and mitotic entry. Tome-1 associates with Skp-1 and is required for degradation of the cdk1 inhibitory tyrosine kinase wee1; Tome-1 therefore appears to be acting as part of an SCF-type E3 for wee1. Degradation of Tome-1 during G1 allows for wee 1 accumulation during interphase, thereby providing a critical link between the APC and SCF pathways in regulation of cdk1/cyclin B activity and thus mitotic entry and exit.  相似文献

With exposure to trace amounts of air and moisture, the Cr2(II, II) complex Cr2(μ-3,5Cl2-form)4, where 3,5Cl2-form is [(3,5-Cl2C6H3)NC(H)N(3,5-Cl2C6H3)], undergoes an oxidative addition reaction. Structural information from the X-ray crystal structure of the edge-sharing bioctahedral (ESBO) Cr2(III, III) product Cr2(μ-OH)2(μ-3,5Cl2-form)22-3,5Cl2-form)2 (1) indicates 1 has a significantly longer Cr–Cr distance [2.732(2) Å] than Cr2(μ-3,5Cl2-form)4 [1.9162(10) Å], but the shortest Cr–Cr distance in an ESBO Cr2(III, III) complex recorded to date.  相似文献

This work describes the long-term acclimation of the halotolerant microalga Dunaliella viridis to different photon irradiance, ranging from darkness to 1500 μmol m−2 s−1. In order to assess the effects of long-term photoinhibition, changes in oxygen production rate, pigment composition, xanthophyll cycle and in vivo chlorophyll fluorescence using the saturating pulse method were measured. Growth rate was maximal at intermediate irradiance (250 and 700 μmol m−2 s−1). The increase in growth irradiance from 700 to 1500 μmol m−2 s−1 did not lead to further significant changes in pigment composition or EPS, indicating saturation in the pigment response to high light. Changes in Photosystem II optimum quantum yield (Fv/Fm) evidenced photoinhibition at 700 and especially at 1500 μmol m−2 s−1. The relation between photosynthetic electron flow rate and photosyntetic O2 evolution was linear for cultures in darkness shifting to curvilinear as growth irradiance increased, suggesting the interference of the energy dissipation processes in oxygen evolution. Carbon assimilation efficiencies were studied in relation to changes in growth rate, internal carbon and nitrogen composition, and organic carbon released to the external medium. All illuminated cultures showed a high capability to maintain a C:N ratio between 6 and 7. The percentage of organic carbon released to the external medium increased to its maximum under high irradiance (1500 μmol m−2 s−1). These results suggest that the release of organic carbon could act as a secondary dissipation process when the xanthophyll cycle is saturated. In 1955, Lüscher proposed a ventilation mechanism forcathedral-shaped termite mounds to exchange respiratory gases. This mechanism was generally accepted, although it had neverbeen tested critically. We tested this mechanism by investigatingtemperatures, CO2 concentrations, and air currents in and aroundtwo types of Macrotermes bellicosus mounds: cathedral-shapedmounds with many ridges and thin walls located in the savannaand dome-shaped mounds without ridges and with thick wallsin the forest. These two mound shapes have two different mechanismsof ventilation, depending on the environmental temperature.In the savanna during the day, sun heats the air in the peripheralair channels inside the ridges of the mound above the centralnest temperatures and produces a temperature gradient in theperipheral air channels with decreased temperatures at thetop of the mound. This temperature gradient leads to convectioncurrents with air rising inside the air channels of the ridgesto the top of the mound, meanwhile exchanging CO2. In contrast,in the savanna during the night and generally in the forest,the temperatures inside the air channels are lower than thoseof the central nest, and no air currents rising upward insidethe air channels were detected. The CO2 concentrations in theair channels of savanna mounds at night and forest mounds ingeneral were higher than during the day in the savanna.
