Clustering of pathogens in the environment leads to hot spots of diseases at local, regional, national, and international levels. Scotland contains regional hot spots of Johne's disease (caused by Mycobacterium avium subsp. paratuberculosis) in rabbits, and there is increasing evidence of a link between paratuberculosis infections in rabbits and cattle. The spatial and temporal dynamics of paratuberculosis in rabbits within a hot spot region were studied with the overall aim of determining environmental patterns of infection and thus the risk of interspecies transmission to livestock. The specific aims were to determine if prevalence of paratuberculosis in rabbits varies temporally between seasons and whether the heterogeneous spatial environmental distribution of M. avium subsp. paratuberculosis on a large scale (i.e., regional hot spots) is replicated at finer resolutions within a hot spot. The overall prevalence of M. avium subsp. paratuberculosis in rabbits was 39.7%; the temporal distribution of infection in rabbits followed a cyclical pattern, with a peak in spring of 55.4% and a low in summer of 19.4%. Spatially, M. avium subsp. paratuberculosis-infected rabbits and, thus, the risk of interspecies transmission were highly clustered in the environment. However, this is mostly due to the clustered distribution of rabbits. The patterns of M. avium subsp. paratuberculosis infection in rabbits are discussed in relation to the host's socioecology and risk to livestock.  相似文献

Environmental and host-associated microbial communities are complex ecosystems, of which many members are still unknown. Hence, it is challenging to study community dynamics and important to create model systems of reduced complexity that mimic major community functions. Therefore, we developed MiMiC, a computational approach for data-driven design of simplified communities from shotgun metagenomes. We first built a comprehensive database of species-level bacterial and archaeal genomes (n = 22 627) consisting of binary (presence/absence) vectors of protein families (Pfam = 17 929). MiMiC predicts the composition of minimal consortia using an iterative scoring system based on maximal match-to-mismatch ratios between this database and the Pfam binary vector of any input metagenome. Pfam vectorization retained enough resolution to distinguish metagenomic profiles between six environmental and host-derived microbial communities (n = 937). The calculated number of species per minimal community ranged between 5 and 11, with MiMiC selected communities better recapitulating the functional repertoire of the original samples than randomly selected species. The inferred minimal communities retained habitat-specific features and were substantially different from communities consisting of most abundant members. The use of a mixture of known microbes revealed the ability to select 23 of 25 target species from the entire genome database. Cryptic species are increasingly being recognized in many organisms. In Brachionus rotifers, many morphologically similar yet genetically distinct species/biotypes have been described. A number of Brachionus cryptic species have been recognized among hatchery strains. In this study, we present a simple, one-step genetic method to detect the presence of those Brachionus sp. rotifers that have been found in hatcheries. With the proposed technique, each of the B. plicatilis sensu stricto, B. ibericus, Brachionus sp. Nevada, Brachionus sp. Austria, Brachionus sp. Manjavacas, and Brachionus sp. Cayman species and/or biotypes can be identified with polymerase chain reaction (PCR) analysis. Based on 233 cytochrome c oxidase subunit I sequences, we reviewed all the available cryptic Brachionus sp. genetic polymorphisms, and we designed six nested primers. With these primers, a specific amplicon of distinct size is produced for every one of the involved species/biotypes. Two highly sensitive protocols were developed for using the primers. Many of the primers can be combined in the same PCR. The proposed method has been found to be an effective and practical tool to investigate the presence of the above six cryptic species/biotypes in both individual and communal (bulk) rotifer deoxyribonucleic acid extractions from hatcheries. With this technique, hatchery managers could easily determine their rotifer composition at the level of cryptic species and monitor their cultures more efficiently. Lipocalin-2 is a novel adipokine that appears to play a role in the development of insulin resistance. Serum lipocalin-2 levels are elevated in obese patients. Obesity and insulin resistance are cardinal characteristics of the polycystic ovary syndrome (PCOS). However, there are limited data on serum lipocalin-2 levels in patients with PCOS. The marine finfish industry worldwide depends greatly on the mass culture of Brachionus rotifers. Recently, molecular data have revealed a more complicated view about the species status of Brachionus rotifers than previous mainly morphological assessments. Under this view, Brachionus rotifers are comprised of many morphologically similar, albeit genetically differentiated, cryptic members of larger groups. A redefinition of the cultured rotifer species/biotypes is therefore needed if aquaculture is to reach higher levels of standardization and predictability. In this work, restriction fragment length polymorphism (RFLP) and single-strand conformational polymorphism (SSCP) methods are applied to the COI and 16S rRNA mitochondrial genes. A detailed COI restriction map was constructed, using sequence data from all known representatives of Brachionus phylogroups. Therefore, it is the first time that such an extended restriction database has been produced. Several restriction endonucleases are proposed for the discrimination of the different Brachionus species/biotypes. Furthermore, eight different SSCP gel alleles are described for the 16S region. Using these data, five Brachionus species/biotypes were identified in 78 samples collected from laboratories and hatcheries around the world. This study examined the effects of a progressive resistance training program in addition to soccer training on the physical capacities of male adolescents. Eighteen soccer players (age: 12-15 years) were separated in a soccer (SOC; n = 9) and a strength-soccer (STR; n = 9) training group and 8 subjects of similar age constituted a control group. All players followed a soccer training program 5 times a week for the development of technical and tactical skills. In addition, the STR group followed a strength training program twice a week for 16 weeks. The program included 10 exercises, and at each exercise, 2-3 sets of 8-15 repetitions with a load 55-80% of 1 repetition maximum (1RM). Maximum strength ([1RM] leg press, bench-press), jumping ability (squat jump [SJ], countermovement jump [CMJ], repeated jumps for 30 seconds) running speed (30 m, 10 x 5-m shuttle run), flexibility (seat and reach), and soccer technique were measured at the beginning, after 8 weeks, and at the end of the training period. After 16 weeks of training, 1RM leg press, 10 x 5-m shuttle run speed, and performance in soccer technique were higher (p < 0.05) for the STR and the SOC groups than for the control group. One repetition maximum bench press and leg press, SJ and CMJ height, and 30-m speed were higher (p < 0.05) for the STR group compared with SOC and control groups. The above data show that soccer training alone improves more than normal growth maximum strength of the lower limps and agility. The addition of resistance training, however, improves more maximal strength of the upper and the lower body, vertical jump height, and 30-m speed. Thus, the combination of soccer and resistance training could be used for an overall development of the physical capacities of young boys.  相似文献

A parthenogenetic Artemia population from Torre Colimena, southernItaly, originally reported in 1998, is characterized by a multidisciplinaryapproach including cyst and naupliar biometry, morphometry ofadults, scanning electron microscopy (SEM) of brood pouch, cytogeneticsand 16S rRNA PCR-RFLP analysis. We confirmed parthenogeneticstatus, inferred ploidy level and determined phenotypic andmolecular relationships of this population through comparisonswith other asexual Artemia strains as well as bisexual speciesfound (A. salina) or introduced (A. franciscana) in the Mediterraneanbasin. Cyst and naupliar sizes for Torre Colimena are amongthe smallest recorded for asexual Artemia while the oppositeis true for chorion thickness. Discriminant analysis of adultbody measurements shows increased differentiation (89.5% forthe first four out of the twelve functions produced) from tetraploidparthenogenetic strains and bisexual species (A. salina andA. franciscana). Scanning electron micrographs of brood pouchreveal the characteristic morphology of asexual strains, whilechromosome observations of instar-I nauplii unequivocally establishdiploidy. Restriction patterns give evidence that the TorreColimena population shares an identical set of mitochondrialDNA (mtDNA) polymorphisms with a diploid Spanish parthenogeneticstrain and it is well differentiated from other tetraploid aswell as bisexual auto- and allochthonous strains (A. salinaand A. franciscana) from the Mediterranean. The present studymay serve as a reference methodological framework for multidisciplinarycharacterizations and biodiversity assessments in the genusArtemia.  相似文献
