Previous work showed that treatment of plateau-phase Chinese hamster ovary cells with the radiomimetic double-strand cleaving agent bleomycin induced very small deletions as well as interchromosomal reciprocal translocations, both of which could be ascribed to errors in end joining of DNA double-strand breaks. In an attempt to assess the possible role of TP53 in suppressing such repair errors, bleomycin-induced mutagenesis at the HPRT locus was examined in immortalized 184B5 human mammary epithelial cells (TP53(+)), and in a TP53-defective derivative, 184B5-E6tfxc6. For both cell lines, the most frequent bleomycin-induced mutations were base substitutions, with no apparent targeting to major bleomycin lesions. However, both lines also sustained single-base deletions that were targeted to expected sites of double-strand breaks, suggesting that they arose by end-joining repair of the breaks. Surprisingly, only a few large deletions or rearrangements, and no interchromosomal events involving the HPRT locus were detected among the mutants. The results suggest that in both cell lines, errors in double-strand break repair resulting in heritable large deletions and rearrangements are rare. Spectral karyotyping of bleomycin-treated 184B5 cells showed that a significant number of translocations were present shortly after bleomycin exposure, but their frequency decreased upon continued culture of the cells. Thus, for these cells, the lack of induced interchromosomal rearrangements can be explained in part by selection against such events as the cells proliferate. 相似文献

The maximal endothelial dependent relaxation of isolated aortic rings to cumulative doses of acetylcholine was significantly decreased in the Cyclosporine-A (CSA, 20 mg kg(-1) day(-1)) treated animals compared to olive oil (CSA vehicle) treated control. Administration of antihypertensive drugs like diltiazem, enalapril or propranolol to CSA treated animals augmented the endothelial damage induced by CSA. These drugs also increased the bioavailability of CSA. However, administration of losartan to CSA treated animals produced a significant increase in endothelial dependent relaxation as compared to CSA treated control but did not affect the bioavailability of CSA significantly. The objective of this work was to prepare and evaluate ketorolac tromethamine-loaded albumin microspheres using a factorial
design. Albumin microspheres were prepared by emulsion cross-linking method. Selected formulations were characterized for
their entrapment efficiency, particle size, surface morphology, and release behavior. Analysis of variance (ANOVA) for entrapment
efficiency indicated that entrapment efficiency is best fitted to a response surface linear model. From the statistical analysis
it was observed that as the drug:polymer (D∶P) ratio and volume of glutaraldehyde increased, there was a significant increase
in the encapsulation efficiency. Scanning electron microscopy of the microspheres revealed a spherical, nonporous and uniform
appearance, with a smooth surface. Based on the entrapment efficiency and physical appearance, 9 formulations were selected
for release study. The maximum particle size observed was below 40 μm. The release pattern was biphasic, characterized by
an initial burst effect followed by a slow release. All selected microspheres, except those having less polymer proportion
(D∶P ratio is 1∶1), exhibited a prolonged release for almost 24 hours. On comparingr
2 values for Higuchi and Peppas kinetic models, different batches of microspheres showed Fickian, non-Fickian, and diffusion
kinetics. The release mechanism was regulated by D∶P ratio and amount of cross-linking agent. From the experimental data obtained
with respect to particle size and extent of drug relaase, it could be concluded that the prepared microspheres are useful
for once-a-day intramuscular administration of ketorolac tromethamine.

X-ray crystallography, although a powerful technique for determining the three-dimensional structure of proteins, poses inherent problems in assigning the primary structure in residues Asp/Asn and Glu/Gln since these cannot be distinguished decisively in the electron density maps. In our recently published X-ray crystal structure of the Sclerotium rolfsii lectin (SRL) at 1.1 A resolution, amino acid sequence was initially deduced from the electron density map and residues Asp/Asn and Glu/Gln were assigned by considering their hydrogen bonding potential within their structural neighborhood. Attempts to verify the sequence by Edman sequencing were not successful as the N terminus of the protein was blocked. Mass spectrometry was applied to verify and resolve the ambiguities in the SRL X-ray crystal structure deduced sequence. From the Matrix assisted laser desorption/ionization time-of-flight-mass spectrometry (MALDI TOF-MS) and liquid chromatography-electrospray ionization-tandem mass spectrometry (LC-ESI-MS/MS) analysis of tryptic and chymotryptic peptides of SRL, we could confirm and correct the sequence at five locations with respect to Asp/Asn and Glu/Gln. Analysis data also confirmed the positions of Leu/Ile, Gln/Lys residues and the sequence covering 118 of the total 141 residues accounting to 83.68% of the earlier deduced sequence of SRL. 相似文献

The present work embodies epidermal structure, structure and ontogeny of stomata in five genera embracing sixteen species of the Capparidaceae namely Cleome (8 species) Capparis (5 species), Cadaba (1 species), Crataeva (1 species) and Maerua (1 species). The epidermal cells are polygonal, isodiametric or elongated arranged irregularly, with evenly or unevenly thickened, sinuous, straight or arched anticlinal walls. Two main types of trichomes: glandular (four types) and eglandular (five types) are noticed. The stomatal types include cyclocytic, triacytic, staurocytic, tetracytic, anomocytic, anisocytic, paracytic and with a single subsidiary cell. The ontogeny of stomata with a single subsidiary cell is perigenous or mesoperigenous, of paracytic mesoperigenous or mesogenous, of anisocytic is mesoperigenous or mesogenous, while that of the other types is perigenous. Abnormalities observed are: single guard cell; aborted guard cells; complete or incomplete division of guard cells; contiguous stomata; giant stomata and cytoplasmic connections.
