Anti-trichomonal IgA antibodies were estimated by enzyme-linked immunosorbent assay (ELISA) in serum and vaginal secretions of 25 symptomatic and 25 asymptomaticTrichomonas vaginalis positive patients before and after treatment and in 25 age-matched controls. Significantly higher levels of antitrichomonad IgA antibodies were found inT. vaginalis positive patients when compared to control subjects, especially in vaginal secretions. In addition, a significant decrease in these antibodies was observed after treatment, which was more pronounced in vaginal secretions. It seems that anti-trichomonal IgA antibodies in serum and more so in vaginal secretions are directly related to and specific to the presence ofT. vaginalis in the urogenital tract.  相似文献

Ribosome biogenesis is a complex process that is facilitated by a large number of assembly factors. In this issue, Andrade et al ( 2018 ) provide evidence that a widely conserved RNA chaperone, Hfq, acts as a ribosomal assembly factor in bacteria. Hfq is known to support regulation of stress response genes by small RNAs. Andrade et al ( 2018 ) show that the absence of Hfq results in higher levels of immature 30S ribosomes and error‐prone translation, suggesting that Hfq globally affects the quality of protein synthesis when bacteria are under stress.  相似文献

In this study, zymographic analysis for xylanase and pectinase enzymes has been carried out using agrowaste residues, wheat bran and citrus peel as well as their extracts. Isozymic forms of xylanase as well as pectinase enzyme displayed comparable zymographic bands onto agar petriplates containing either commercial substrates (xylan and pectin), agrowaste-based substrates (wheat bran and citrus peel), or polysaccharides extracted from these agrowastes (crude xylan and pectin extracted from wheat bran and citrus peel, respectively), indicating the fact that agro residues and their extracts can be utilized as a substitute of cost-intensive commercial substrates, xylan and pectin for zymographic analysis. This is the first report revealing the zymographic analysis of xylano–pectinolytic enzymes using agro-based solid residues particles or polysaccharides extracted from agro-based residues.  相似文献

Results of experiments to determine the sensitivity of Channa punciatus to different levels of sodium chloride (NaCl) (1.2, 3.2 and 6.2g 1−1) are reported. It was found that C. punctatus which is otherwise considered a hardy fish is more sensitive to higher levels of sodium chloride stress. Thus 10% of the LC50 96 h value (1.2 g 1−1 NaCl) was found stressful as evidenced by loss in body weight, while 25 and 50% of the LC50 96 h value (3.2 and 6.2 g 1−1 NaCl) even resulted in mortality. Haematological and associated haematopoietic investigation confirmed stress symptoms in all the parameters tested. Thus microcytic hypochromic anemia together with leucopenia was indicated at the two higher sublethal levels, i.e., 25% and 50% of LC50 96 h value. Differential leucocyte count showed that leucopenia is mainly contributed by thrombocytic and neutrophilic decrease. Other cell types, viz., eosinophils, basophils, large and small lymphocytes, showed a relatively significant (P < 0.01) increase in number.
Haematopoietic studies revealed that the two penultimate stages in both RBC and neutrophil development closely follow the trend of their counterparts in the peripheral blood. However, the stem cells of both these cell types recorded significant increase under sodium chloride stress, which indicated homeostatic response. Biochemical studies revealed a depletion in blood glucose level as well as in liver and muscle glycogen reserves.
Restoration to normal control conditions after 6 weeks of exposure to different levels of sodium chloride stress resulted in a cessation of mortality (if any), gain in body weight and more or less normality in all the haematological parameters within a period of 2 weeks.  相似文献

The chloroplast homolog of the 54 kDa subunit of signal recognition particle is required for the in vitro targeting of chlorophyll a/b binding proteins (LHCP) to the thylakoid membrane. To explore the function of cpSRP54 in vivo, plants that are mutated in cpSRP54 function were generated. Dominant negative forms of cpSRP54 altered in single amino acids within the conserved guanine nucleotide binding domain were expressed in Arabidopsis. Transformed plants contained less than 30% of the wild-type level of cpSRP54 protein. As a consequence of the reduced cpSRP54 protein content, the first emerging leaves were yellow and contained immature chloroplasts. Although the chlorophyll (chl) content of the leaves was reduced by 75%, the chl a/b ratio was unaffected, indicating a role of cpSRP54 in the biogenesis of proteins besides LHCP. Many chloroplast proteins were less abundant in the first emerging leaves, including non-pigmented proteins, thylakoid proteins known to be targeted by alternative pathways, and soluble proteins. These observations indicate that the cpSRP54 mutation also has a pleiotropic effect on chloroplast biogenesis. Whereas the level of cpSRP54 remained low as the plants aged, leaves emerging subsequently had a wild-type appearance, suggesting that the adult plants compensated for the reduction in cpSRP54 protein.  相似文献

BackgroundTill to date whether adiponectin AdipoQ gene variation (rs 1501299) is associated with cardiovascular disease, still remains controversial. Therefore, we aimed to relate the SNP (rs1501299) of adiponectin gene and oxidative stress in context to CVD in Punjabi women of North West India.MethodsIn the present case-control study menopausal women with CVD as cases (n=265) and menopausal women without CVD as controls (n=258) were recruited. Genotyping of rs1501299 single nucleotide polymorphism of adiponectin gene was carried out by RFLP-PCR analysis. Biochemical parameters were analyzed according to the standard procedures.ResultsDistribution of homozygous TT genotype of normolipidemic (p=0.001) and hyperlipidemic (p=0.001) women with CVD was significantly more frequent as compared to women without CVD. rs1501299 T allele carriers with CVD also showed significant (p=0.001) higher frequency distribution as compared to women without CVD. Under recessive model of inheritance TT mutant type homozygotes conferred ~9 fold higher risk [p=0.001; OR= 9.60 (2.92-31.58)] towards CVD susceptibility for MDA>1.50; ~11 fold higher risk [p=0.007; OR= 11.11 (1.49-82.83)] towards CVD for LDL carbonyl protein>15.04 and ~9 fold higher risk [p=0.001; OR= 9.75 (2.30-41.22)] towards CVD susceptibility for SOD≤5.55. Under logistic regression analysis oxidative stress and TT genotype were significantly correlated with CVD.ConclusionsOur study revealed significant association of AdipoQ (rs1501299) gene polymorphism and oxidative stress with cardiovascular disease in Punjabi women of North West India. However, additional studies are required to support these findings.  相似文献

BackgroundKit lot change in clinical biochemistry labs leads to variations in patient results. This study planned to identify variations during 60 reagent lot changes in our laboratory during the period from June 2018 to May 2019.MethodsA statistical analysis was performed to identify the difference between patient samples results variations and QC results. The long term drift was analyzed using a regression test.ResultsThere was a significant difference between the patient and QC results in 16.7% of reagent lot changes. Moreover, the extent of variation in QC results was 3.3%. No long-term drift was seen in three analytes which were studied using regression analysis.ConclusionsOur results showed that, during reagent kit lot change, along with QC material, the patient samples should also be run in order to identify the variation. However, this practice is presently ignored by most of the laboratories. There was no accumulated effect in our laboratory due to reagent kit lot change.  相似文献
