Background99mTc sestamibi scanning and aspiration biopsy can predict the histopathological result of a thyroid nodule fairly accurately.ObjectiveTo determine the accuracy of 99mTc sestamibi scanning in detecting malignancy in patients with thyroid nodule confirmed by definitive histopathological report after thyroidectomy.Material and methodsA total of 69 patients with a solitary thyroid nodule were studied. In all patients, fine needle aspiration, total thyroidectomy for suspected thyroid cancer, and histological analysis of the surgical specimen were performed. There were 54 patients with a positive 99mTc sestamibi scan; of these, malignancy was confirmed by histological analysis in 25 and excluded in 29. There were 15 patients with a negative 99mTc sestamibi scan; of these, three had a final diagnosis of cancer and 12 were confirmed as cancer-free.ResultsThe diagnostic accuracy of 99mTc sestamibi scanning in detecting malignancy in thyroid nodules was determined through a statistical analysis. 99mTc sestamibi scan for thyroid cancer had a sensitivity of 89.28% and a specificity of 29.25%. The positive predictive value was 46.29% and the negative predictive value was 80%.ConclusionsWe believe that 99mTc sestamibi scan should be routinely used in all patients with a thyroid nodule and an indeterminate result on fine needle aspiration. This procedure is most useful in excluding malignancy in patients with a negative 99mTc sestamibi scan.  相似文献

Although there is no doubt about the influence of the genetic background in the onset of the allergic diseases, Epigenome-Wide Association Studies are needed to elucidate the possible relationship between allergic diseases and epigenomic dysregulation. In this study we aimed to analyze the epigenetic patterns, in terms of DNA methylation, of three well-characterized populations of house dust mite allergic subjects, aspirin-intolerant asthmatics and controls. As a first, genome-wide phase, we used the HELP assay to study the methylation patterns in CD19 (+) B lymphocytes in these populations, and found that there are reproducible epigenetic differences at limited numbers of loci distinguishing the groups, corroborated by bisulphite MassArray in a second validation phase of an expanded 40 subject group. These validated epigenetic changes occur at loci characterized as important for the immune response. One such locus is a new candidate gene, CYP26A1, which shows differential methylation patterns and expression levels between groups. Our results suggest that epigenomic dysregulation may contribute to the susceptibility to allergic diseases, showing for the first time differences in DNA methylation between allergic and non-allergic healthy subjects, both globally and at specific loci. These observations indicate that the epigenome may offer new pathophysiological insights and therapeutic targets in atopic diseases.  相似文献

Coherences were observed between 15N3 of cytosine and its trans amino proton (H42) using a modified gradient-based heteronuclear single quantum coherence (HSQC) pulse sequence optimized for three-bond proton-nitrogen couplings. The method is demonstrated with a 22-nucleotide RNA fragment of the P5abc region of a group I intron uniformly labeled with 15N. Use of intraresidue 15 N3-amino proton couplings to assign cytosine 15 N3 signals complements the recently proposed JNN HNN COSY [Dingley, A.J. and Grzesiek, S. (1998) J. Am. Chem. Soc., 120, 8293–8297] method of identifying hydrogen-bonded base pairs in RNA.  相似文献

Páramos are high elevation tropical ecosystems in northern Andes, with large water yield and water regulation. One of the main and representative species growing in these páramos is the genus Espeletia, known as frailejones. There is a lack of knowledge of Espeletia ecophysiology, maybe due to its unusual anatomical modifications and the specific climatic conditions of these ecosystems. Therefore, it is important to determine the relationships between the anatomical modifications of Espeletia, its physiological functioning, and its contribution to the ecohydrologic functioning of páramos. Consequently, we studied the physiology of frailejones in two Colombian páramos, focused on the identification of conductive tissues inside the stems, calculated the age, and measured sapflow, using the heat ratio method. Results show that Espeletia spp. have a central pith that increases with height, as the size of secondary xylem decreases. Frailejones respond quickly to the changing conditions of weather factors controlling transpiration such as solar radiation, temperature, and fog presence. However, although environmental factors favor transpiration, the sapflow tends to decrease—a particular behavior of the Espeletia transpiration processes—since this occurs chaotically over time, including sapflow at night. The transformation of sapflow velocity to depth of water in a basin shows that the water lost through their transpiration is very low, which contributes to the high runoff ratio of páramo ecosystems. For the first time, we determine by radiocarbon the real ages of three E. hartwegiana, and their mean growth rates to range between 3.8 and 6.9 cm year?1.  相似文献

Genetic control of seed dormancy in barley (Hordeum vulgare L.) has mostly been described in terms of quantitative variation. Although some molecular markers for dormancy QTL have been identified, the corresponding genes involved in the regulation of the process have not been cloned. Induced barley mutants may constitute useful material to study the physiology and genetics of seed dormancy. The objective of this study was to identify the genetic control of this trait in a mutant (TL43) produced in the barley cv. Triumph. This mutant was selected for reduced dormancy and reduced sensitivity to abscisic acid (ABA). Two sets of F6 barley lines were selected for high and low levels of dormancy from a cross between the original dormant parent and the sodium azide-induced non-dormant TL43 mutant. Unexpectedly, given the near-isogenic nature of these two genotypes, polymorphism was detected for an SSR located in the centromeric region of chromosome 6(6H) out of a total of 92 molecular markers evenly distributed along the genome. Fortunately, upon three cycles of intensive divergent selection, every dormant and non-dormant F5 line consistently showed the genotype for this region identical to Triumph and TL43, respectively. Based on the mutagenic effect presumably attributed to sodium azide, mostly single point mutations, it cannot be clearly established if such extensive genomic variation on chromosome 6(6H) is due to the mutagenic treatment or may be an introgression from an unknown source. The means that could originate such heterogeneity are discussed; however, regardless of its origin, this genomic region shows a strong association with the expression of seed dormancy and provides an additional genetic locus for further studies of the mechanistic basis of this complex trait. In addition, since TL43 shows reduced sensitivity to ABA, the response to this hormone was determined on the F6 seed from the two sets of selected F5 lines. The results confirmed that the initial level of dormancy in the seed lot is the most important factor in determining ABA sensitivity.  相似文献

Stone tool use by wild chimpanzees of West Africa offers a unique opportunity to explore the evolutionary roots of technology during human evolution. However, detailed analyses of chimpanzee stone artifacts are still lacking, thus precluding a comparison with the earliest archaeological record. This paper presents the first systematic study of stone tools used by wild chimpanzees to crack open nuts in Bossou (Guinea-Conakry), and applies pioneering analytical techniques to such artifacts. Automatic morphometric GIS classification enabled to create maps of use wear over the stone tools (anvils, hammers, and hammers/ anvils), which were blind tested with GIS spatial analysis of damage patterns identified visually. Our analysis shows that chimpanzee stone tool use wear can be systematized and specific damage patterns discerned, allowing to discriminate between active and passive pounders in lithic assemblages. In summary, our results demonstrate the heuristic potential of combined suites of GIS techniques for the analysis of battered artifacts, and have enabled creating a referential framework of analysis in which wild chimpanzee battered tools can for the first time be directly compared to the early archaeological record.  相似文献

Aim To test the importance of the Lago Mare stage of the Messinian Salinity Crisis for the dispersal and diversification of European leuciscins (Cyprinidae: Leuciscinae). Location Europe. Methods Cytochrome b sequences of European leuciscins were employed to investigate phylogenetic relationships among species, using Bayesian inference, and to estimate times of diversification, using a relaxed molecular clock. The distributions of 190 European leuciscins were compiled, and regional species compositions were compared using a taxonomic similarity index and an area cladogram. Results Leuciscins restricted to the Iberian and Italian peninsulas and the West and South Balkan regions are phylogenetically more closely related to northern European species than to species from another southern European area. Application of a relaxed molecular clock to a Bayesian phylogeny indicates that most southern clades originated and diversified prior to the Messinian. Southern European regions are taxonomically distinct from one another, and from a more taxonomically homogeneous group of areas that includes Anatolia, East Balkans, Middle East, North Europe and West Russia. Main conclusions The scenario of a Messinian period of dispersal of Paratethyan fauna into Mediterranean regions, via the Lago Mare, predicts a rapid period of diversification and a pattern of close association among southern European faunas. Phylogenetic relationships among leuciscins, the timing of cladogenic events, and the taxonomic similarity among geographical regions do not conform to this expectation. The depth of clades endemic to southern Europe, together with the high levels of endemism in these regions, suggests that the faunas in these regions diverged prior to the Messinian and have evolved largely in isolation from one another. Our results support a model of gradual colonization of Mediterranean regions since the Oligocene. Subsequent connections between adjacent areas may have occurred in the Messinian or Pleistocene.  相似文献
