Chromatin of chicken erythrocyte nuclei was extracted by digestion with micrococcal nuclease. The length distribution of the soluble chromatin was determined by gel electrophoresis and electron microscopy. These results were fitted with a theoretical distribution which was an outcome of the domain model proposed by Igo-Kemenes and Zachau (Igo-Kemenes, T. and H.G. Zachau (1977) Cold Spring Harbour Symp. Quant. Biol. 42, 109–118). A domain length of 45 kbp was obtained.  相似文献

Arsenic is a ubiquitously found metalloid that commonly contaminates drinking water and agricultural food. To understand the ecotoxicological effects of arsenic in environment, it is essential to ameliorate the deleterious effects on human and animal health, particularly on the immune response. We investigated the effects of inorganic arsenic (iAs) on the immune response of chicken splenocytes. Both 1 and 10 mM concentrations of sodium arsenite treatment significantly reduced (P < 0.001) splenocyte proliferation and phagocytic activity compared to concanavalin A (ConA) stimulated cells at 24, 48 and 72 h of incubation. Nitrous oxide (NO) production was significantly higher (P < 0.001) at 24 h and subsequently declined in the higher dose group, while there was a gradual decline from 24 to 72 h in the lower dose group. Comparison of two different concentration of arsenic treatment also revealed time dependent differences. Relative quantification of expression of IFN?? and IL2 revealed that both genes were significantly down regulated (P < 0.001) at both concentrations at each time point. iNOS gene was rapidly down regulated in splenocytes at 24 h at the high doses of As treated splenocyte, a gradual decreasing trend at low doses. Down regulation of IL-2 gene expression in response to As was further evidenced by a significant reduction (P < 0.001) in the release of IL-2 into the splenocyte culture medium. We suggest that arsenic, a potent immunotoxic agent, modulates non-specific immune responses and alters the expression of cytokines in a dose and time dependent manner.  相似文献

The respiratory quinone composition of the parasitic protozoa Leishmania donovani promastigote was investigated. 1′-oxomenaquinone-7, a chlorobiumquinone was found to be the major isoprenoid quinone. Substantial level of ubiquinone-9 was also present. Isolation and identification of the quinone from the purified plasma membrane yielded mainly 1′-oxomenaquinone-7 and ubiquinone-9; menaquinone was not detected. Membrane bound 1′-oxomenaquinone-7 could be destroyed by near-ultraviolet irradiation, with a concomitant loss or stimulation of plasma membrane electron transport activities. The abilities of different quinones to restore α-lipoic acid and ferricyanide reductase activity in near UV-irradiated cell preparations were compared. The order was; conjugate of chlorobiumquinone and sphingosine base ? conjugate of 2-methyl-3-(1′-oxooctadecyl)-1,4-napthoquinone and octadecylamine >> chlorobiumquinone ? 2-methyl-3-(1′-oxooctadecyl)-1,4-napthoquinone > menaquinone-4 ? ubiquinone-10. After irradiation with near-UV light, transmembrane α-lipoic acid reduction was inhibited, while transmembrane ferricyanide reduction was stimulated. The result obtained indicates that chlorobiumquinone mediates the plasma membrane electron transport between cytosolic reductant and oxygen as well as α-lipoic acid. UV-inactivation of chlorobiumquinone shuts down the plasma membrane oxygen uptake and diverts the electron flux towards ferricyanide reduction via ubiquinone-9. Chlorobiumquinone is the only example of a polyisoprenoid quinone containing a side chain carbonyl group from photosynthetic green-sulphur bacteria. Recent work has revealed numerous genes of trypanosomatid sharing common ancestry with plants and/or bacteria. These observations pose some fascinating questions about the evolutionary biology of this important group of parasitic protozoa.  相似文献

Ten palm leaf impressions are documented from the latest Maastrichtian (late Cretaceous) to early Danian (earliest Paleocene) sediments (K-Pg, c. 66–64 Ma) of the Mandla Lobe of the Deccan Inter-trappean Beds, Madhya Pradesh, central India. The palmate leaf shape along with a definite well-preserved costa support their placement in the subfamily Coryphoideae of the family Arecaceae. We place all recovered palm leaf specimens in the fossil genus Sabalites, report seven species of coryphoid palms and describe two new species namely, Sabalities umariaensis sp. nov. and Sabalites ghughuaensis sp. nov. The fossils indicate that coryphoid palms were highly diverse in central India by the latest Cretaceous. These and earlier reported coryphoid palm fossils from the same locality indicate that they experienced a warm and humid tropical environment during the time of deposition. These discoveries confirm the presence of a diversity of Coryphoideae in Gondwana prior to the India-Eurasia collision and provide information about coryphoid biogeographical history over geological time. Based on megafossil remains, we trace coryphoid palm migration pathways from India to mainland Southeast (SE) Asia and other parts of Asia after the docking of the Indian subcontinent with Eurasia early in the Paleogene.  相似文献
