Summary A Saccharomyces cerevisiae strain harbouring the recombinant plasmid pSMF38TMA was cultured in a jar fermentor under the control of glucose concentration. In the recombinant plasmid, the mouse -amylase gene was fused to the S. cerevisiae SUC2 promoter. When glucose concentration in the medium was controlled at 10 g/l, the gene expression was completely repressed. On the other hand, the -amylase was produced and secreted in the medium at a very high level, around 200 mg/l as evaluated from the specific activity of commercially available human salivary amylase, when the glucose was kept at 0.15 g/l. This amount was almost 20-fold that obtained at 10 g/l glucose. The specific growth rate of the yeast in this culture was almost 60% of that attained with 10 g/l glucose. To obtain higher cell growth and productivity, the yeast was at first cultured at 2 g/l glucose and the concentration was then lowered to 0.15 g/l. By this control of the glucose concentration, on-off regulation of gene expression from the SUC promoter could be attained.  相似文献

The DNA coding for human nerve growth factor (hNGF) was chemically synthesized and introduced into Saccharomyces cerevisiae. Expression and secretion of hNGF was obtained by use of the yeast phosphoglycerate kinase-encoding gene promoter and the pre-pro sequence of the yeast alpha-mating factor. Immunoblotting with antiserum raised against a protein A-hNGF fusion protein, allowed the detection of an immunoreactive material secreted into the culture medium. A preparation from the culture medium, partially purified by ion-exchange column chromatography, stimulated neurite outgrowth from rat pheochromocytoma PC12h cells.  相似文献

Smooth muscle of guinea-pig taenia caecum was desensitized by treatment with 10(-4)M carbachol or 10(-4)M histamine for 30 min in Ca-free solution containing 2mM EGTA. Phosphatidylinositol turnover stimulated by carbachol was not reduced by desensitization with either carbachol or histamine, while the turnover stimulated by histamine was reduced by desensitization with histamine, but not with carbachol. These results are consistent with our previous report (1) that heterologous desensitization induced by carbachol occurs at intracellular Ca stores and homologous desensitization by histamine occurs at H1 receptors.  相似文献

We devised a simple and effective purification method for the microdetermination of 6,15-diketo-13,14-dihydro-prostaglandin F1alpha (DK), a metabolite of prostacyclin (PGI2). [18O]DK was synthesized from the repeated base-catalyzed hydrolysis of methyl ester derivatives in [18O] water to obtain an internal standard for the gas chromatography/selected ion monitoring (GC/SIM) of DK. The methyl ester-methoxime-tert-butyldimethylsilyl ether derivative was prepared, then gas chromatography/selected ion monitoring was carried out by monitoring the ion at m/z 613.4 for DK and at m/z 617.4 for an internal standard. A good linear response over the range of 10 pg to 10 ng was demonstrated. We detected DK to a level of about 297.8 pg/ml in human plasma. This method can be used to determine DK in biological samples.  相似文献

A mature sporophyte of Bryopsis plumosa (Hudson) C. Agardh forms a huge number of zoospores in its cell continuum. Zoospore formation starts with the division of a single giant nucleus and subsequent repeated mitosis. We found that an elevation of photosynthetic activity triggered the division of a mature giant nucleus. Transfer to short-day conditions was not necessary. Giant nuclei did not divide in darkness or in the presence of 1 μM DCMU. Giant nuclei of as many as 90% of sporophtyes started to divide following the addition of 5 mM NaHCO3 to the growth medium under continuous white light (6–12 W m−2). Frequency of nuclear division increased with increased light intensity. By combining those parameters that promoted the division of giant nuclei, we developed the "two-step culture method" which is composed of preliminary and main cultures. This new method guarantees that giant nuclei of more than 90% of all sporophytes synchronously divide between 72 and 96 h after the transfer to the main culture (continuous white light of 12 W m−2 in PES medium supplemented with 5 mM NaHCO3).  相似文献

We developed a determination method for human urinary 12-hydroxyeicosatetraenoic acid (12-HETE) using LC-MS-MS. This method, which includes simple extraction and detection in the SRM mode, allows precise and accurate determination of 12-HETE. There was a significant sex difference in urinary 12-HETE levels. Chiral analysis of 12-HETE using LC-MS-MS with column-switching technique revealed that the major enantiomer was 12(S)-HETE. Furthermore, the urinary level in patients with diabetes mellitus (DM) was analyzed. The present in vivo findings indicate that there could be difference in production of 12(S)-HETE between genders and 12(S)-HETE may play a role in the pathogenesis of DM.  相似文献

The responsiveness of non-dormant, upper cocklebur (Xanthiumpennsylvanicum Wallr.) seeds to various germination stimulants,such as CO2 C2H4 CS(NH2)2, BA and enriched O2, decreased withincreasing periods of water imbibition and was completely lostin the state of secondary dormancy. Unlike CO2 BA and CS(NH2)2however, C2H2 and enriched O2 effectively prevented the developmentof secondary dormancy, and their combination was the most effectivefor stimulating the germination of seeds which had undergoneimbibition for a long time. CS(NH2)2 and BA were effective,not by themselves but either under anaerobiosis or elevatedO2 tension. Growth of the axial and cotyledonary segments excisedfrom aged seeds remained responsive to these germination stimulantsand could be further stimulated by exogenous C2H2. With imbibitionat a lower temperature, the seeds maintained high germinationin response to various stimulants and a high rate of C2H2 andCO2 production during a long period of water imbibition. Theseresults are discussed in terms of the two possible causes forthe loss of responsiveness or induction of the secondary dormancy. (Received June 27, 1978; )  相似文献

Abstract Electron microscopic observation demonstrated that linear DNA plasmids, pGKL1 and pGKL2, were replicated by a strand displacement mechanism similar to adenovirus and Bacillus subtilis ø 29 phage. Moreover, their DNA replication was prevented by α-factor, a mating hormone which prevents the replication of chromosomal DNA and 2 μm plasmid in Saccharomyces cerevisiae mating type a cells. This result suggests that the replication of pGKL plasmids is controlled by the same genes that control the initiation or maintenance of chromosomal DNA and 2 μm plasmid replications.  相似文献

Facultatively alkaliphilic Bacillus sp. strain YN-2000 was isolated from an indigo ball. Although the strain has been extensively investigated as a representative strain of alkaliphilic bacillus, its taxonomic position is not yet known. Morphological, biochemical, and physiological characteristics and chemotaxonomic properties indicated that the strain was closely related to Bacillus cohnii; this was confirmed by the high homology of the 16S rRNA sequence and the construction of a phylogenetic tree on the basis of the 16S rRNA sequence and DNA–DNA relatedness data. Strain YN-2000 contained a larger amount of unsaturated fatty acids compared with Bacillus subtilis and the obligate alkaliphile, Bacillus alcalophilus, regardless of its culture pH. When the cells were grown at pH 10, the unsaturated fatty acid content and anteiso-/iso-branched fatty acid ratio became lower than those at pH 7. This result suggests that membrane fluidity decreases when the cells are grown at pH 10 compared to those of pH 7. In the cells of strain YN-2000 grown at pH 10, the cell-surface aspect was rougher, the cell shape was longer, and the cell-surface layer was thicker compared with those of the cells grown at pH 7. Effects of troglitazone, an antidiabetic thiazolidinedione that enhances insulin sensitivity, on thromboxane (TX) production were assessed in human erythroleukemia (HEL) cells and human platelets. Measurement of TX was performed by using the gas chromatography/selected ion monitoring (GC/SIM) method. We found that troglitazone reduced the TX production from HEL cells and human platelets. Furthermore, troglitazone also reduced arachidonic acid (AA)-induced TX production from HEL cell and thrombin-induced TX release from platelets. In addition, we compared the effect of troglitazone with that of alpha-tocopherol and BRL 49653. Other thiazolidinedione compound BRL 49653 had effects similar to troglitazone, but alpha-tocopherol had no effect on TX production. Our findings suggest that the thiazolidinedione group had an antithrombotic effect and was beneficial in preventing vascular complications often observed in diabetes mellitus.  相似文献
