In cultured mammalian cells, foreign DNA can be integrated into the host genome. Foreign DNA is frequently de novo methylated in specific patterns with successive cell generations. The sequence-specific methylation of promoter sequences in integrated foreign DNA is associated with the long-term inactivation of eukaryotic genes. We have now extended these experiments to studies on transgenic mice. As in previous work, a construct (pAd2E2AL-CAT) has been used which consists of the late E2A promoter of adenovirus type 2 (Ad2) DNA fused to the prokaryotic gene for chloramphenicol acetyltransferase (CAT). This construct has been integrated in the non-methylated in the 5'-CCGG-3' premethylated form in the genomes of transgenic mice. DNA from various organs was analyzed by HpaII/MspI cleavage to assess the state of methylation in 5'-CCGG-3' sequences. The results demonstrate that the transgenic construct is in general stable. Non-methylated constructs have remained partly non-methylated for four generations or can become de novo methylated at all or most 5'-CCGG-3' sequences in the founder animal. Preimposed patterns of 5'-CCGG-3' methylation have been preserved for up to four generations beyond the founder animal. In the testes of two different founder animals and two F1 males, the transgenic DNA has become demethylated by an unknown mechanism. In all other organs, the transgenic DNA preserves the preimposed 5'-CCGG-3' methylation pattern. In the experiments performed so far we have not observed differences in the transmission of methylation patterns depending on whether the transgene has been maternally or paternally inherited. The 5'-CCGG-3' premethylated transgene does not catalyze CAT activity in several organs, except in one example of the testes of an animal in which the transgenic construct has become demethylated. In contrast, when the nonmethylated construct has been integrated and remained largely non-methylated, CAT activity has been detected in extracts from some of the organs.  相似文献

Triglyceride-rich lipoproteins derived from ten normo- and hyperlipidemic apoE-2 homozygotes were analyzed for their composition, beta-VLDL content, and their ability to induce cholesteryl ester storage in macrophages. In six of these probands apoE sequence analysis revealed that the cysteine residues were at positions 112 and 158 of the amino acid sequence (Rall et al. 1983. J. Clin. Invest. 71: 1023-1031). ApoE-2 of these six and the other four patients was further analyzed by SDS electrophoresis to exclude the presence of apoE-2* (Rall et al. 1982. Proc. Natl. Acad. Sci. USA. 79: 4696-4700). The relative serum concentrations of free and esterified cholesterol transported in the d less than 1.006 g/ml and d 1.006-1.019 g/ml lipoproteins of the apoE-2 homozygotes was significantly higher as compared to controls. Compositional analysis of these lipoproteins revealed a relative reduction of triglycerides and a relative increase of cholesteryl esters as compared to controls. In most patients, with increasing serum triglyceride levels the cholesteryl ester concentration increased in d less than 1.006 g/ml and d 1.006-1.019 g/ml lipoproteins. However, in three patients with a low content of beta-VLDL, the increase in the d less than 1.006 g/ml fraction cholesterol was mostly due to free cholesterol and not due to cholesteryl esters. The degree of the macrophage cholesteryl ester accumulation induced by d less than 1.006 g/ml lipoproteins was mostly dependent on the concentration of the beta-migrating fraction (beta-VLDL). The amount of beta-VLDL and pre-beta-VLDL contained in the d less than 1.006 g/ml fraction was determined densitometrically after electrophoretic separation. It could be demonstrated that the beta-VLDL content in the d less than 1.006 g/ml fraction of the apoE-2 homozygous patients was largely independent of serum triglyceride and serum cholesterol levels. When macrophages were incubated with the IDL fraction (d 1.006-1.019 g/ml) from the apoE-2 patients, no significant increase in cellular cholesteryl esters above control levels was observed. Studies with purified lipoprotein lipase (LPL) and hepatic triglyceride lipase (HTGL) clearly revealed that both enzymes interacted with apoE-2 VLDL (binding, hydrolysis) to a lesser degree compared to control preparations. However, the apoE-2 VLDL preparations containing a low content of beta-VLDL were better substrates for LPL and HTGL than those containing a high beta-VLDL content. It is concluded from our studies that the plasma beta-VLDL content in apoE-2 homozygotes is a major determinant for cholesteryl ester accumulation in macrophages.(ABSTRACT TRUNCATED AT 400 WORDS)  相似文献

Biosynthesis of sucrosyl-oligosaccharides (raffinose, stachyose) was traced in source leaves of Cucumis melo after 14C-photoassimilation. The main carbon compound exported was 14C-labeled stachyose. No oligosaccharide synthesis was detected in young, importing leaves. Mesophyll protoplasts, isolated from mature leaves which had previously photosynthesized 14CO2, did not contain 14C-oligosaccharides but contained [14C]-sucrose and 14C-hexoses. Isolated minor-vein-enriched fractions from the same leaves, however, showed nearly 30% of the 14C of the neutral fraction to be in oligosaccharides. Isolated, viable mesophyll protoplasts incubated with NaH14CO3 also failed to incorporate radioactivity into oligosaccharides, although sucrose and galactinol synthesis was unimpaired. Galactinolsynthase activity in leaf extracts and in mesophyll protoplasts was 16.8 mol·h-1·mg-1 protein and 13.8 mol·h-1·mg-1 protein, respectively. Galactosyltransferase (EC 2.4.1.67), which synthesizes stachyose from raffinose and galactinol, had an activity of 50 nmol·h-1·mg-1 protein in leaf extracts and was also present in the minor-vein-enriched fraction, but could not be detected in mesophyll protoplast lysates. The results indicate that mesophyll cells may not be the site of stachyose synthesis although precursor compounds like sucrose and galactinol are synthesized there.Abbreviation HPLC high-performance liquid chromatography  相似文献

Zusammenfassung Bei Erstbelichtung des Senfkeimlings mit Dauer-Dunkelrot tritt der P730-abhängige Anstieg der Phenylalanindesaminase-Aktivität erst mit einer lag-Phase von 1,5 Std ein (Abb. 3, 4 unten). Bei einer Zweitbelichtung (Programm: Erstbelichtung — längere Dunkelperiode — Zweitbelichtung) fehlt die lag-Phase (Abb. 4, oben). Die Enzymaktivität steigt sofort linear an. Da der Anstieg der Enzymaktivität wahrscheinlich auf eine de novo Synthese von RNS und Enzymprotein zurückzuführen ist (Tabelle), so erscheint der Schluß berechtigt, daß P730 sehr rasch eine differentielle Genaktivierung mit anschließender Enzymsynthese bewirken kann, falls die Gene der Aktivierung durch P730 zugänglich sind. Die relativ lange lag-Phase nach Einsetzen der Erstbelichtung benötigt das P730 offenbar dazu, die potentiell aktiven Gene (P730) für das P730 zugänglich zu machen. Das Problem der primären lag-Phase ist in einer vorangegangenen Arbeit zur P730-abhängigen Anthocyansynthese ausführlich diskutiert worden (vgl. Lange, Bienger und Mohr, 1967).
Phytochrome-mediated enzyme formation (Phenylalanine deaminase) as a rapid process
Summary In previous papers we have reported (Mohr and Durst, 1966a, b) that synthesis of phenylalanine deaminase (EC 4.3.1.5), an important enzyme of phenolic metabolism, can be stimulated by the physiologically active phytochrome (=P730) in the mustard seedling. The data of the present paper suggest that induction of this enzyme is a rapid process if the gene in question is easily accessible for the activating action of P730.The seedlings were irradiated with continuous standard far-red light. Longtime irradiation with far-red will maintain a low but virtually constant level of P730 in the seedling over an extended period of time. At the moment when the far-red light is turned off the action of P730 will virtually cease. — Fig. 3 and Fig. 4, lower part, show the kinetics of enzyme induction by P730 in an etiolated seedling. The initial (or primary) lag-phase after the onset of far-red is 1.5 hours. If, however, a seedling which has been pre-irradiated with 12 hours of far-red is kept in darkness for 6 hours and is then re-irradiated with far-red no lag-phase for the action of the second irradiation can be found. Enzyme activity increases immediately after the onset of far-red. Since the action of the second irradiation as measured by increase of enzyme activity can be inhibited by relatively low doses of Puromycin and Cycloheximide (table) we conclude that the re-appearance of P730 leads to de novo synthesis of enzyme protein. — Application of Actinomycin D (10 g/ml) only partially inhibits the action of the second irradiation as measured by increase of enzyme activity. This finding was to be expected. In preceding papers (e.g. Mohr and Bienger, 1967) it has been concluded that genes which have once been activated by P730 remain less sensitive towards Actinomycin D even when P730 has disappeared. Taking into account all available data the conclusion seems to be justified that the induction of enzyme synthesis by P730 (i.e. differential gene activation followed by enzyme synthesis) is a rapid process if the genes are accessible for the action of P730. The relatively long initial lag-phase (1.5 hours) is needed to make the potentially active genes (P730) accessible for the action of P730. The problem of how the initial lag-phase can be understood has been dealt with more in detail in a previous paper on phytochrome-mediated anthocyanin synthesis (Lange, Bienger and Mohr, 1967).

Zusammenfassung Pinguicula lusitanica wurde in vitro auf stickstoff- und phorphorfreiem Mineralsalzagar kultiviert; jede Pflanze stand für sich in einem Erlenmeyerkolben.Als die Pflanzen 8 Wochen alt waren, wurden 20 Exemplare innerhalb von 5 Wochen viermal mit Pinuspollen gefüttet. 20 gleichgroße und gleichalte dienten als Kontrollen.Durch die Fütterung steigerte sich die Blattzahl und der Durchmesser der Blattrosette, die Blätter wurden intensiver grün und alterten langsamer.Vor allem wurde durch die Pollenfütterung die Blütenbildung ausgelöst. Schon nach der 2. Fütterung traten die ersten Knospen auf, eine Woche darauf blühten bereits 70% und noch vor der letzten Fütterung alle gefütterten Pflanzen, von den ungefütterten dagegen keine einzige. In dem anschließenden halben Jahr entwickelten die 20 gefütterten Pflanzen im ganzen 127 Blüten; die größte Blütenzahl einer Einzelpflanze was 14. Die nichtgefütterten Pflanzen waren auch jetzt noch rein vegetativ.
Flowering of in vitro cultures of Pinguicula lusitanica after feeding with pinus pollen
Summary Plants of Pinguicula lusitanica were grown in individual Erlenmeyer flasks on an inorganic agar medium containing no nitrogen or phosphorus. After 8 weeks of culture, twenty of the plants were fed Pinus pollen 4 times over a period of 5 weeks.As a result of the feeding, the number of leaves as well as the diameters of the rosettes were increased. The leaves became turned a deeper green and aged more slowly.The most spectacular effect of the pollen feeding was an initiation of flowering. The first buds were already visible after the second feeding. All of the treated plants flowered before the last feeding, whereas none of the untreated plants flowered. During the following 6 months, the treated plants developed 127 flowers, the largest number on a single specimen being 14. Even after this period of time the untreated plants remained vegetative.

Nonspiking interneurons were investigated in a tethered, walking insect, Carausius morosus, that was able to freely perform walking movements. Experiments were carried out with animals walking on a lightweight, double-wheel treadmill. Although the animal was opened dorsally, the walking system was left intact. Intracellular recordings were obtained from the dorsal posterior neuropil of the mesothoracic ganglion. Nonspiking inter-neurons, in which modulations of the membrane potential were correlated with the walking rhythm, were described physiologically and stained with Lucifer Yellow. Interneurons are demonstrated in which membrane potential oscillations mirror the leg position or show correlation with the motoneuronal activity of the protractor and retractor coxae muscles during walking. Other interneurons showed distinct hyperpolarizations at certain important trigger points in the step cycle, for example, at the extreme posterior position. Through electrical stimulation of single, nonspiking interneurons during walking, the motoneuronal activity in two antagonistic muscles—protractor and retractor coxae—could be reversed and even the movement of the ipsilateral leg could be influenced. The nonspiking interneurons described appear to be important premotor elements involved in walking. They receive, integrate, and process information from different leg proprioceptors and drive groups of leg motoneurons during walking.  相似文献

A detailed kinetic study of the inhibitory effects ofl- andd-enantiomers of cysteate, cysteine sulphinate, homocysteine sulphinate, homocysteate, and S-sulpho-cysteine on the neuronal, astroglial and synaptosomal high-affinity glutamate transport system was undertaken.d-[3H] Aspartate was used as the transport substrate. Kinetic characterisation of uptake in the absence of sulphur compounds confirmed the high-affinity nature of the transport systems, the Michaelis constant (K m) ford-aspartate uptake being 6 M, 21 M and 84 M, respectively, in rat brain cortical synaptosomes and primary cultures of mouse cerebellar granule cells and cortical astrocytes. In those cases where significant effects could be demonstrated, the nature of the inhibition was competitive irrespective of the neuronal versus glial systems. The rank order of inhibition was essentially similar in synaptosomes, neurons and astrocytes. Potent inhibition (K iK m) of transport in each system was exhibited byl-cysteate, andl- andd-cysteine sulphinate whereas substantially weaker inhibitory effects (K i>10–1000 times the appropriateK m value) were exhibited by the remaining sulphur amino acids. In general, inhibition: (i) was markedly stereospecific in favor of thel-enantiomers (except for cysteine sulphinate) and (ii) was found to decrease with increasing chain length. Computer-assisted molecular modelling studies, in which volume contour maps of the sulphur compounds were superimposed on those ofd-aspartate andl-glutamate, demonstrated an order of inhibitory potency which was, qualitatively, in agreement with that obtained quantitatively by in vitro kinetic studies.Special issue dedicated to Dr. Elling Kvamme  相似文献
