Oligodendrocyte progenitor cells (OPCs) undergo marked morphological changes to become mature oligodendrocytes, but the metabolic resources for this process have not been fully elucidated. Although lactate, a metabolic derivative of glycogen, has been reported to be consumed in oligodendrocytes as a metabolite, and to ameliorate hypomyelination induced by low glucose conditions, it is not clear about the direct contribution of lactate to cell cycling and differentiation of OPCs, and the source of lactate for remyelination. Therefore, we evaluated the effect of 1,4‐dideoxy‐1,4‐imino‐d‐arabinitol (DAB), an inhibitor of the glycogen catabolic enzyme glycogen phosphorylase, in a mouse cuprizone model. Cuprizone induced demyelination in the corpus callosum and remyelination occurred after cuprizone treatment ceased. This remyelination was inhibited by the administration of DAB. To further examine whether lactate affects proliferation or differentiation of OPCs, we cultured mouse primary OPC‐rich cells and analyzed the effect of lactate. Lactate rescued the slowed cell cycling induced by 0.4 mM glucose, as assessed by the BrdU‐positive cell ratio. Lactate also promoted OPC differentiation detected by monitoring the mature oligodendrocyte marker myelin basic protein, in the presence of both 36.6 mM and 0.4 mM glucose. Furthermore, these lactate‐mediated effects were suppressed by the reported monocarboxylate transporter inhibitor, α‐cyano‐4‐hydroxy‐cinnamate. These results suggest that lactate directly promotes the cell cycling rate and differentiation of OPCs, and that glycogen, one of the sources of lactate, contributes to remyelination in vivo. J. The present study was designed to evaluate whether gestational exposure to an EMF targeting the head region, similar to that from cellular phones, might affect embryogenesis in rats. A 1.95‐GHz wide‐band code division multiple access (W‐CDMA) signal, which is one applied for the International Mobile Telecommunication 2000 (IMT‐2000) system and used for the freedom of mobile multimedia access (FOMA), was employed for exposure to the heads of four groups of pregnant CD(SD) IGS rats (20 per group) for gestational days 7–17. The exposure was performed for 90 min/day in the morning. The spatial average specific absorption rate (SAR) for individual brains was designed to be 0.67 and 2.0 W/kg with peak brain SARs of 3.1 and 7.0 W/kg for low (group 3) and high (group 4) exposures, respectively, and a whole‐body average SAR less than 0.4 W/kg so as not to cause thermal effects due to temperature elevation. Control and sham exposure groups were also included. At gestational day 20, all dams were killed and fetuses were taken out by cesarean section. There were no differences in maternal body weight gain. No adverse effects of EMF exposure were observed on any reproductive and embryotoxic parameters such as number of live (243–271 fetuses), dead or resorbed embryos, placental weights, sex ratios, weights or external, visceral or skeletal abnormalities of live fetuses. Ulva limnetica Ichihara et Shimada, sp. nov. (Ulvales, Ulvophyceae) is described from the Ryukyu Islands, Japan, and is characterized by thalli that are: (i) branched, tubular, fragile and wrinkled; (ii) up to 80 cm in height and up to 2 cm in diameter; (iii) light to yellowish green in color; and (iv) having an asexual reproduction by means of quadriflagellate swarmers. Rhizoidal cells bear tubular extensions on the outside of the cell layer in the stipe. Ulva limnetica is distinguished from species with similar thalli by chloroplast disposition, branching pattern, number of pyrenoids per cell and gross morphology. It is also distinguished by sequences of the nuclear-encoded 18S ribosomal RNA gene, internal transcribed spacer 2 region and the plastid-encoded large subunit of ribulose-1,5-bisphosphate carboxylase/oxgenase gene ( rbc L). Ulva limnetica was clustered with other Ulva species in an early diverging lineage within the genus.  相似文献

We surveyed ectomycorrhizae, ectomycorrhizal fungal fruit bodies, and soil physical properties in one heavily damaged and two lightly damaged pine stands on Mt. Tsukuba, central Japan. The rate of ectomycorrhizal root tips was not different between heavily and lightly damaged pine stands. For ectomycorrhizae, Cenococcum geophilum had high relative abundance in the heavily damaged pine stand. The number of ectomycorrhizal fungal fruit bodies in the heavily damaged pine stand was much lower than that in the lightly damaged pine stands.  相似文献

Two Rhizoclonium strains thriving in contrasting spring types (slightly hot spring/ambient, shaded pool spring) and biogeographic areas (El-Farafra Oasis in the Western Desert of Egypt/River-Po Plain in Northern Italy) were studied in depth based on field and cultured material, bright-field and fluorescence microscopy, and phylogenetic molecular analyses (SSU and LSU rRNA gene sequences). This polyphasic approach revealed that the two Rh. strains clearly belonged to the Rhizoclonium clade but differed in some of their key diagnostic features. The Egyptian Rhizoclonium strain, isolated from the spring-fed (Ain El-Balad) agricultural ditch in El-Farafra Oasis, was described with the working name ‘Rhizoclonium sp. 10.6 μm from a desert, slightly-hot spring’ based on its smaller cell diameter, ecological and phylogenetic molecular traits compared to allied morphospecies. Moreover, it was highlighted that the L/D ratio can be more than 3.0 within this group. The Italian Rhizoclonium strain, collected from the Fontanile Valle Re-shaded, pool ambient spring (Emilia-Romagna Region), was virtually identical to other strains identified as Rh. hieroglyphicum from China and Japan, due to its highly supported congruence in morphological characteristics and phylogenetic position. This study provided the first partial LSU and SSU rRNA gene sequences for European Rh. hieroglyphicum based on available literature. Phenotypic plasticity of rhizoid formation was also observed in both Rhizoclonium strains studied using culturing approaching techniques. Our investigation also confirms that a lot of work with a variety of approaches is still needed to assess the ecological preferences, morphological plasticity, and phylogenetics of freshwater Rhizoclonium taxa worldwide.  相似文献
