Fms-like tyrosine kinase 3-ligand (FL) is a growth factor that may expand dendritic cell and regulatory T cell populations. We hypothesised that FL-induced regulatory T cells would protect mice from experimental rapidly progressive glomerulonephritis. To determine if FL was able to enhance regulatory T cell populations, C57BL/6 mice received 10 days of daily intraperitoneal injections of either FL or phosphate buffered saline. To induce accelerated autologous-phase anti-mouse glomerular basement membrane glomerulonephritis, mice were sensitized to sheep globulin 4 days prior to the induction of glomerulonephritis with sheep anti-mouse glomerular basement membrane globulin, and experiments ended 10 days later. FL was administered before, throughout and during the sensitization phase of this glomerulonephritis model. Renal disease and systemic immunity to the nephritogenic antigen were assessed. FL increased regulatory T cell and plasmacytoid dendritic cell proportions within spleen and lymph nodes. FL administration prior to glomerulonephritis did not protect mice from renal injury. When FL was given throughout the model, FL treated mice had reduced survival, with more interstitial neutrophils and glomerular CD11c+ cells than controls. Systemic immune responses showed increased IL-17A production from splenocytes, with more CD11c+ cells, but reduced plasmacytoid dendritic cell proportions in spleen and lymph nodes, despite increased regulatory T cell proportions. Under homeostatic conditions, FL expanded regulatory T cell and plasmacytoid dendritic cell populations, but FL enhanced systemic inflammatory responses and conventional dendritic cell populations when given during experimental glomerulonephritis, suggesting selective attempts to suppress pathogenic immunity by dendritic cell manipulation may be harmful.  相似文献

Aims Major patterns and determinants of the species richness of Sphingidae in the Malesian archipelago were investigated, including a distinction of richness patterns between subfamilies and range‐size classes. Location Southeast Asia, Malesia. Methods Using a compilation of specimen‐label data bases, geographic information system (GIS)‐supported estimates of distributional ranges for all Sphingidae species of Southeast Asia were used to assess the species richness of islands. Range maps for all species and checklists for 114 islands can be found at http://www.sphingidae‐sea.biozentrum.uni‐wuerzburg.de . Potential determinants of the species richness of islands were tested with general linear models. Results The estimated species richness of islands in the region is determined by biogeographical association, seasonality, availability of rain forest and island size. Species–area relationships are linear on a semi‐logarithmic representation, but not on a double‐logarithmic scale. Species richness of all sphingid subfamilies is influenced by biogeography. The presence of large rain‐forest areas affects mainly Smerinthinae, whereas distance from continental Asia is conspicuously irrelevant for this group. Widespread rather than geographically restricted species shape the overall distribution patterns of species richness. The altitudinal range of islands does not significantly affect species‐richness patterns, but its potential effects on geographically restricted species are discussed. Main conclusions As well as being affected by climatic and vegetation parameters, sphingid species richness is strongly influenced by a historical, directional dispersal process from continental Southeast Asia to the Pacific islands. This process did not apply equally to species of different taxonomic groups or range sizes. Widespread species decline in species richness towards the south‐east, whereas geographically restricted species exhibit an inverse pattern of species richness, probably because speciation becomes more important in this group within the more isolated island groups.  相似文献

Regulatory T cells (Tregs) must express appropriate skin-homing adhesion molecules to exert suppressive effects on dermal inflammation. However, the mechanisms whereby they control local inflammation remain unclear. In this study we used confocal intravital microscopy in wild-type and Foxp3-GFP mice to examine adhesion of effector T cells and Tregs in dermal venules. These experiments examined a two-challenge model of contact sensitivity (CS) in which Treg abundance in the skin progressively increases during the course of the response. Adhesion of CD4(+) T cells increased during CS, peaking 8-24 h after an initial hapten challenge, and within 4 h of a second challenge. At these time points, 40% of adherent CD4(+) T cells were Foxp3(+) Tregs. CD4(+) T cell adhesion was highly dependent on ICAM-1, and consistent with this finding, anti-ICAM-1 prevented Treg adhesion. Skin TGF-β levels were elevated in skin during both challenges, in parallel with Treg adhesion. In the two-challenge CS model, inhibition of ICAM-1 eliminated Treg adhesion, an effect associated with a significant increase in neutrophil adhesion. Similarly, total CD4(+) T cell depletion caused an increase in adhesion of CD8(+) T cells. Because Treg adhesion was restricted by both of these treatments, these experiments suggest that adherent Tregs can control adhesion of proinflammatory leukocytes in vivo. Moreover, the critical role of ICAM-1 in Treg adhesion provides a potential explanation for the exacerbation of inflammation reported in some studies of ICAM-1-deficient mice.  相似文献

There are over one million described invertebrate species on Earth, the majority of which are likely to inhabit the highly biodiverse rain forests around the equator. These are some of the most vulnerable ecosystems on Earth due to the pressures of deforestation and climate change with many of their inhabitants at risk of extinction. Invertebrates play a major role in ecosystem functioning from decomposition and nutrient cycling to herbivory and pollination; however, while our understanding of these roles is improving, we are far from being able to predict the consequences of further deforestation, climate change, and biodiversity loss due to the lack of comparative data and the high proportion of species which remain to be discovered. As we move into an era of increased pressure on old-growth habitats and biodiversity, it is imperative that we understand how changes to invertebrate communities, and the extinction of species, affect ecosystems. Innovative and comprehensive methods that approach these issues are needed. Here, we highlight priorities for future tropical terrestrial invertebrate research such as the efficiency of sustainable land management, exploration of innovative methods for better understanding of invertebrate ecology and behavior, and quantifying the role of invertebrates in ecosystem functioning.  相似文献

Montane birds face significant threats from a warming climate, so determining the environmental factors that most strongly influence the composition of such assemblages is of critical conservation importance. Changes in temperature and other environmental conditions along elevational gradients are known to influence the species richness and abundance of bird assemblages occupying mountains. However, the role of species‐specific traits in mediating the responses of bird species to changing conditions remains poorly understood. We aimed to determine whether different bird species responded differently to changing environmental conditions in a relatively understudied biodiversity hotspot in subtropical rainforest on the east coast of Australia. We examined patterns in avian species richness and abundance along two rainforest elevational gradients using monthly point counts between September 2015 and October 2016. Environmental data on temperature, wetness, canopy cover and canopy height were collected simultaneously, and trait information on body size and feeding guild membership for each bird species was obtained from the Handbook of Australian, New Zealand and Antarctic Birds. We used a generalized linear mixed modelling (GLMM) framework to determine the drivers of species richness and abundance and to quantify species’ trait–environment interactions. GLMMs indicated that temperature alone was significantly positively correlated with species richness and abundance. Species richness declined with increasing elevation. When modelling abundance, we found that feeding guild membership did not significantly affect species’ responses to environmental conditions. In contrast, the predicted abundance of a species was found to depend on its body size, due to significant positive interactions between this trait, temperature and canopy cover. Our findings indicate that large‐bodied birds are likely to increase in abundance more rapidly than small‐bodied birds with continued climatic warming. These results underline the importance of temperature as a driving factor of avian community assembly along environmental gradients.  相似文献

The mannose/glucose-binding Dolichos lablab lectin (designated DLL) has been purified from seeds of Dolichos lablab (hyacinth bean) to electrophoretic homogeneity by affinity chromatography on an ovalbumin- Sepharose 4B column. The purified lectin gave a single symmetric protein peak with an apparent molecular mass of 67 kDa on gel filtration chromatography, and five bands ranging from 10 kDa to 22 kDa upon SDS-PAGE. N-Terminal sequence analysis of these bands revealed subunit heterogeneity due to posttranslational proteolytic truncation at different sites mostly at the carboxyl terminus. The carbohydrate binding properties of the purified lectin were investigated by three different approaches: hemagglutination inhibition assay, quantitative precipitation inhibition assay, and ELISA. On the basis of these studies, it is concluded that the Dolichos lablab lectin has neither an extended carbohydrate combining site, nor a hydrophobic binding site adjacent to it. The carbohydrate combining site of DLL appears to most effectively accommodate a nonreducing terminal alpha-d-mannosyl unit, and to be complementary to the C-3, C-4, and C-6 equatorial hydroxyl groups of alpha-d-mannopyranosyl and alpha-d-glucopyranosyl residues. DLL strongly precipitates murine IgM but not IgG, and the recent finding that this lectin interacts specifically with NIH 3T3 fibroblasts transfected with the Flt3 tyrosine kinase receptor and preserves human cord blood stem cells and progenitors in a quiescent state for prolonged periods in culture, make this lectin a valuable tool in biomedical research.   相似文献

Successional processes in forest gaps created by tree falls are often considered a principal mechanism maintaining species diversity within forests. Although insects are important as mediators of forest recovery processes, there have been few observations of how they differ between forest gaps and non-gaps across latitudes. Here we tested the impacts of gap habitat on beetle assemblage composition at three locations spanning sub-tropical and tropical biomes (28°S, 16°S, 5°N: Lamington and Daintree in Australia, and Danum in Borneo). Beetles were collected by flight interception traps and sorted to family and feeding guilds. Despite clear differences in environmental conditions between gaps and non-gaps at all three location, we found weak and inconsistent differences in beetle family and feeding guild composition across a latitudinal gradient. PERMANOVA results showed no significant differences in beetle family and feeding guild composition between gaps and non-gaps. For beetle families, however, the interaction effect (between gap vs. non-gap treatment and location) was significant. Post-hoc analysis and NMDS ordinations showed significant and clear separation of family assemblages between gaps and non-gaps within the Danum samples but not within samples from either the Lamington or Daintree sites. Using IndVal, when all three locations were combined, Chrysomelidae and Tenebrionidae were found to be indicators for gaps and Scarabaeinae and Leiodidae for non-gaps. Analysed separately, gap indicators were only Chrysomelidae at Daintree, whereas non-gap indicators were Scarabaeinae at Lamington, Scarabaeinae, all other Scarabaeidae, Anthicidae, Scydmaeninae, and Hybosoridae at Danum. We suggest the scale of changes in insect composition and richness with the creation of a natural light gap in forests contrast with those resulting from anthropogenic disturbance such as logging and clearing of forests and the greater adaptability of forests to small-scale changes rather than large-scale changes.  相似文献
