One group of mantellid frogs from Madagascar (subgenus Pandanusicola of Guibemantis) includes species that complete larval development in the water-filled leaf axils of rainforest plants. This group consists of six described species: G. albolineatus, G. bicalcaratus, G. flavobrunneus, G. liber, G. pulcher, and G. punctatus. We sequenced the 12S and 16S mitochondrial rRNA genes ( approximately 1.8 kb) from multiple specimens (35 total) of all six species to assess phylogenetic relationships within this group. All reconstructions strongly supported G. liber as part of the Pandanusicola clade, even though this species does not breed in plant leaf axils. This result confirms a striking reversal of reproductive specialization. However, all analyses also indicated that specimens assigned to G. liber include genetically distinct allopatric forms that do not form a monophyletic group. Most other taxa that were adequately sampled (G. bicalcaratus, G. flavobrunneus, and G. pulcher) likewise consist of several genetically distinct lineages that do not form monophyletic groups. These results suggest that many of the recognized species in this group are complexes of cryptic species.  相似文献

Summary X-linked retinitis pigmentosa (XLRP) is a series of hereditary dystrophic diseases of the retina that occur in three clinically distinguishable variants: the classic form (McK-31360), a type known as choroidoretinal dystrophy (McK-30330), and a variant with golden-metallic or tapetal reflex in the heterozygote (McK30320). Controversy exists as to whether these phenotypic differences are due to clinical variability in disease expression, heterogeneity in disease alleles at a single locus, or a multiplicity of loci for XLRP. We have studied a single large kindred segregating for XLRP with the metallic fundus reflex in the heterozygote with restriction fragment length polymorphisms (RFLPs) from the short arm of the human X chromosome, and found measurable linkage to DXS7 (=12.5 cMorgans at LOD=2.5), the same RFLP previously shown by others to be tightly linked to the other forms of XLRP at =3cM. Although these estimates appeared to be different, each fell just within the 95% probability interval of the other and, therefore, were insufficient to prove or disprove that the metallic sheen form of XLRP is allelic with other forms of XLRP. Additional RFLPs at the DXS43 and the ornithine transcarbamoylase loci provided three-point crosses for determining the relative positions of DXS7 and XLRP, and supported an order that placed this form of XLRP distal to DXS7 on the Xp. Until the question of genetic heterogeneity is resolved, careful phenotypic characterization of the clinical type of XLRP present in families being used for linkage analyses is advisable.Presented in part at the American Society of Human Genetics meeting, Toronto, Canada, November 1, 1984  相似文献

This study sought to characterize muscle loading and fatigue during static shoulder abductions with varying force. In a supine posture, participants maintained fixed shoulder abductions against a time-varying external resistance, generated by a dynamometer-spring mechanism. Patterns (cumulative distribution) of the external resistance were varied by selecting different 10th and 90th percentiles of the distribution. Dynamometer angular velocities were also varied, to reflect different rates of cyclic muscle contraction. The degree of local fatigue development was assessed by common measures, including endurance time, strength reduction, and perceived discomfort. Myoelectric (EMG) signals were continuously obtained from the middle deltoid muscle throughout experimental exercise (60min max). Changes in EMG root-mean-square (RMS) and spectral measures (derived from 1-s windows at peaks in the cyclic contractions) were used as manifestations of muscle fatigue. For each minute, the RMS signal was further reduced using two methods, the cumulative probability distribution of EMG (CPDE) and exposure variation analysis (EVA). The former resulted in three percentile values (10th, 50th, and 90th), whereas the latter method resulted in 10 different measures (grouped by EMG activity level and duration). A main finding of the study was the applicability of several common fatigue indicators for these cyclic, repetitive exertions. Overall, the use of CPDE and EVA to characterize task differences and predict muscle fatigue was found to have limited value.  相似文献

Sendai virus envelopes have been a useful tool in studying the mechanism of membrane-membrane fusion and have served as a vehicle for introducing foreign molecules (e.g., membrane proteins) into recipient cells. Reconstituted Sendai virus envelopes are routinely obtained following solubilization of virus particles with Triton X-100. This detergent has a low critical micellar concentration which precludes it from being the best detergent of choice in reconstitution studies. Nevertheless, it has remained in use since other detergents such as sodium deoxycholate and sodium cholate rendered the resultant vesicles inactive. Triton X-100 may be suboptimal for studies of some proteins that need be coreconstituted with the viral envelopes. Thus, alternative advantageous detergents, which retain the envelope fusogenic activity, have been sought. In this study we show that the synthetic detergent 3-[(3-cholamidopropyl)dimethylammonio]-1-propanesulfonate (Chaps) effectively solubilizes the Sendai virions, and that the vesicles formed by simple reconstitution protocols appear structurally and biochemically similar to those obtained with Triton X-100. The resultant vesicles retain functional integrity as assessed in both fusion and hemolysis assays. This protocol seems to be useful in sendai envelope-mediated reimplantation of Fc epsilon receptors into the plasma membranes of rat basophilic leukemia cells.  相似文献

A lambda gt11 expression library was screened with a polyclonal antiserum directed against Wla Wolfgram protein which corresponds to the 2',3'-cyclic nucleotide 3'-phosphohydrolase. This antiserum also recognizes epitopes of the high protein subunit of neurofilaments (NF-H). An NF-H cDNA clone (pNF-H1: 1.7 kb) was isolated and characterized. Using pNF-H1 as a probe, a second cDNA clone (pNF-H2: 3 kb) was isolated from the lambda gt11 library. The two clones were sequenced and pNF-H2 was found to encode 80% of rat NF-H protein (coil-2 and C-terminal region). The C-terminal region contains an unusual sequence with stretches of repeats of Lys-Ser-Pro which represent possible phosphorylation sites. Specific localization in neurons of the corresponding mRNA was demonstrated by in situ hybridization using the pNF-H1 as a probe.  相似文献

The endemic Seychelles treefrog, Tachycnemis seychellensis (Duméril & Bibron), is restricted to four of the granitic islands of the Seychelles: La Digue, Mahe, Praslin and Silhouette. Megalixalus infrarufus Gunther 1869 is a junior synonym of Eucnemis ( Tachycnemis ) seychellensis Dumeril Bibron, 1841. Significant variation in colour and morphometric characteristics exists within and between island populations. The patterns of geographic variation revealed support the hypothesis that the distribution of Tachycnemis seychellensis in the granitic Seychelles reflects vicariance through fragmentation of the Seychelles Microcontinent 10,000 years B. P. by marine transgression. However, the possibility of low rates of postfragmentation dispersal between islands cannot be ruled out. The close relationship of the nearby Mahe and Silhouette populations probably reflects prefragmentation gene flow over relatively short distances and postfragmentation stasis due to large population size and similar environments. The small body size and colour similarities of the Praslin and La Digue populations may result from prefragmentation gene flow between these close populations, but the relatively great differences in morphometric traits suggest rapid divergence in isolation perhaps as a result of genetic drift and strong selection. It is argued that the four island populations represent a single species and that subspecies should not be named.  相似文献

Specimens of the endemic New Zealand frog Leiopelma hochstetteri from Tapu on North Island were found to have six, nine or ten supernumerary chromosomes in their karyotypes. In comparison with previously published data, these results further indicate probable geographic variation in supernumerary chromosome number between populations. Increased numbers of supernumeraries in these frogs is correlated with apparent decrease of centromeric heterochromatin in the five large metacentric chromosomes of the karyotype, as detected by C-banding. Meiosis was abnormal in a male with a high number of supernumeraries. In lampbrush preparations from a single female with one supernumerary univalent, the supernumerary often had a denser, beaded appearance in comparison with the regular bivalents. Evidence is consistent with the notion that these supernumerary chromosomes may have arisen from centromeric fragments.  相似文献
