RNA interference (RNAi) has revolutionised approaches to gene function determination. From a parasitology perspective, gene function studies have the added dimension of providing validation data, increasingly deemed essential to the initial phases of drug target selection, pre-screen development. Notionally advantageous to those working on nematode parasites is the fact that Caenorhabditis elegans research spawned RNAi discovery and continues to seed our understanding of its fundamentals. Unfortunately, RNAi data for nematode parasites illustrate variable and inconsistent susceptibilities which undermine confidence and exploitation. Poly(ADP‐ribosylation) is a post‐transductional modification that regulates protein's function. Most of the proteins subjected to this control mechanism belong to machineries involved in DNA damage repair, or DNA interacting proteins. Poly(ADP‐ribose) polymers are long chains of even 100 monomer length that can be branched at several positions but, not withstanding its importance, nothing is known concerning its structure. To understand, which are the geometrical parameters that confer to the polymer the structural constraints that determine its interaction with the target proteins, we have performed molecular dynamics of three chains of different length, made by 5, 25, and 30 units, the last one being branched. Analysis of the simulations allowed us to identify the main intra‐ and inter‐monomer dihedral angles that govern the structure of the polymer that however, does not reach a unique definite conformation. © 2013 Wiley Periodicals, Inc. Aims
Cervical cancer is the third most frequent cancer in women worldwide, mostly treated with cisplatin-based chemoradiotherapy. Since it is known that folate metabolism might interfere with cisplatin effectiveness, we intended to study the influence of the Gamma Glutamyl Hydrolase -401C > T polymorphism in treatment response in cervical cancer.Methods
We retrospectively reviewed the clinical data of 167 patients with bulky cervical cancer submitted to cisplatin-based chemoradiotherapy. The genotypes of GGH -401C > T SNP were determined by real-time PCR and statistical analysis was performed by χ2 test and survival analysis.Results
The genotypes of GGH-401C > T were significantly associated with the response to platinum-based chemoradiotherapy. Treatment response was higher in patients carrying the CC genotype, who presented a significant increased chance of treatment response (survival time in months/genotype: 91 for CC Vs 72 for CT/TT; p = 0.035, log rank test). A Cox regression analysis accordingly showed that the presence of the T allele was significantly linked to a worse treatment response (HR = 3.036; CI 95% 1.032-8.934, p = 0.044).Conclusions
The results of our study suggested the potential interest of GGH -401C > T as a predictive factor of the outcome of cervical carcinoma treated with cisplatin-based chemoradiotherapy. 相似文献97.

The genotoxicity and cytotoxicity of water in small urban basins was evaluated by the Salmonella/microsome assay and micronucleus test in V79 cells. The results showed that the cytotoxic effect was the most significant response in areas with medium to heavy urban occupation for both assays evaluated. Water samples from these areas include different concentrations of chloroform, bromodichloromethane, toluene, ethylbenzene, m,p-xylene and 1,4-dichlorobenzene. As to genotoxic damage, the presence of mainly direct-acting frameshift mutagens was detected in areas with less urban concentration and showed genotoxic activity in V79 cells in more heavily urbanized areas. Water organic extracts, evaluated using a microsuspension procedure, showed frameshift mutagenic activity in the presence of hepatic metabolization that increased as the population density grow. Chronic toxicity studies of sediment samples with the microcrustacean Daphnia magna showed that, while survival was not highly affected, reproductive inhibition was found in 92% of the observations. A retrospective diagnosis of water quality using traditional physicochemical parameters that defined the differential contribution of urban wastes at the three sites was associated with the biological assays. It became clear that the biological assays were of significant benefit in the diagnosis of risks of contamination of hydrographic basins by pollutants from urban non-point sources. 相似文献

The aim of this work was to identify the main allergy-related Ascomycetes fungal spores present in the atmosphere of Porto,
using different and complementary techniques. The atmospheric sampling, performed in the atmosphere of Porto (Portugal) from
August 2006 to July 2008, indicated Cladosporium, Penicillium, Aspergillus and Alternaria as the main fungal spore taxa. Alternaria and Cladosporium peaks were registered during summer. Aspergillus and Penicillium highest values were registered from late winter to early spring. Additionally, the Andersen sampler allowed the culture and
isolation of the collected viable spores subsequently used for different identification approaches. The internal-transcribed
spacer region of the nuclear ribosomal repeat unit sequences of airborne Ascomycetes fungi isolates revealed 11 taxonomically
related fungal species. Among the identified taxa, Penicillum and Aspergillus presented the highest diversity, while only one species of Cladosporium and Alternaria, respectively, were identified. All selected fungal spore taxa possessed phosphatase, esterase, leucine arylamidase and β-glucosidase
enzymatic activity, while none had lipase, cystine arylamidase, trypsin or β-glucuronidase activity. The association between
the spore cell wall morphology, DNA-based techniques and enzymatic activity approaches allowed a more reliable identification
