Macronuclear chromatin of Blepharisma japonicum compared to that of Tetrahymena pyriformis

Abstract The macronuclear chromatin of the ciliate Blepharisma japonicum was studied by electrophoretic and immunological techniques as well as by micrococcal nuclease digestion and circular dichroism spectroscopy. Under these experimental conditions the macronuclear chromatin of B. japonicum was compared to that of Tetrahymena pyriformis . Data obtained through this analysis showed that the macronuclear chromatin of B. japonicum is structurally more relaxed than that of T. pyriformis . A perchloric acid soluble polypeptide, referred to as P3, is the only polypeptide of B. japonicum chromatin to appear immunologically related to the H1 histone of T. pyriformis . It is suggested that this P3 polypeptide in B. japonicum should be considered functionally equivalent to the T. pyriformis H1 histone, even though it differed from it both in terms of molecular mass and relative concentration.     

Short-term impact of dry olive mill residue addition to soil on the resident microbiota

The short-term response of the resident soil bacterial and fungal communities to the addition of 5% (w/w) of either dry olive mill residue (DOR), DOR treated with Phlebia sp. (PTDOR) or DOR previously extracted with water (WEDOR) was investigated. As opposed to bacteria, the diversity of fungi increased upon the amendments as assessed by denaturing gradient gel electrophoresis of 18S rDNA. Over the first 30 days, phospholipid fatty acids analyses indicated a gradual decrease in the relative abundances of Gram⁺ bacteria (from 44.8% to 37.9%) and a concomitant increase of Gram⁻ bacteria (from 37.3% to 51.2%) in DOR-amended soil. A considerable increase in the fungal/bacterial ratio was observed after 7 days in DOR, WEDOR and PTDOR-amended soils with respect to the control (0.316, 0.165 and 0.265, respectively, vs. 0.011). The overall microbial activity was stimulated by the amendments as indicated by the higher activity levels of both dehydrogenase and fluorescein diacetate hydrolase. These results indicate that DOR at the application level examined is not toxic on soil microorganisms.     

Some notes on the terminology of brassinosteroids

In this paper the definitions of brassinolide, brassinolide activity andbrassins are reviewed, and definitions for the terms brassin, naturalbrassinosteroids and brassinosteroid analogues, based on biosynthetic reasoningand structure similarity are proposed.     

A New Basal Subfamily of mariner Elements in Ceratitis rosa and Other Tephritid Flies

Several copies of highly related transposable elements, Crmar2, Almar1, and Asmar1, are described from the genomes of Ceratitis rosa, Anastrepha ludens, and A. suspensa, respectively. One copy from C. rosa, Crmar2.5, contains a full-length, uninterrupted ORF. All the other copies, from the three species contain a long deletion within the putative ORF. The consensus Crmar2 element has features typical of the mariner/Tc1 superfamily of transposable elements. In particular, the Crmar2 consensus encodes a D,D41D motif, a variant of the D,D34D catalytic domain of mariner elements. Phylogenetic analysis of the relationships of these three elements and other members of the mariner/Tc1 superfamily, based on their encoded amino acid sequences, suggests that they form a new basal subfamily of mariner elements, the rosa subfamily. BLAST analyses identified sequences from other diptera, including Drosophila melanogaster, which appear to be members of the rosa subfamily of mariner elements. Analyses of their molecular evolution suggests that Crmar2 entered the genome of C. rosa in the recent past, a consequence of horizontal transfer.
