A rapid and effective RP‐UHPLC‐DAD method for enantioseparation of three flavanones, i.e., flavanone, naringenin, and hesperetin, was developed and validated. Chromatographic separation of the analytes was performed using a Chiralpak AD‐3R analytical column under reverse phase conditions with methanol as the mobile phase. The method was validated in the concentration range of 0.2 to 50 µg/mL for enantiomers of flavanone and 0.5 to 50 µg/mL for enantiomers of naringenin and hesperetin. The limits of quantification were between 0.03 to 0.5 µg/mL. Intraday and interday precision were below 14% and accuracy varied from 0.04 to 8.17%. The growth of microorganisms is affected by cultivation conditions, concentration of carbon and nitrogen sources and the presence of trace elements. One of the new possibilities of influencing the production of cell mass or lipids is the use of lanthanides. Lanthanides are biologically non-essential elements with wide applications in technology and industry and their concentration as environmental contaminants is therefore increasing. Although non-essential, lanthanides have been proposed (and even used) to produce beneficial effects in plants but their mechanisms of action are unclear. Recently, it was suggested that they may replace essential elements or operate as potent blockers of Ca2+ channels. We tested the effect of low concentrations of lanthanides on traditional biotechnologically useful yeast species (Kluyveromyces polysporus, Saccharomyces cerevisiae, Torulospora delbrueckii), and species capable of high accumulation of lipids (Rhodotorula glutinis, Trichosporon cutaneum, Candida sp., Yarrowia lipolytica). Low concentrations of lanthanum and monazite were conducive to an increase in cell mass and lipids and also higher production of palmitoleic acid, commonly used in cosmetics and medicine, and ω6-linoleic acid which is a precursor of thromboxanes, prostaglandins and leucotrienes.  相似文献

In order to study the mechanism of the enhancement of solution chemiluminescence, the kinetics of the decay of the oxidant and the chemiluminescence emission were followed for oxidations by permanganate, manganese dioxide sol and Mn3+(aq) of glyoxylic acid, using stopped‐flow spectrophotometry. Results are reported for the glyoxylic acid oxidized under pseudo first‐order conditions and in an acidic medium at 25 °C. For permanganate under these conditions, the decay is sigmoidal, consistent with autocatalysis, and for manganese dioxide sol and Mn3+ it is pseudo first order. The effects of the presence of aqueous formaldehyde and Mn2+ were observed and a fit to a simple mechanism is discussed. It is concluded that chemiluminescent enhancement in these systems is best explained by reaction kinetics. Copyright © 2014 John Wiley & Sons, Ltd.  相似文献

OBJECTIVE: Inwardly-rectifying K(+) (Kir) channels are responsible for maintaining membrane potentials in a variety of cell types including endothelial cells where they modulate endothelium-dependent vasorelaxation. The goal of this study is to determine the functional expression of Kir channels in porcine bone marrow-derived side population (BM-SP) cells that demonstrate phenotypes of endothelial progenitor cells (EPCs). We further asses the hypercholesterolemia sensitivity of Kir channels in BM-SP cells, which may play a key role in hypercholesterolemia-mediated regulation of EPCs. METHODS: To assess the effect of hypercholesterolemia on Kir channels in BM-SP, Kir currents were recorded in SP cells sorted from the bone marrow of healthy or hypercholesterolemic animals. RESULTS: We found Kir channels constitute the major conductance in porcine bone marrow-derived side population (BM-SP) cells. These cells are defined by their efficiency of Hoechst dye efflux and have been reported to differentiate into multiple cell lineages including endothelium in vivo. We demonstrate here that porcine BM-SP cells differentiate to an endothelial lineage (CD31(+), vWF(+)) supporting the hypothesis that these cells are endothelial progenitor cells. Also, BM-SP cells express Kir with biophysical properties recapitulating those in mature endothelial cells, but with a much higher current density. Flow cytometric (FACS) analysis indicated that the number of SP cells was unaffected by hypercholesterolemia. However, hypercholesterolemia significantly inhibited Kir channels in BM-SP cells. CONCLUSIONS: We successfully demonstrate that BM side population cells represent an origin of endothelial progenitor cells. This study further shows, for the fist time, that the functional expression of Kir channels in bone marrow (BM)-derived SP. Moreover, we demonstrate that hypercholesterolemia condition significantly suppresses the Kir channels in BM-SP cells, suggesting that hypercholesterolemia-mediated regulation of Kir channels may be an important factor not only in dysfunction of mature endothelium but also in dysfunction of BM-SP cells.  相似文献

Response of the invasive species Heracleum mantegazzianum to experimental removal of tissues was studied in the Czech Republic. The study aimed at determining (i) how efficiently plants respond, in terms of quantity and quality of produced fruit, to the removal of different amounts of generative and/or vegetative tissues; and (ii) whether regeneration ability depends on the time of treatment. Total number of fruits and their mean weight were reduced by removal of leaves, but germination percentage and rate of germination did not differ from control. More vigorous individuals compensated for the loss of tissues to a higher degree, and the advantage of being larger increased with severity of the treatment. Of 40 plants with umbels completely removed, 18 (45.0%) regenerated and produced on average 103.4 ± 220.1 (mean ± SD) fruits. Total fruit numbers and total fruit weight of regenerating plants significantly differed neither among treatments nor from the control, but some treatments resulted in poorer germination compared to the control. Umbels removed from plants at the beginning of fruit development and left at the locality produced 18.6% of fruit numbers of control plants, and 24% of these seeds germinated. Control by removing umbels from plants must ensure that they are collected and destroyed. From the management viewpoint, there is a trade-off between later umbel removal, resulting in more efficient reduction in fecundity but necessity to handle more developed fruits, and early treatment, leading to a high regeneration, that produces seed of sufficient quantity and very little affected in terms of quality.  相似文献

A cDNA library for Myzus persicae has served to identify sequences coding for cuticular proteins (CPs) with RR-1 and RR-2 consensus. Two putative CPs showed a common RR-2 chitin binding domain (CBD) but differed in their C and N terminals. Two other predicted CPs showed a typical RR-1 CBD but differed in size and sequence of the C and N terminals. An additional sequence encoding for a protein that showed terminal amino acid repeats similar to those of putative CPs from M. persicae, but lacked the R & R consensus, was also described. A comparison of the sequences obtained from the cDNA library with those attained from the genomic DNA, confirmed their identity as cuticular proteins genes. Presence of introns was revealed in the Mpcp4 and Mpcp5 genes coding for CPs with an RR-1 consensus. The Mpcp4 has a single large intron, while the Mpcp5 has two shorter ones. Introns were not found in the Mpcp2 and Mpcp3 genes encoding for CPs with RR-2 consensus. Differences were also noticed for 3' UTR and 5' UTR of both the RR-1 and RR-2 CPs. CPs genes were expressed in bacteria, and the resulting protein was identified as a CP by amino acid sequencing.  相似文献

Background  

Unsupervised annotation of proteins by software pipelines suffers from very high error rates. Spurious functional assignments are usually caused by unwarranted homology-based transfer of information from existing database entries to the new target sequences. We have previously demonstrated that data mining in large sequence annotation databanks can help identify annotation items that are strongly associated with each other, and that exceptions from strong positive association rules often point to potential annotation errors. Here we investigate the applicability of negative association rule mining to revealing erroneously assigned annotation items.  相似文献
