The effects of habitat restoration measures designated to promote farmland biodiversity have been documented at the field scale, but little is known about their role in restoring the agricultural mosaic. In this study, we analyzed the effects of wildflower strips (WFS) at the field scale and in the landscape context on butterflies in a Swiss arable landscape. Three hypotheses were tested: (1) butterfly diversity and abundance are higher in WFS than in conventional fields; (2) butterfly diversity and abundance are enhanced by the amount, proximity and connectivity of WFS in the landscape context; (3) additional factors influence butterfly diversity and abundance according to individual site conditions and landscape context characteristics linked to other landscape elements. WFS had more species and individuals of butterflies than conventional habitats. However, promoted species were mainly generalists; few specialists were enhanced. The diversity of all butterflies and of generalists increased linearly with percent cover of WFS, reflecting an effect of restoration measures depending on the landscape context. The influence of proximity and connectivity of WFS were, however, not significant. The occurrence of specialists was conditioned by plant species richness, while the effect of WFS for overall diversity was affected by the amount of grassland in the surroundings. We conclude that to increase the effectiveness of biodiversity‐orientated restoration measures, their implementation should be steered toward increasing the share of WFS in the landscape. However, the combination of WFS with additional restoration measures might be needed to halt the decline of specialist species. 相似文献

In temperate regions, populations of Aedes aegypti survive the cold season in the egg stage. In the present work, we studied the cold‐season mortality of Ae. aegypti eggs and their subsequent hatching pattern in Buenos Aires city. Eggs were exposed during the winter season (three months) in three neighborhoods located along a gradient of distance toward the Río de la Plata River, coincident with a gradient of activity of Ae. aegypti. Results showed mortalities lower (30.6%) than those from tropical regions during the dry season. Significant differences were detected among the egg mortalities of each site with a maximum value at the site nearest the Río de la Plata River (50%), and a minimum value at the most continental site (9%). Post‐experimental hatching response of eggs differed between sites, with the highest proportion of hatched eggs during the first immersion in the site nearest to the river and the lowest proportion in the most continental site. The hatching proportion also differed between age classes, with older (early‐laid) eggs hatching later than new (late‐laid) ones. Our results provide the first information of Ae. aegypti egg mortality in temperate South America and support the hypothesis that differences in egg mortality are associated with abundance patterns of Ae. aegypti in Buenos Aires city. 相似文献

Current viral vectors for gene therapy are associated with serious safety concerns, including leukemogenesis, and nonviral vectors are limited by low gene transfer efficiency. Here we investigate the therapeutic utility of chemically modified mRNA as an alternative to DNA-based gene therapy. A combination of nucleotide modifications abrogates mRNA interaction with Toll-like receptor (TLR)3, TLR7, TLR8 and retinoid-inducible gene I (RIG-I), resulting in low immunogenicity and higher stability in mice. A single intramuscular injection of modified murine erythropoietin mRNA raises the average hematocrit in mice from 51.5% to 64.2% after 28 days. In a mouse model of a lethal congenital lung disease caused by a lack of surfactant protein B (SP-B), twice weekly local application of an aerosol of modified SP-B mRNA to the lung restored 71% of the wild-type SP-B expression, and treated mice survived until the predetermined end of the study after 28 days. 相似文献

Family studies suggest a genetic component to the etiology of chronic kidney disease (CKD) and end stage renal disease (ESRD). Previously, we identified 16 loci for eGFR in genome-wide association studies, but the associations of these single nucleotide polymorphisms (SNPs) for incident CKD or ESRD are unknown. We thus investigated the association of these loci with incident CKD in 26,308 individuals of European ancestry free of CKD at baseline drawn from eight population-based cohorts followed for a median of 7.2 years (including 2,122 incident CKD cases defined as eGFR <60ml/min/1.73m2 at follow-up) and with ESRD in four case-control studies in subjects of European ancestry (3,775 cases, 4,577 controls). SNPs at 11 of the 16 loci (UMOD, PRKAG2, ANXA9, DAB2, SHROOM3, DACH1, STC1, SLC34A1, ALMS1/NAT8, UBE2Q2, and GCKR) were associated with incident CKD; p-values ranged from p = 4.1e-9 in UMOD to p = 0.03 in GCKR. After adjusting for baseline eGFR, six of these loci remained significantly associated with incident CKD (UMOD, PRKAG2, ANXA9, DAB2, DACH1, and STC1). SNPs in UMOD (OR = 0.92, p = 0.04) and GCKR (OR = 0.93, p = 0.03) were nominally associated with ESRD. In summary, the majority of eGFR-related loci are either associated or show a strong trend towards association with incident CKD, but have modest associations with ESRD in individuals of European descent. Additional work is required to characterize the association of genetic determinants of CKD and ESRD at different stages of disease progression. 相似文献

Background
Reactive astrocytes are implicated in the development and maintenance of neuroinflammation in the demyelinating disease multiple sclerosis (MS). The sphingosine kinase 1 (SphK1)/sphingosine1-phosphate (S1P) receptor signaling pathway is involved in modulation of the inflammatory response in many cell types, but the role of S1P receptor subtype 3 (S1P3) signaling and SphK1 in activated rat astrocytes has not been defined.Methodology/Principal Findings
Using immunohistochemistry we observed the upregulation of S1P3 and SphK1 expression on reactive astrocytes and SphK1 on macrophages in MS lesions. Increased mRNA and protein expression of S1P3 and SphK1, as measured by qPCR and Western blotting respectively, was observed after treatment of rat primary astrocyte cultures with the pro-inflammatory stimulus lipopolysaccharide (LPS). Activation of SphK by LPS stimulation was confirmed by SphK activity assay and was blocked by the use of the SphK inhibitor SKI (2-(p-hydroxyanilino)-4-(p-chlorphenyl) thiazole. Treatment of astrocytes with a selective S1P3 agonist led to increased phosphorylation of extracellular signal-regulated kinase (ERK)-1/2), which was further elevated with a LPS pre-challenge, suggesting that S1P3 upregulation can lead to increased functionality. Moreover, astrocyte migration in a scratch assay was induced by S1P and LPS and this LPS-induced migration was sensitive to inhibition of SphK1, and independent of cell proliferation. In addition, S1P induced secretion of the potentially neuroprotective chemokine CXCL1, which was increased when astrocytes were pre-challenged with LPS. A more prominent role of S1P3 signaling compared to S1P1 signaling was demonstrated by the use of selective S1P3 or S1P1 agonists.Conclusion/Significance
In summary, our data demonstrate that the SphK1/S1P3 signaling axis is upregulated when astrocytes are activated by LPS. This signaling pathway appears to play a role in the establishment and maintenance of astrocyte activation. Upregulation of the pathway in MS may be detrimental, e.g. through enhancing astrogliosis, or beneficial through increased remyelination via CXCL1. 相似文献950.
