研究了体外培养一种孟加拉传统香蕉(Musa spp.Cv. Kanthali)的茎尖组织。茎尖的原始细胞表面经无菌处理(0.1%HgCl2处理12min) ,接种6~15d后外植体地下茎部分仍有微生物污染(大部分是细菌) ,杀死了85%的外植体。为确定无污染培养基,将等量外植体分别浸泡在含400mg/L氨苄青霉素和200mg/L庆大霉素(两种光谱抗生素)的培养基中1h。结果表明,经抗生素处理的外植体完全没有污染,但培养3周后不能再生。进行二次继代培养后,其中一部分外植体吸收了培养基并胀大,颜色由苍白转变成浅绿或深绿。三次继代培养后数天,不再观察到外植体的生长,所有经抗生素处理过的外植体都开始死亡。在未经抗生素处理的活外植体中,单个茎发育的最佳培养基是:MS 4.0mg/L BA 0.5mg/L KT 15% CW,平均生长时间为18~21d,但再生率很低,只有30%。茎细胞增殖的最佳培养基是:MS 4.0mg/L BA 2.0mg/LIAA 15% CW,每个茎平均只萌发3~4个芽。最后,在添加0.5mg/LIBA的一半浓度的MS培养基中,体外培养茎最大生根率达到90%。  相似文献

Freestanding composite structures with embedded transition metal dichalcogenides (TMDCs) as the active material are highly attractive in the development of advanced electrodes for energy storage devices. Most 3D electrodes consist of a bilayer design involving a core–shell combination. To further enhance the gravimetric and areal capacities, a 3D trilayer design is proposed that has MoSe2 as the TMDC sandwiched in‐between an inner carbon nanotube (CNT) core and an outer carbon layer to form a CNT/MoSe2/C framework. The CNT core creates interconnected pathways for the e?/Na+ conduction, while the conductive inert carbon layer not only protects the corrosive environment between the electrolyte and MoSe2 but also is fully tunable for an optimized Na+ storage. This unique heterostructure is synthesized via a solvothermal‐carbonization approach. Due to annealing under a confined structural configuration, MoSe2 interlayer spaces are expanded to facilitate a faster Na+ diffusion. It is shown that an ≈3 nm thick carbon layer yielded an optimized anode for a sodium‐ion battery. The 3D porosity of the heterostructure remains intact after an intense densification process to produce a high areal capacity of 4.0 mAh cm?2 and a high mass loading of 13.9 mg cm?2 with a gravimetric capacity of 347 mAh g?1 at 500 mA g?1 after 500 cycles.  相似文献

From the world's tiny flying bugs to gigantic dobsonflies, inflight locomotion of a flying creature requires complex biomechanical strategies to cope with air turbulence. These unpredictable changes in ambient airflow strength and direction may destabilize body posture and orientation. To record this behaviour in further detail, we scientifically examined how houseflies (Musca domestica) respond to air turbulence. We then, three-dimensionally reconstructed body and wings motion of continuously perturbated houseflies using high-speed videography under laboratory condition. The findings confirmed that houseflies, in general, do not initiate flight when average ambient air speed exceeds ~0.63 ms?1 at approximately ~2% of relative turbulent intensity. This finding contrasts with flies which immediately take-off after being released. During mild turbulent conditions, flies performed take-off but with severe and active modulation of body postures. In addition, the body roll angle fluctuates more severely (18.5-fold increase) compared to yaw (7-fold of increment) and pitch (6.4-fold of increment) during turbulence, highlighting that body roll stability is highly sensitive. This research extends our current knowledge on flies' behaviours during turbulence and how insects achieve their superior flight performance.  相似文献

The basic principle of their organization appears very simple, but they are actually of high complexity and represent very dynamic structures. The interactions between bioactive molecules and lipids are important for numerous processes, from drug bioavailability to viral fusion. The cell membrane is a carefully balanced environment and any change inflicted upon its structure by a bioactive molecule must be considered in conjunction with the overall effect that this may have on the function and integrity of the membrane. Background

The study was conducted to evaluate the in vitro thrombolytic activity, and in vivo analgesic, anti-inflammatory and antipyretic potentials of different hydrocarbon soluble extracts of Litsea glutinosa leaves for the first time widely used in the folkloric treatments in Bangladesh. This work aimed to create new insights on the fundamental mechanisms of the plant extracts involved in these activities.

Results

In thrombolytic activity assay, a significant clot disruption was observed at dose of 1 mg/mL for each of the extracts (volume 100 μL) when compared to the standard drug streptokinase. The n-hexane, ethyl acetate, chloroform, and crude methanolic extracts showed 32.23 ± 0.26, 37.67 ± 1.31, 43.13 ± 0.85, and 46.78 ± 0.9% clot lysis, respectively, whereas the positive control streptokinase showed 93.35 ± 0.35% disruption at the dose of 30,000 I.U. In hot plate method, the highest pain inhibitory activity was found at a dose of 500 mg/kg of crude extract (15.54 ± 0.37 sec) which differed significantly (P <0.01 and P <0.001) with that of the standard drug ketorolac (16.38 ± 0.27 sec). In acetic acid induced writhing test, the crude methanolic extract showed significant (P <0.01 and P <0.001) analgesic potential at doses 250 and 500 mg/kg body weight (45.98 and 56.32% inhibition, respectively), where ketorolac showed 64.36% inhibition. In anti-inflammatory activity test, the crude methanolic extract showed significant (P <0.001) potential at doses 250 and 500 mg/kg body weight (1.51 ± 0.04 and 1.47 ± 0.03 mm paw edema, respectively), where ketorolac showed 1.64 ± 0.05 mm edema after 3 h of carrageenan injection. In antipyretic activity assay, the crude extract showed notable reduction in body temperature (32.78 ± 0.46°C) at dose of 500 mg/kg-body weight, when the standard (at dose 150 mg/kg-body weight) exerted 33.32 ± 0.67°C temperature after 3 h of administration.

Conclusions

Our results yield that the crude hydroalcoholic extract has better effects than the other in all trials. In the context, it can be said that the leaves of L. glutinosa possess remarkable pharmacological effects, and justify its traditional use as analgesic, antipyretic, anti-inflammatory, and thrombolytic agent.  相似文献

Background

Current study has been designed to evaluate the chemical composition of essential and fixed oils from stem and leaves of Perovskia abrotanoides and antioxidant and antimicrobial activities of these oils.

Results

GC-MS analysis of essential oil identified 19 compounds with (E)-9-dodecenal being the major component in stem and hexadecanoic acid in leaves. In contrast, GC-MS analysis of fixed oil showed 40 constituents with α-amyrin the major component in stem and α-copaene in leaves. The antioxidant activity showed the highest value of 76.7% in essential oil from leaves in comparison with fixed oil from stem (45.9%) through inhibition of peroxidation in linoleic acid system. The antimicrobial assay tested on different microorganisms (e.g. E. coli, S. aureus, B. cereus, Nitrospira, S. epidermis, A. niger, A. flavus and C. albicans) showed the higher inhibition zone at essential oil from leaves (15.2 mm on B. cereus) as compared to fixed oil from stem (8.34 mm on S. aureus) and leaves (11.2 mm on S. aureus).

Conclusions

The present study revealed the fact that essential oil analyzed from Perovskia abrotanoides stem and leaves could be a promising source of natural products with potential antioxidant and antimicrobial activities, as compared to fixed oil.  相似文献

Secondary metabolites from plants are a good source for the NSAID drug development. We studied the analgesic activity of ethanolic extract of Erythrina variegata L. (Fabaceae) followed by molecular docking analysis. The analgesic activity of Erythrina variegata L. is evaluated by various methods viz., acetic acid-induced writhing test, hot plate and tail immersion test. Subsequently, molecular docking analysis has been performed to identify compounds having activity against COX-1 and COX-2 enzymes by using GOLD docking fitness. The result of preliminary phytochemical screening revealed that the extract contains alkaloids and flavonoids. In analgesic activity tests, the extract at the doses of 50, 100 and 200 mg/kg body weight (b.w.) produced a increase in pain threshold in a dose dependent manner. In acetic acid induced writhing test, the inhibitory effect was similar to the reference drug diclofenac sodium. The extract showed 18.89% writhing inhibitory effect at the dose 200 mg/kg b.w., whereas diclofenac sodium showed 79.42% inhibition of writhing at a dose of 10 mg/kg b.w. The results of tail immersion and hot plate test also showed potential analgesic activity of the extract which is also comparable to the standard drug morphine (5 mg/kg b.w.). Docking studies shows that phaseollin of Erythrina variegata L. has the best fitness score against the COX-1 which is 56.64 and 59.63 for COX- 2 enzyme. Phaseollin of Erythrina variegata L. detected with significant fitness score and hydrogen bonding against COX-1 and COX-2 is reported for further validation.  相似文献

Neonates with intrauterine growth retardation (IUGR) are susceptible to decreases in cellular immunity. In recent years, a growing body of evidence indicates that Hsp70 may serve as a danger signal to the innate immune system and promote receptor-mediated apoptosis. Using neonatal pigs with IUGR, we investigated immune function of pigs and expression of heat shock protein 70 (Hsp70), nuclear factor-kappa B (NF-κB), and forkhead box O 3a (FoxO3a) in the intestinal tract. Samples from the blood, duodenum, jejunum, and ileum of normal body weight (NBW) piglets and IUGR piglets were collected at day 7 after birth. Furthermore, to test whether Hsp70 is associated with regulation of NF-κB and FoxO3a, Hsp70 was silenced using small RNA interference (siRNA) in IEC-6 cells.
