Phylogenetic diversity (PD, the diversity of lineages) and functional diversity (FD, the diversity of functional traits or groups in a biological community) reflect important yet poorly understood attributes of species assemblages. Until recently, few studies have examined the spatial variation of PD and FD in natural communities. Yet the relationships between PD and FD and area (termed PDAR and FDAR), like the analogous species–area relationship (SAR), have received less attention and may provide insights into the mechanisms that shape the composition and dynamics of ecological communities. In this study, we used four spatial point process models to evaluate the likely roles of the random placement of species, habitat filtering, dispersal limitation, and the combined effects of habitat filtering and dispersal limitation in producing observed PDARs and FDARs in two large, fully mapped temperate forest research plots in northeast China and in north‐central USA. We found that the dispersal limitation hypothesis provided a good approximation of the accumulation of PD and FD with increasing area, as it did for the species area curves. PDAR and FDAR patterns were highly correlated with the SAR. We interpret this as evidence that species interactions, which are often influenced by phylogenetic and functional similarity, may be relatively unimportant in structuring temperate forest tree assemblages at this scale. However, the scale‐dependent departures of the PDAR and FDAR that emerged for the dispersal limitation hypothesis agree with operation of competitive exclusion at small scales and habitat filtering at larger scales. Our analysis illustrates how emergent community patterns in fully mapped temperate forest plots can be influenced by multiple underlying processes at different spatial scales.  相似文献

The Ran protein regulates nucleocytoplasmic transport mediated by the karyopherin family of nuclear transport factors. Ran is converted to the active, GTP bound form in the nucleus and then binds to a conserved domain found in all karyopherins. This interaction induces cargo binding for exportins and cargo release for importins. In either case, the Ran.GTP is then transported to the cytoplasm by the karyopherin, where it is hydrolyzed to Ran.GDP. To ask whether Ran could function as a nuclear mRNA export factor, we fused Ran to the MS2 coat protein and inserted MS2 RNA-binding sites into an unspliced cat mRNA that is normally sequestered in the nucleus. Coexpression of MS2-Ran induced cat mRNA export and CAT enzyme expression as effectively as, for example, an MS2-Rev fusion protein. MS2-Ran dependent nuclear mRNA export was reduced by inhibitors specific for Crm1, but not blocked as was seen with MS2-Rev. Consistent with the hypothesis that Crm1 is not the only karyopherin cofactor for MS2-Ran mediated mRNA export, we show that not only Crm1 but also CAS, transportin, importin beta and exportin t can all export mRNA from the nucleus when tethered via the MS2 RNA-binding domain. In contrast, two shuttling hnRNPs, hnRNP A1 and hnRNP K, proved unable to function as nuclear RNA export factors when expressed as MS2 fusions. Together, these data argue that karyopherins that normally function to transport proteins into or out of the nucleus are also capable of exporting tethered mRNA molecules.  相似文献

We suggest that the conscious use of information that is "hidden" in distinct structures in nature itself and in data extracted from nature (=pattern) during the process of modeling (=pattern-oriented modeling) can substantially improve models in ecological application and conservation. Observed patterns, such as time-series patterns and spatial patterns of presence/absence in habitat patches, contain a great deal of data on scales, site-history, parameters and processes. Use of these data provides criteria for aggregating the biological information in the model, relates the model explicitly to the relevant scales of the system, facilitates the use of helpful techniques of indirect parameter estimation with independent data, and helps detect underlying ecological processes. Additionally, pattern-oriented models produce comparative predictions that can be tested in the field. We developed a step-by-step protocol for pattern-oriented modeling and illustrate the potential of this protocol by discussing three pattern-oriented population models: (1) a population viability analysis for brown bears ( Ursus arctos ) in northern Spain using time-series data on females with cubs of the year to adjust unknown model parameters; (2) a savanna model for detecting underlying ecological processes from spatial patterns of tree distribution; and (3) the incidence function model of metapopulation dynamics as an example of process integration and model generalization. We conclude that using the pattern-oriented approach to its full potential will require a major paradigm shift in the strategies of modeling and data collection, and we argue that more emphasis must be placed on observing and documenting relevant patterns in addition to attempts to obtain direct estimates of model parameters.  相似文献

Objective: Although the obesity epidemic is progressing in European children too, there is no consensus on the population‐specific prevalence of comorbidities or efficient diagnostic strategies. Therefore, weight‐related risk factors, their interrelationship, and association with biological parameters were assessed in a large group of overweight (OW) children, documented by an electronic database. Methods and Procedures: Data of 26,008 children (age 12.6 ± 2.9 years, 56% females) presented for OW (BMI > 90th percentile) or obesity (>97th percentile) in 98 specialized centers were evaluated using a simple software (Adipositas Patienten Verlaufsbeobachtung (APV)) for standardized longitudinal documentation. After local anonymization, data were transmitted for central analysis including multiple logistic regression. Results: A total of 5.9% of the children were normal weight, 41% obese (OB), and 37% extremely OB (>99.5th percentile, XXL; 41% of the girls). In 50%, at least one risk factor and in 11% a cluster of two were found, comprising increased blood pressure (BP): 35.4%, dyslipidemia: 32% (total cholesterol: 14.1%, low‐density lipoprotein (LDL)‐cholesterol: 15.8%, high‐density lipoprotein (HDL)‐cholesterol: 11.1%, triglycerides: 14.3%), impaired glucose tolerance (IGtT): 6.5% and suspicion of diabetes: 0.7%. The degree of OW was inversely associated with HDL‐cholesterol and directly with clustered risk factors, impaired glucose metabolism, increased BP and triglycerides (odds ratios (ORs) XXL vs. normal = 6.15, >10, 4.3, 3.0 and 2.5, respectively), but not with LDL‐cholesterol. Discussion: In a very large cohort of young Europeans risk factors for cardiovascular (CV) diseases are frequently found, related to the degree of OW and tend to cluster, thus a comprehensive screening is justified in all OW or OB children. Electronic patient documentation is feasible in a large obesity care network.  相似文献

Nuclear export of the incompletely spliced mRNAs encoded by several complex retroviruses, including human immunodeficiency virus type 1 (HIV-1), is dependent on a virally encoded adapter protein, termed Rev in HIV-1, that directly binds both to a cis-acting viral RNA target site and to the cellular Crm1 export factor. Human endogenous retrovirus K, a family of ancient endogenous retroviruses that is not related to the exogenous retrovirus HIV-1, was recently shown to also encode a Crm1-dependent nuclear RNA export factor, termed K-Rev. Although HIV-1 Rev and K-Rev display little sequence identity, they share the ability not only to bind to Crm1 and to RNA but also to form homomultimers and shuttle between nucleus and cytoplasm. We have used mutational analysis to identify sequences in the 105-amino-acid K-Rev protein required for each of these distinct biological activities. While mutations in K-Rev that inactivate any one of these properties also blocked K-Rev-dependent nuclear RNA export, several K-Rev mutants were comparable to wild type when assayed for any of these individual activities yet nevertheless defective for RNA export. Although several nonfunctional K-Rev mutants acted as dominant negative inhibitors of K-Rev-, but not HIV-1 Rev-, dependent RNA export, these were not defined by their inability to bind to Crm1, as is seen with HIV-1 Rev. In total, this analysis suggests a functional architecture for K-Rev that is similar to, but distinct from, that described for HIV-1 Rev and raises the possibility that viral RNA export mediated by the approximately 25 million-year-old K-Rev protein may require an additional cellular cofactor that is not required for HIV-1 Rev function.  相似文献

The relationship between heterogeneous nuclear RNA (hnRNA) and messenger RNA (mRNA) synthesis has been studied as a function of the development of the sea urchin embryo through the use of methyl incorporation. Several parameters in the metabolism of capped hnRNA and mRNA of early blastula and late gastrula stages have been investigated by measuring the kinetics of transfer of methyl groups from S-adenosylmethionine to the 5′ cap structures in nuclear and cytoplasmic RNA:
  • 1 The rate constants for the decay of hnRNA caps and the synthesis of mRNA caps are equal to within experimental error. This equality indicates a flux of precursor hnRNA caps to mRNA caps with a very high degree of conservation of the hnRNA caps. This conservation holds for each embryonic stage.
  • 2 From literature data on the labeling kinetics of GTP and mRNA, we have calculated the decay constant of a putative mRNA precursor component of hnRNA. The value of this constant is very close to that for the decay constant of hnRNA caps. Hence, all hnRNA caps and some portion of their associated hnRNA sequences behave kinetically as the pre-mRNA fraction. This kinetically ascribed pre-mRNA comprises approximately 30% of the hnRNA mass.
  • 3 The part of the hnRNA which does not serve as precursor to mRNA turns over at least twice as rapidly as the pre-mRNA fraction.
  • 4 During development from early blastula to late gastrula, the rate of hnRNA cap synthesis drops from 2 × 103 molecules/min/cell to half of this value. This decline is parallel to the decline in total hnRNA synthesis and thereby confirms the constant degree of capping of hnRNA, as previously reported. We infer that the pre-mRNA fraction of hnRNA remains nearly constant during this developmental period.

We present a technique for estimating size-age relations and size-dependent mortality patterns of long-lived plants. The technique requires two sets of size data of individual (non-marked) plants that should be collected with a time-lag of several years in the same area of a study site. The basic idea of our technique is to assume general (three parameter) families of size-dependent functions which describe growth and mortality that occurred between the two data gathering events. We apply these growth and mortality functions to the size data of the early data set and construct predicted size-class distributions to compare it, in a systematic way, to the size-class distribution of the later data set. In a next step we calculate the size-age relations from the resulting growth functions, which yield the smallest difference between observed and predicted size-class distribution. Applying this technique to size data of five dominant shrub species at the Tierberg study site in the semiarid Karoo, South Africa produced new insight into the biology of these species which otherwise cannot be obtained without frequent measurements of marked plants. We could relate characteristics of growth behavior and mortality, for certain subgroups of the five species, to the life-history attributes evergreen vs. deciduous, succulent vs. woody, and early reproductive vs. late reproductive. The results of our pilot-study suggest a broad applicability of our technique to other shrublands of the world. This requires at least one older record of (individual) shrub-size data and performance of resampling.  相似文献

Abstract. We used a spatially explicit simulation model to examine the impact of small-scale disturbance (created by the digging of aardvarks, Orycteropus afer, and bat-eared foxes, Otocyon megalotis or as a management action) on the temporal and spatial dynamics of a typical Karoo shrub plant community, and to gather insight into the interplay between disturbance structure and population dynamics. Establishment, growth, mortality, seed dispersal and competitive interactions were modelled over long time-scales in annual time-steps under the influence of stochastic and unpredictable rainfall. Three disturbance regimes were included, varying the type, rate and size of the small-scale disturbances. The impact of a disturbance regime on long-term community dynamics depends on complex interactions between disturbance characteristics and life-history attributes of component species. Plant density decreased with overall disturbance rates; this effect was independent of the type of disturbance. A given type and rate of disturbance did not influence all species within a guild (e.g. colonizer species) in the same way. The reason for these differences was that species responded not only to the disturbance but to changes in competition intensity from other species and changes in their reproductive potential relative to other species as well. Such interactions resulted in a sequential change in dominant species within guilds as disturbance rates increased. An increase in the overall disturbance rate did not always produce the trend in evenness expected from the intermediate disturbance hypothesis, but was influenced by the relative abundance of different types of disturbance.  相似文献

Background

Chagas disease control campaigns relying upon residual insecticide spraying have been successful in many Southern American countries. However, in some areas, rapid reinfestation and recrudescence of transmission have occurred.

Methodology/Principal Findings

We conducted a cross-sectional survey in the Bolivian Chaco to evaluate prevalence of and risk factors for T. cruzi infection 11 years after two rounds of blanket insecticide application. We used a cubic B-spline model to estimate change in force of infection over time based on age-specific seroprevalence data. Overall T. cruzi seroprevalence was 51.7%. The prevalence was 19.8% among children 2–15, 72.7% among those 15–30 and 97.1% among participants older than 30 years. Based on the model, the estimated annual force of infection was 4.3% over the two years before the first blanket spray in 2000 and fell to 0.4% for 2001–2002. The estimated annual force of infection for 2004–2005, the 2 year period following the second blanket spray, was 4.6%. However, the 95% bootstrap confidence intervals overlap for all of these estimates. In a multivariable model, only sleeping in a structure with cracks in the walls (aOR = 2.35; 95% CI = 1.15–4.78), age and village of residence were associated with infection.

Conclusions/Significance

As in other areas in the Chaco, we found an extremely high prevalence of Chagas disease. Despite evidence that blanket insecticide application in 2000 may have decreased the force of infection, active transmission is ongoing. Continued spraying vigilance, infestation surveillance, and systematic household improvements are necessary to disrupt and sustain interruption of infection transmission.  相似文献
