The interaction between two different monoclonal antibodies (Mabs) and their corresponding bispecific antibodies (Babs) with immobilized antigens was investigated using an optical biosensor (IAsys). The analyzed panel of affinity-purified antibodies included two parental Mabs (one of which was specific to human IgG (hIgG), and another one to horseradish peroxidase (HRP)), as well as Babs derived thereof (anti-hIgG/HRP). Babs resulting from the fusion of parental hybridomas bear two antigen-binding sites toward two different antigens and thus may interact with immobilized antigen through only one antigen-binding site (monovalently). Using an IAsys biosensor this study shows that the bivalent binding of Mabs predominates over the monovalent binding with immobilized HRP, whereas anti-hIgG parental Mabs were bound monovalently to the immobilized hIgG. The observed equilibrium association constant (K
ass) values obtained in our last work [1] by solid-phase radioimmunoassay are consistent with those constants obtained by IAsys. The K
ass of anti-HRP Mabs was about 50 times higher than that of anti-HRP shoulder of Babs. The dissociation rate constant (k
diss) for anti-HRP shoulder of Babs was 21 times higher than k
diss for anti-HRP Mabs. The comparison of the kinetic parameters for bivalent anti-HRP Mabs and Babs derived from anti-Mb/HRP and anti-hIgG/HRP, allowed to calculate that 95% of bound anti-HRP Mabs are bivalently linked with immobilized HRP, whereas only 5% of bound anti-HRP Mabs are monovalently linked. In general, the data obtained indicate that Babs bearing an enzyme-binding site may not be efficiently used instead of traditional antibody–enzyme conjugates in the case of binding of bivalent Mabs. 相似文献

Interhelical packing modulates conformational flexibility in the lactose permease of Escherichia coli 总被引:1,自引:0,他引:1
A key to obtaining an X-ray structure of the lactose permease of Escherichia coli (LacY) (Abramson, J., Smirnova, I., Kasho, V., Verner, G., Kaback, H. R., and Iwata, S. (2003) Science 301, 549-716) was the use of a mutant in which Cys154 (helix V) is replaced with Gly. LacY containing this mutation strongly favors an inward-facing conformation, which binds ligand with high affinity, but catalyzes little transport and exhibits few if any of the ligand-dependent conformational changes observed with wild-type LacY. The X-ray structure demonstrates that helix V crosses helix I in the approximate middle of the membrane in such a manner that Cys154 lies close to Gly24 (helix I). Therefore, it seems likely that replacing Cys154 with Gly may lead to tighter packing between helices I and V, thereby resulting in the phenotype observed. Consistently, replacement of Gly24 with Cys in the C154G mutant rescues significant transport activity, and the mutant exhibits properties similar to wild-type LacY with respect to substrate binding and thermostability. However, the only other replacements that rescue transport to any extent whatsoever are Val and Asp, both of which are much less effective than Cys. The results suggest that, although helix packing probably plays an important role with respect to the properties of the C154G mutant, the ability of Cys at position 24 to rescue transport activity of C154G is more complicated than simple replacement of bulk between positions 24 and 154. Rather, activity is dependent on more subtle interactions between the helices, and mutations that disrupt interactions between helix IV and loop 6-7 or between helices II and IV also rescue transport in the C154G mutant. 相似文献

After the prolonged (about 30 years) absence of Crimean-Congo hemorrhagic fever (CCHF) morbidity in the Stavropol territory cases of this infection were registered, and received laboratory confirmation, in summer of 1999-2000. At the end of the 1999 season antibodies to CCHF virus were detected among cattle-breeders in all 7 inspected regions of the territory. According to the data of the determination of virus contamination of Ixodes ticks (the season of 2000), the circulation of CCHF on the territory of 14 regions out of 24 expected was established. An essential factor of the exacerbation of the epidemic situation was a rise in the number of Hyalomma marginatum ticks, the main vector of the causative agent of CCHF in the south of Russia. 相似文献

Eighteen compounds were identified by GC-MS of their trimethylsilyl derivatives in n-butanolic extract from the biomass of Botryllus schlosseri. Three of them, 5-oxoproline, 5-hydroxyhydantoin, and kinurenic acid, were found in marine invertebrates for the first time. In addition to cellulose, the biomass was also shown to contain complex water-soluble sulfated polysaccharides. These were extracted and fractionated, and sulfate content and monosaccharide composition were determined in the fractions; fucose, xylose, galactose, mannose, glucose, glucosamine, galactosamine, and uronic acids were found. Identification of interaction partners opens a way to direct functional characterization of proteins. Several cDNAs coding for potential partners of protein kinase MAK-V/Hunk were isolated using two-hybrid cloning in yeast. Based on the partner properties, MAK-V/Hunk was assumed to play a role in tumorigenesis and tumor progression. This impact was attained by medicinal laser application to alexandrite, dye, erbium and gallium arsenid in various conditions on cultures of pulmonic fibroblasts of human embryo. Obtained results were estimated by transmission and scanning microscopy. The highest cell destruction was observed in dye and alexandrite, being less expressed in gallium arsenid under laser irradiation. Impulsive action of erbium laser did not cause any substantional cell destruction. So, laser application to dye alexandrite and gallium arsenide for healing scar and contracture damages is completely justified for skin pathogeny. 相似文献

The effect of three pleasant fine women's fragrances on the mood of women was assessed. The three fragrances had similar hedonic values but different sensory characteristics. The fragrances elicited nonspecific effects on mood. All three fragrances’ impact on mood was influenced by initial mood and lasted up to three hours, i.e., as long as the scent was still noticeable. Trends revealed that all three fragrances decreased negative affect and increased vigor. Also, independent of their hedonic value, the three fragrances elicited specific mood patterns. With the “woody citrus coniferous” scent, respondents experienced higher hostility and tension. The “floral chypre citrus” fragrance decreased anger and confusion. The “floral woody” scent had the lowest Total Mood Disturbance score and lowered depression, tension and confusion. 相似文献
