Multidrug resistance P-glycoprotein (MDR1) is a membrane proteinof 150-170 kDa that catalyzes the ATP-driven efflux of hydrophobic xenobiotics, including fluorescent dyes, from cells. Expressed in manyepithelial tissues and in the endothelia of the blood-brain barrier,the MDR1 protein provides major routes of detoxification. We found thattaste cells of the rat vallate papilla (VP; posterior tongue) had onlya slow increase in fluorescence due to uptake of the hydrophobic dyecalcein acetoxymethyl ester. However, the development of fluorescencewas accelerated two- to threefold by substrates and/orinhibitors of MDR1, such as verapamil, tamoxifen, and cyclosporin A,and by addition of the transport-blocking antibody to MDR1, UIC2.Western blots of vallate tissue rich in taste buds withthe MDR1-specific monoclonal antibodies C219 and C494 revealed animmunoreactive protein at ~170 kDa. In contrast, the lingual epithelium surrounding the VP showed a much weaker band with these antibodies. Furthermore, using the antibodies C494 and UIC2 with tissuesections, MDR1-like immunoreactivity was found in taste cells. Theseresults show that MDR1 is present and functional in vallate taste cellsof the rat. MDR1-related transport may achieve active elimination ofxenobiotics from the sensory cells and thereby protect the peripheraltaste organs from potentially harmful molecules contained in ananimal's food.

The lake number was used to describe the mixing condition forthree consecutive years (1992–1994) in Lake Tegel, Berlin,and compared to the successions of diatoms, dinoflagellatesand cyanobacteria, the main phytoplankton groups in the lake,as well as to diversity and similarity indices. Using both diversityand similarity indices in juxtaposition provides an indicationof the growth type of prevailing species (r- or K-strategists)and the degree of competition in the aqua-ecological system.A genera] pattern of these indices can be recognized as threephases: (i) high diversity—during spring, summer and autumn,interrupted by (ii) phases of low diversity during the latespring clear-water phase as the number of spring species plummeted,and (iii) during the late summer, climax populations of K-strategists.On a smaller time scale, similarity and diversity proved toreact sensitively to disturbances at frequencies intermediatein relation to the generation times of the phytoplankton. Thissupports the ‘intermediate disturbance hypothesis’,as proposed for phytoplankton by Padisak et al. [(eds) IntermediateDisturbance Hypothesis in Phytoplankton Ecology. Kluwer Academic,1993]. Diversity may remain quite high even for extended periodsduring summer climax situations, in conjunction with a highdegree of similarity, if deeper mixing of the epilimnion occursat time intervals of 2–3 weeks, as during the summer of1993. This enables the prevalence of ‘ruderal’ species,together with some motile K-strategists who actively seek optimaldepths for photosynthesis. During such summer situations describedby frequent occurrences of lower lake numbers, the epilimnionof Lake Tegel is mixed deeply enough to support ruderals, butnot too vigorously to counteract competitive advantages of motilespecies. The body mass index (BMI) is a complex phenotype representing the amount of fat mass, lean mass, body build and proportions, and it is likely to be affected by various metabolic processes, hormonal effects, energy intake and expenditure, and interactions within and among these broad categories of etiologic factors. Nonetheless, several previous studies have reported evidence for major gene segregation for the BMI in various populations. Data on a random sample of Caucasian families participating in the National Heart, Lung, and Blood Institute (NHLBI) Family Heart Study were analyzed to document the extent of familial resemblance and to investigate whether a similar monogenic inheritance pattern could be detected. Genetic analysis was carried out on age- and sex-adjusted BMI values. Familial correlations were significant implying a maximal heritability, including all genetic and environmentally inherited additive factors, of 41% to 59%. Segregation analysis revealed the presence of two maximum likelihood solutions, one characterized as a recessive Mendelian gene and the other as a major effect with an ambiguous transmission pattern. The presence of two such solutions is consistent with detection of two separate factors, each influencing the BMI distribution in a substantive manner. The evidence also supports a multifactorial background for BMI and suggests that the frequencies of these two factors, one of which appears to be a gene, may vary among diverse populations in the United States.  相似文献

The within‐field spatio‐temporal distributions and relationships of two pest insects with stem‐mining larvae, Psylliodes chrysocephala (L.) (Coleoptera: Chrysomelidae) and Ceutorhynchus pallidactylus (Marsham) (Coleoptera: Curculionidae), and their larval endoparasitoids, Tersilochus microgaster (Szépligeti) and Tersilochus obscurator Aubert (Hymenoptera: Ichneumonidae), respectively, were studied in a crop of winter oilseed rape [Brassica napus L. (Brassicaceae)]. The insects were sampled, at different life stages, at 40 spatially referenced points within the oilseed rape in 1998–99 and in the following crop of winter wheat. Distributions were analysed and compared using SADIE (Spatial Analysis by Distance IndicEs) and by tests of edge distribution. Tersilochus microgaster emerged from overwintering by the end of March and T. obscurator from mid April to mid May 2000. Tersilochus microgaster parasitized 10.8% of P. chrysocephala larvae, peak parasitism occurring in May. The distribution of each species was patchy and irregular. Psylliodes chrysocephala and its parasitoid were closely associated, but new generation C. pallidactylus and T. obscurator were less so, probably because of host mortality caused by the parasitoid. The two host–parasitoid pairs showed distributions that were polarized with respect to each other. The genesis of these spatial patterns is discussed in relation to the influence of wind direction, plant size, and interspecific interactions. The potential of these parasitoids as biocontrol agents in integrated pest management is discussed. Opportunities for conservation biocontrol in oilseed rape offered by spatio‐temporal targeting of insecticides to avoid parasitoids and by reduced soil tillage are examined.  相似文献

Oxidative stress is known to cause oxidative protein modification and the generation of reactive aldehydes derived from lipid peroxidation. Extent and kinetics of both processes were investigated during oxidative damage of isolated rat liver mitochondria treated with iron/ascorbate. The monofunctional aldehydes 4-hydroxynonenal (4-HNE), n-hexanal, n-pentanal, n-nonanal, n-heptanal, 2-octenal, 4-hydroxydecenal as well as thiobarbituric acid reactive substances (TBARS) were detected. The kinetics of aldehyde generation showed a lag-phase preceding an exponential increase. In contrast, oxidative protein modification, assessed as 2,4-dinitrophenylhydrazine (DNPH) reactive protein-bound carbonyls, continuously increased without detectable lag-phase. Western blot analysis confirmed these findings but did not allow the identification of individual proteins preferentially oxidized. Protein modification by 4-HNE, determined by immunoblotting, was in parallel to the formation of this aldehyde determined by HPLC. These results suggest that protein oxidation occurs during the time of functional decline of mitochondria, i.e. in the lagphase of lipid peroxidation. This protein modification seems not to be caused by 4-HNE.  相似文献

The 325-residue outer membrane protein OmpA of Escherichia coli has been proposed to consist of a membrane-embedded moiety (residues 1 to about 170) and a C-terminal periplasmic region. The former is thought to comprise eight transmembrane segments in the form of antiparallel -strands, forming an amphiphilic connected by exposed turns. Several questions concerning this model were addressed. Thus no experimental evidence had been presented for the turns at the inner leaflet of the membrane and it was not known whether or not the periplasmic part of the polypeptide plays a role in the process of membrane incorporation. Oligonucleotides encoding trypsin cleavage sites were inserted at the predicted turn sites of the ompA gene and it was shown that the encoded proteins indeed become accessible to trypsin at the modified sites. Together with previous results, these data also show that the turns on both sides of the membrane do not possess specifically topogenic information. In two cases one of the two expected tryptic fragments was lost and could be detected at low concentration in only one case. Therefore, bilateral proteolytic digestion of outer membranes can cause loss of -strands and does not necessarily produce a reliable picture of protein topology. When ompA genes were constructed coding for proteins ending at residue 228 or 274, the membrane assembly of these proteins was shown to be partially defective with about 20% of the proteins not being assembled. No such defect was observed when, following the introduction of a premature stop codon, a truncated protein was produced ending with residue 171. It is concluded that (1) the proposed -barrel structure is essentially correct and (2) the periplasmic part of OmpA does not play an active role in, but can, when present in mutant form, interfere with membrane assembly.  相似文献

Species composition and diversity of phytoplankton were studied for several years in two lakes which differ with respect to mixing conditions and nutrient limitation: Schlachtensee regularly stratifies very stably. In contrast, size and wind-exposure predispose Lake Tegel to deeper mixing; additionally, stratification is artificially destabilized by aeration. As the duration of aeration was varied, the study period includes interannual changes in mixing conditions. For both lakes, it also covers trophic change due to restoration; this was especially pronounced in Schlachtensee.Results show that mixing conditions affect species composition on two levels: on a superordinate level, lake morphology or hydrology govern stability of stratification and susceptibility to perturbation, and hence the extent to which motile species can develop. In Schlachtensee, species with some means of actively seeking preferred depths usually dominated during summer stratification: Planktothrix agardhii during the hypertrophic phase, and flagellates since restoration. In contrast, in Lake Tegel deeper mixing as a generally prevailing condition favored non-motile species. Their seasonal pattern was remarkably constant from year to year. Although changes in the extent of mixing were pronounced during the four years studied (1987–1990), these were within a range that affected species composition only slightly: in summer, cyanobacteria and diatoms represented climax species whose dominance was not offset by additional, weather-induced increases of turbulence.On a subordinate level, and within the constraints set by nutrient limitation as well as by grazing pressure, small-scale changes in mixing conditions caused by meteorological cycles were shown to strongly affect species composition and in consequence diversity: Results for the fouryear post-restoration study period at Schlachtensee show that considerable interannual variations of species composition and diversity can be attributed to variations in the frequency of meteorological changes. In accordance with the intermediate disturbance hypothesis (IDH), diversity was lowest during 1989, the year with the longest and most pronounced cycles of fair weather (14 to 27 fair days on end). However, the mechanism for this was rarely a decline of diversity caused by competitive exclusion within single long phases of stable conditions, as conceived by the intermediate disturbance hypothesis. Instead, diversity responded to changes in mixing conditions with a variety of patterns — often with low values during phases of increased mixing and with high values under quiescent conditions, especially during the first calm days just after increased mixing. Thus, not disturbance as such, but rather the rate of change between phases of disturbance and quiescence appears to determine the frequency of high diversity indices.In Lake Tegel, high diversity indices were somewhat more frequent in 1989, the year during which thermal stratification was most stable. For species adapted to frequent or continuous mixing, interjected calm phases with unusually high stability of thermal stratification may represent a disturbance of accustomed conditions. Thus, in turbulent Lake Tegel, meteorological cycles appear to act in reversal to the IDH, in a sense which may be termed intermediate quiescence hypothesis.Phosphorus limitation due to successful restoration was found to decrease winter and vernal diversity in Schlachtensee. Presumably, without nutrient constraints, new populations could grow more rapidly in response to the rapid changes of physical parameters during this season. In contrast, restoration has increased summer diversity, as phosphorus concentrations no longer allow the virtual monocultures of Planktothrix agardhii which prevailed previously. In Lake Tegel, the reduction of phosphorus concentration down to 60 µg/l P in 1989 limited biomass, but this level was still too high to significantly alter species composition or diversity.  相似文献

Interest in a single gene etiology for obesity, as assessed by the body mass index (BMI), has been spurred recently by reports of a putative recessive major gene for extreme values, which accounts for as much as 40% of the variance. The major gene hypothesis was evaluated here in the Québec Family Study, a random sample of 375 French-Canadian volunteer families. This report represents one component in a more complete investigation of obesity in these families. In contrast to the recent studies, a major gene hypothesis for BMI was not verified here. Although there was a major effect, it did not conform to a Mendelian pattern of transmission. A multifactorial component (i.e., polygenic and/or common environmental factors) accounted for 42% of the phenotypic variance. In addition, evidence of heterogeneity between the generations was found. The heterogeneity was traced to the major non-Mendelian component (which accounted for 0.01% of the variance in parents and over 40% in offspring) rather than to the multifactorial one. These results would suggest that a simple recessive gene mixed model may not be sufficient to explain the familial distribution of the BMI. Several factors which may have contributed to these results include temporal trends and surrogate effects such as those related to variation in body composition and energy balance components.
