A mathematical model (Kliemann, W. 1987.Bull. math. Biol.
49, 135–152.) that predicts the quantitative branching pattern of dendritic tree was evaluated using the apical and basal dendrites
of rat hippocampal neurons. The Wald statistics for χ2-test was developed for the branching pattern of dendritic trees and for the distribution of the maximal order of the tree.
Using this statistic, we obtained a reasonable, but not excellent, fit of the mathematical model for the dendritic data. The
model's predictability of branching patterns was greatly enhanced by replacing one of the assumptions used for the original
model “splitting of branches for all dendritic orders is stochastically independent”, with a new assumption “branches are
more likely to split in areas where there is already a high density of branches”. The modified model delivered an excellent
fit for basal dendrites and for the apical dendrites of hippocampal neurons from young rats (30–34 days postpartum). This
indicates that for these cells the development of dendritic patterns is the result of a purely random and a systematic component,
where the latter one depends on the density of dendritic branches in the brain area considered. For apical dendrites there
is a trend towards decreasing pattern predictability with increasing age. This appears to reflect the late arrival of afferents
and subsequent synaptogenesis proximal on the apical dendritic tree of hippocampal neurons. 相似文献

The metabolism of the weak carcinogen 7-methylbenz[c]acridine (7MBAC) was examined in rat liver microsomes from 3-methylcholanthrene(MC)-induced animals by the use of mixed 14C- and 2H-labelled substrate. The three metabolites identified by spectroscopic and chromatographic examination were 7-OHMBAC and two dihydrodiols. Polygonal networks in cultured chick endoderm cells are ordered arrays of actin microfilaments situated just beneath the dorsal cell surface. Each strut is formed from a bundle of microfilaments and 5-7 bundles intersect at each node. Dense bodies are seen in nodes and some struts. At its periphery the network is attached to the substrate at the termini of long radial struts. Most of the network is resistant to detergent extraction. Sliding microfilaments can explain the observed behaviour of networks in live cells. 相似文献

An EMG biofeedback program was developed for a 56-year-old Parkinsonism patient who exhibited pathological lip hypertonia and retraction. The program was designed to achieve the following goals: (1) to demonstrate a reduction in postural lip hypertonicity and (2) to demonstrate a reduction in lip hypertonicity during a series of increasingly complex speech activities. To achieve the first goal, contrastive tasks of full contraction and relaxation were utilized. Each posture was sustained while voltage measurements were made at specific intervals. Procedures to modify lip retraction during speech included five tasks in which the patient was to monitor the audio feedback signal. The tasks involved: prolongation of a neutral vowel, consonant-vowel combinations, monosyllabic words, sentences, and a paragraph-reading task. Data collected over six biofeedback sessions are presented. Trend analyses showed consistent muscular reduction within each task. The following explanations for the decrease in the patient's hypertonicity were discussed: (1) reduction of anisometric contraction, (2) reduction of isometric contraction, (3) relearning of agonistic-antagonistic muscle balance. 相似文献

In order to study cell proliferation in intact human cervical epithelium, a technique involving metaphase arrest has been utilized. Metaphase accumulation is observed following intraepithelial administration of vincristine sulphate, at a predetermined optimum dose of 50 μg/ml. A significant delay before the onset of stathmokinetic activity is demonstrated; thereafter linearity of accumulation is apparent over a 4-hr period following injection. The technique appears applicable to the estimation and comparison of cell production rates in vivo of normal cervical epithelium, cervical intraepithelial neoplasia, wart-affected cervical epithelium and early invasive carcinoma. However, individual values are likely to be imprecise in isolation, indicating the need to study relatively large numbers of subjects in each group. Some practical difficulties are discussed. 相似文献

An application of the metaphase-arrest technique to human cervical epithelium, in vivo, was utilized to obtain cell birth rate data for seventy-six patients with normal and pathological cervical epithelium. Mean cell production rates for basal and parabasal layers of normal epithelium were 0.91 and 0.92 cells/1000 cells/hr respectively. Histologically normal epithelium adjacent to CIN (cervical intra-epithelial neoplasia) had a significantly higher rate for the parabasal layer compared to the ‘normal’ group (P < 0.05). Values for the lower two-thirds of CIN III were 8–10 times higher than for normal epithelium, with microinvasive carcinoma having the highest rates of all. Values for wart-affected cervical epithelium (NCWVI) were intermediate between normal and CIN, but there was activity in the superficial layer, possibly reflecting activity of the papilloma virus. Large variation in birth rates between individuals in the same histological category was noted for each group, this being particularly the case in six patients with early invasive carcinoma. The data is used to attempt to elucidate some of the uncertainties surrounding the aetiology and biological behaviour of cancer of the cervix and its precursors. Atlas cDNA cell interaction arrays (CLONTECH) were used to examine degenerate tissue from four patients with Achilles tendon disorders, in order to identify changes in expression of genes important in cell-cell and cell-matrix interactions. The greatest difference between normal (post-mortem) and degenerate tissue samples was in the level of MMP-3 (stromelysin) mRNA, which was down-regulated in all the degenerate samples. Quantitative RT-PCR assay of RNA extracted from paired 'normal' and degenerate Achilles tendon tissue samples taken from tendons during surgery mirrored the results of the arrays. Levels of MMP-3 mRNA were lower, whereas levels of type-I and type-III collagen mRNAs were significantly higher, in the degenerate compared to the 'normal' samples. Immunoblotting of proteins extracted from the same tendon samples showed that three of four normal tissue samples taken from individuals without apparent tendon disorder had much higher levels of MMP-3 protein than 'normal' or degenerate samples from patients with tendinosis. We suggest that MMP-3 may play an important role in the regulation of tendon extracellular matrix degradation and tissue remodelling. 相似文献
