The ability to extrapolate from the known to the unknown is essential if we are to use the turnover of overall biodiversity, as opposed to a few well-known groups, to inform conservation planning. We investigated the usefulness of using evolutionary relationships of plants as a surrogate for the turnover of their associated beetle assemblages. If plant traits that are important to insects are phylogenetically conserved, it follows that there will be a positive relationship between insect faunal dissimilarity and plant evolutionary distance. We collected beetles using pyrethrum knock-down methods from 40 plant species belonging to four plant families in the Sydney region of Eastern Australia. We developed a novel approach for estimating variance in the dissimilarity of beetle assemblages, as explained by plant phylogeny, by using phylogenetic eigenvectors as explanatory variables in a distance-based redundancy analysis. We found a highly significant relationship between faunal dissimilarity and plant evolutionary distance for the entire beetle assemblage, the herbivorous component, and the non-herbivorous component, indicating that beetles generally showed some preference for particular plant clades as habitat, regardless of feeding guild. When comparing observed dissimilarities with those predicted from 40 jack-knife replicates of a Generalised Dissimilarity Model, we were often able to predict beetle turnover from plant phylogenetic relationships, although the reliability of this result was highly variable. Nevertheless, the broad response of beetle assemblages to plant evolutionary relatedness indicates real potential for plant phylogenetic pattern to act as a useful surrogate for insect biodiversity, especially when supplemented with other environmental correlates.  相似文献

The renal glomerulus is one of the few sites within the microvasculature in which leukocyte recruitment occurs in capillaries. However, due to the difficulty of directly visualizing the glomerulus, the mechanisms of leukocyte recruitment to glomerular capillaries are poorly understood. To overcome this, we rendered murine kidneys hydronephrotic to allow the visualization of the functional glomerular microvasculature during an inflammatory response. These experiments demonstrated that following infusion of anti-glomerular basement membrane (GBM) Ab, leukocytes became adherent in glomerular capillaries via a process of immediate arrest, without undergoing prior detectable rolling. However, despite the absence of rolling, this recruitment involved nonredundant roles for the P-selectin/P-selectin glycoprotein ligand-1 and beta2 integrin/ICAM-1 pathways, suggesting that a novel form of the multistep leukocyte adhesion cascade occurs in these vessels. Anti-GBM Ab also increased glomerular P-selectin expression and induced a P-selectin-independent increase in platelet accumulation. Moreover, platelet depletion prevented both the increase in glomerular P-selectin, and the leukocyte recruitment induced by anti-GBM Ab. Furthermore, depletion of neutrophils and platelets also prevented the increase in urinary protein excretion induced by anti-GBM Ab, indicating that their accumulation in glomeruli contributed to the development of renal injury. Finally, infusion of wild-type platelets into P-selectin-deficient mice restored the ability of glomeruli in these mice to support leukocyte adhesion. Together, these data indicate that anti-GBM Ab-induced leukocyte adhesion in glomeruli occurs via a novel pathway involving a nonrolling interaction mediated by platelet-derived P-selectin.  相似文献

CD100, a member of the semaphorin family, is a costimulatory molecule in adaptive immune responses by switching off CD72's negative signals. However, CD100's potential pathogenetic effects in damaging immune responses remain largely unexplored. We tested the hypothesis that CD100 plays a pathogenetic role in experimental immune complex glomerulonephritis. Daily injection of horse apoferritin for 14 days induced immune complex formation, mesangial proliferative glomerulonephritis and proteinuria in CD100-intact (CD100+/+) BALB/c mice. CD100-deficient (CD100-/-) mice were protected from histological and functional glomerular injury. They exhibited reduced deposition of Igs and C3 in glomeruli, reduced MCP-1 and MIP-2 intrarenal mRNA expression, and diminished glomerular macrophage accumulation. Attenuated glomerular injury was associated with decreased Ag-specific Ig production, reduced CD4+ cell activation and cytokine production. Following Ag injection, CD4+ cell CD100 expression was enhanced and dendritic cell CD86 expression was up-regulated. However, in CD100-/- mice, dendritic cell CD86 (but not CD80) up-regulation was significantly attenuated. Following i.p. immunization, CD86, but not CD80, promotes early Ag-specific TCR-transgenic DO11.10 CD4+ cell proliferation and IFN-gamma production, suggesting that CD100 expression enables full expression of CD86 and consequent CD4+ cell activation. Transfer of CD100+/+ DO11.10 cells into CD100-/- mice resulted in decreased proliferation demonstrating that CD100 from other sources in addition to CD100 from Ag-specific CD4+ cells plays a role in initial T cell proliferation. Although T cell-B cell interactions also may be relevant, these studies demonstrate that CD100 enhances pathogenetic humoral immune responses and promotes the activation of APCs by up-regulating CD86 expression.  相似文献

IL-18 is an important cofactor in Th1 immune responses and it has additional roles in inflammation. Recent reports suggest the contribution of IL-18 to immune responses may vary between mouse strains and immune contexts. We investigated the contribution of IL-18 to T-cell activation and joint inflammation in Ag-induced arthritis (AIA) in C57Bl/6 mice. AIA and cutaneous delayed-type hypersensitivity (DTH) reactions were induced in wild-type (WT) and IL-18-/- C57Bl/6 mice, and Ag-specific T-cell proliferation and IFN-gamma and IL-4 production were measured. The humoral immune response was measured as serum antibody to the disease-initiating Ag, methylated BSA (mBSA). Splenocyte production of IL-6 was measured by ELISA. To confirm the dependence of this model on Th1-cell-mediated immunity, IL-12p40-/- mice were similarly studied. WT mice developed synovitis, joint effusion, cartilage destruction and bone damage associated with induction of DTH, and in vitro Ag-specific T-cell proliferation and IFN-gamma production. Unexpectedly, IL-18-/- mice developed AIA and indices of T-cell activation were similar to those of WT mice. In contrast, IL-12p40-/- mice did not develop AIA, DTH or T-cell activation. WT and IL-18-/- mice, but not IL-12p40-/- mice, developed significantly increased serum antibody to mBSA compared with naive controls. WT and IL-18-/- splenocytes produced high levels of IL-6, whereas IL-12p40-/- cells had significantly lower IL-6 production compared with both. In conclusion, IL-18 is redundant both as a Th1 response cofactor and inflammatory cytokine, whereas IL-12p40-/- is a key cytokine, in AIA in C57Bl/6 mice.  相似文献

Paeoniflorin is one of the bioactive ingredients of the roots of Paeonia lactiflora (Paeoniaceae). A comparative study of processed and non-processed commercial samples of dried roots of P. lactiflora indicated a very low level of paeoniflorin in the processed sample and the formation of a new more polar component, sodium paeoniflorin sulphonate, during treatment of the roots with sulphiting agents.  相似文献

Aim  We tested various species-level traits for their potential to explain species' range sizes and dispersal abilities.
Location  Southeast Asia and Malay Archipelago.
Methods  We used published maps of geographical distribution estimates for sphingid moths to calculate range areas and classify species according to their dispersion across (present or historical) water straits in the Malay Archipelago. We tested forewing length (FWL), wing load (thorax width/FWL), presence or absence of a functional proboscis (i.e. adult feeding), larval diet breadth and larval diet composition for univariate correlations with range size and inter-island dispersion. We used multivariate, phylogenetically controlled models to test for independent effects of parameters.
Results  Range size correlated strongly with larval diet breadth, a result that was also confirmed in the multivariate model. Adult feeding had a significant impact on range sizes only within the multivariate model, but not in the univariate correlation. Dispersal class also correlated with larval diet breadth, but was additionally influenced by forewing length, wing load and larval diet composition. A univariate effect of adult feeding became non-significant in the multivariate, phylogenetically controlled model.
Main conclusions  Larval diet breadth is the best predictor of range size as well as inter-island dispersion, confirming the importance of niche breadth on the geographic ranges of species. A number of other factors are shown to have additional impact on predictions of range size or inter-island dispersal ability. Our analyses cannot determine the causal mechanisms of these correlations, but may stimulate further research on the adaptive significance of traits affecting range size and dispersal in this system.  相似文献

A partial dinosaur skeleton from the Upper Triassic (Norian) sediments of South Africa is described and named Antetonitrus ingenipes. It provides the first informative look at a basal sauropod that was beginning to show adaptations towards graviportal quadrupedalism such as an elongated forelimb, a modified femoral architecture, a shortened metatarsus and a changed distribution of weight across the foot. These adaptations allowed the clade to produce the largest-ever terrestrial animals. However, A. ingenipes lacked specializations of the hand found in more derived sauropods that indicate it retained the ability to grasp. Antetonitrus is older than the recently described Isanosaurus from Thailand and is the oldest known definitive sauropod.  相似文献

The contribution of IFN-gamma from bone marrow (BM) and non-BM-derived cells to glomerular and cutaneous delayed-type hypersensitivity (DTH) was studied in mice. Chimeric IFN-gamma mice (IFN-gamma(+/+) BM chimera), in which IFN-gamma production was restricted to BM-derived cells, were created by transplanting normal C57BL/6 (wild-type (WT)) BM into irradiated IFN-gamma-deficient mice. BM IFN-gamma-deficient chimeric mice (IFN-gamma(-/-) BM chimera) were created by transplanting WT mice with IFN-gamma-deficient BM. WT and sham chimeric mice (WT mice transplanted with WT BM) developed crescentic glomerulonephritis (GN) with features of DTH (including glomerular T cell and macrophage infiltration) in response to an Ag planted in their glomeruli and skin DTH following subdermal Ag challenge. IFN-gamma-deficient mice showed significant protection from crescentic GN and reduced cutaneous DTH. IFN-gamma(+/+) BM chimeric and IFN-gamma(-/-) BM chimeric mice showed similar attenuation of crescentic GN as IFN-gamma-deficient mice, whereas cutaneous DTH was reduced only in IFN-gamma(-/-) BM chimeras. In crescentic GN, IFN-gamma was expressed by tubular cells and occasional glomerular cells and was colocalized with infiltrating CD8(+) T cells, but not with CD4(+) T cells or macrophages. Renal MHC class II expression was reduced in IFN-gamma(+/+) BM chimeric mice and was more severely reduced in IFN-gamma-deficient mice and IFN-gamma(-/-) BM chimeric mice. These studies show that IFN-gamma expression by both BM-derived cells and intrinsic renal cells is required for the development of crescentic GN, but IFN-gamma production by resident cells is not essential for the development of cutaneous DTH.  相似文献

This paper describes the ant assemblages sampled from rain forest canopies ranging from southern Victoria through to Cape York Peninsula, Australia, and also in Brunei. Specifically, it examines the influence of decreasing latitude and variations in elevation on the character, richness, and abundance of the arboreal rain forest ant fauna, and also the relative contribution of ants to the total arthropod community. The sites that were examined included: cool temperate Nothofagus cunninghamii forest from a range of locations in Victoria; cool temperate N. moorei forest at both Werrikimbe and Styx River, New South Wales; notophyll vine forest in Lamington National Park, southeast Queensland; high elevation notophyll vine forest in Eungella National Park, central Queensland; complex notophyll vine forest at Robson Creek, Atherton Tablelands, north Queensland; complex mesophyll vine forest at Cape Tribulation, north Queensland; and mixed dipterocarp forest in Brunei. Although these sites represent a gradient increasingly tropical in character, botanically speaking, Eungella is less tropical than Lamington because of its high elevation. All samples were obtained by fogging the canopy with a rapid‐knockdown pyrethrin pesticide. In all cases, circular funnels were suspended beneath the foliage of individual trees or small plots of mixed canopy. Arthropods were collected four hours after fogging. Following ordinal sorting, ants were identified and counted to morphospecies level. The resulting catch were then standardized across sites as numbers caught per 0.5 m2 sampling funnel. Generic and species richness were higher at the lowland tropical Cape Tribulation sites than at the sites to the south and was comparable with values in the Brunei site. Species richness was negatively correlated with latitude and elevation. Within the Australian rain forest, the lowland/highland break appears to be the strongest predictor of ant relative abundance, with a weaker latitudinal relationship superimposed.  相似文献

Aim  The effects of resolution and spatial extent on range measures were explored in estimates of the geographic distribution of tropical hawkmoths. Furthermore, data were tested for phylogenetic autocorrelation.
Location  South-East Asia.
Methods  Various range measures, such as geographic information system (GIS)-supported range estimates, minimum convex polygons, latitudinal and longitudinal extents, and their products, were derived from original distribution records and compared to each other. A taxonomic classification of the species was used to analyse phylogenetic effects on range sizes.
Results  Range size measures exhibit a strongly right-skewed frequency distribution with many geographically restricted species and few widespread taxa. Rankings from GIS-supported, comprehensive range size estimates do not deviate greatly from more crude measurements of lower resolution. Comprehensive ranges and ranges within South-East Asia are correlated strongly, but already at this rather large scale the ranking of species changes considerably. Other measures of occupancy with an increasingly more localized consideration of 'range' show decreasing strengths of correlation. We found a weak, but significant, autocorrelation in range area data: related groups of species have ranges of similar size.
Main conclusions  Spatial resolution did not affect range ranking greatly in our data. However, macroecological studies based only on parts of species' ranges must be viewed critically, particularly if their extent is small compared to comprehensive ranges. Phylogenetic non-independence of range size data must be considered in comparative analyses.  相似文献
